Известен способ приготовления питательной среды для культивирования сальмонелл (Лабинская А.С. Микробиология с техникой микробиологических исследований. М., 1978, С.362). Среда обогащения "М" (магниевая среда с малахитовым зеленым в прописи Г.П.Калины и В.Л.Шигановой). При этом поэтапно готовят три раствора:
Раствор А:
Пептон отечественный (семипалатинский) - 0,42 г
Хлорид Na - 0,7 г
Фосфат K - 0,15 г
Дрожжевой диализат - 0,9 мл
Дистиллированная вода - 89 мл
Раствор Б:
Хлорид Mg - 0,36 г
Дистиллированная вода - 9 мл
Раствор В:
Бриллиантовый зеленый (водный р-р 0,5%) - 0,09 мл

Стерилизуют текучим паром дважды (100oC 30 мин). При этом для приготовления 1 л питательной среды исходные компоненты берут в следующем соотношении, г/л:
Na фосфорнокислый двузамещенный - 8,2 - 8,7
K фосфорнокислый однозамещенный - 2,2 - 2,7
Бентонитовая глина - 100 - 150
Дистиллированная вода - До 1 л
Питательная среда слегка опалесцирует, не имеет специфического запаха, pH среды 7,4. В полученную питательную среду дозированно вносят суспензию бактерий с целью накопления биомассы. Заражающая доза 1000 микробных клеток в 1 мл среды.
