Abstract
Description
The invention relates to medicine, namely to ophthalmology and can be used to treat diseases and injuries of the cornea.
Known eye drops "Keratonic" containing polysaccharides in the form of sorbitol, sodium chloride, riboflavin and water (RF patent N 2056821).
The disadvantages of these drops is that they do not have a specific effect on reparative processes in the cornea, since they do not contain specific active stimulants of corneal repair.
The objective of the invention is the creation of a dosage form with pronounced therapeutic properties in diseases and injuries of the cornea with a prologged effect.
The technical result is achieved by the fact that Corneolon eye drops, including the complex polysaccharide, sodium chloride and water, according to the invention, additionally contain anhydrous glucose, water-soluble cellulose derivatives, and sulfated glycosaminoglycans are used as complex polysaccharides, in the following ratio of components, wt.% :
Sulfated glycosaminoglycans (sGAG) - 0.01 - 0.1
Sodium Chloride - 0.9
Anhydrous glucose - 0.01 - 1
Water-soluble cellulose derivatives - 0.1 - 0.5
Purified Water - Else
The introduction of sGAG into drops is due to the fact that they are the main components of the cornea of the human and animal eyes; with injuries and diseases of the cornea, their level in the tissue decreases significantly. The composition of sGAG includes complex polysaccharides: keratan sulfate, chondroitin sulfate, heparan sulfate, heparin and dermatan sulfate.
SAGH actively bind water, which determines their decongestant effect, as well as stimulate corneal repair, affecting cell division. The concentration of sGAG in the solution is in the range from 0.01 to 0.1. A lower concentration has no therapeutic effect, and a concentration above 0.1 may cause unwanted side effects in the form of hyperemia and allergies.
Anhydrous glucose is included in the composition of the drops in a concentration of from 0.1 to 1. At a concentration below 0.1, it does not stabilize the sGAG molecule and does not have a decongestant effect, and at a concentration above 1 it will cause irritation of the cornea and conjunctiva.
Water-soluble cellulose derivatives are introduced in a concentration of 0.1 - 0.5 since a lower concentration than 0.1 does not prolong the action of the main active substances, and at a concentration above 0.5 the solution becomes excessively viscous.
Eye drops "Kornealon" are prepared as follows. First, a sodium chloride solution is prepared, for which 0.9 g of sodium chloride, 0.1 g of anhydrous glucose from 0.1 to 0.5 g of methylcellulose (by dry weight) and from 0.01 to 0 are successively dissolved in 50 ml of purified water , 1 g of lyophilized powder sGAG and stirred on a stirrer until completely dissolved, add water, purified to 100 ml. Sterilized through a 0.22 μm membrane filter.
SGAG is obtained from corneas of farm animals according to the method described in RF patent N 2056821. The determination of authenticity, qualitative and quantitative composition of SSAG is carried out according to Temporary Pharmacopoeia article 422678-96 on staining with 1.9 dimethylene blue method.
249). Anhydrous glucose analysis is carried out by the ferricyanide method as described (Reference Guide for Clinical Biochemistry. 0.1994 p. 249).
Methyl cellulose is analyzed according to TU-6-55-221-137-93 Research Institute "Polymersynthesis", Vladimir. Then the solution is poured into vials and sterilized by boiling.
