Abstract
Description
The invention relates to the chemistry of biologically active compounds, more specifically to compositions having reparative activity. Such compositions are intended for the treatment of wound and burn infections.
The invention may find use as a medicine in medical and veterinary practice.
Well-known compositions of drugs based on aqueous solutions of substances with reparative activity with concentrations that provide their therapeutic effect [1] For example, aqueous solutions of Acemine with a concentration of 25 wt. for oral administration or for microclysters on starch paste, containing 0.2 0.4 g of 1-methyluracil in a volume of 25 to 50 ml.
1-methyluracil [2] 1-methyl-2,4 (1H, 3H) -pyrimidinedione with MM 126.12. The cost of the drug with a purity of 98% and so pl. 236-238 o C is: 100 mg - 15.25 US dollars, 500 mg 48.50 US dollars.
Acemine [3] 6-acetamidohexanecarboxylic acid of the formula: CH 3 CONH (CH 2 ) 5 COOH, MM 173.21, the cost of the drug with a purity of 99% and so pl. 102-104 o C is: 25 g of US $ 23.60, 100 g of 71.20 US dollars.
One of the significant disadvantages of the known compositions based on aqueous solutions of substances with reparative activity is the rapid seeding of their microflora during storage. This leads to the need for either regular sterilization of the prepared solutions, or the use of only freshly prepared solutions under sterile conditions.
The objective of the invention is to create a composition based on aqueous solutions of substances with reparative activity, capable of self-sterilization.
This problem was solved by a composition based on substances with reparative activity, which is an aqueous solution of 1-methyluracil and / or acetaminum with concentrations providing their therapeutic effect, and a polymer antiseptic KATAPOL in an amount of 0.3-0.8 wt.
A distinctive feature of the claimed solution is the introduction of KATAPOL in the amount of 0.3 to 0.8 wt.
CATAPOL is a complex of dimethylbenzylalkylammonium chloride (catamine AB) and a copolymer of vinylpyrrolidone with crotonic acid with a ratio of monomer units of 60 to 81 mol. vinylpyrrolidone, 4 25 mol. sodium salt of crotonic acid and 15 mol. dimethylbenzylalkylammonium salt of crotonic acid with MM (10 60) • 10 3 [4,5] CATAPOL does not have many of the disadvantages of the low molecular weight antiseptic agent Catamine AB: high toxicity and skin-irritating effect.
It is known that 0.1 1/0 wt. KATAPOLA aqueous solutions have a wide spectrum of antimicrobial activity, including against bacteria that are resistant to antibiotics, as well as fungi and algae.
By the Decree of the Pharmacological Committee of the Ministry of Health of the USSR dated June 14, 1985, KATAPOL was recommended for production and use in medical practice as an antiseptic for treating wounded and burn surfaces.
However, preliminary studies conducted under sterile conditions have found that therapeutically active concentrations of KATAPOL inhibit reparative processes.
At the same time, neither 1-methyluracil nor acetamine have antimicrobial activity.
However, it was also found that the introduction of catapol in the declared concentration of 0.3 0.8% in aqueous solutions of 1-methyluracil or atsemin practically does not change their reparative activity with a slight increase in antimicrobial activity, although neither 1-methyluracil nor acetamine have antimicrobial activity .
The study of the reparative activity and antimicrobial action of the claimed compositions showed the presence of new properties of the ingredients of the composition that do not follow from the known level of science and technology.
Firstly, it turned out that in the declared concentration ranges KATAPOL does not show the effect of suppressing reparative activity.
Secondly, in the used concentration range of 1-methyluracil and atsemin, these substances enhance the antimicrobial activity of CATAPOL.
For the first time, new relationships discovered by property attributes make it possible to assert that the proposed invention meets the requirement of "creative level" and "novelty."
To confirm the compliance of the invention with the requirement of "industrial applicability" and to illustrate the essence of the claimed solution, we give examples of its specific implementation.
Example 6. 0.8 g of 1-methyluracil is added to a 100 ml volumetric flask, water is added to the mark. At the same time, 100 ml of 20 wt. KATAPOL solution by dilution of 20 ml of a 10% solution. Both solutions are mixed. The composition has a pH of 6.15. The content of CATAPOL 0.92 wt. 1-methyluracil - 0.45 wt.
Example 9. 4.0 g of Acemine are added to a 100 ml volumetric flask, then 5 ml of a 10% KATAPOL solution and water is added to the mark. The solution was stirred for 30 minutes. The resulting composition has a pH of 3.0. The content of CATAPOL 0.44 wt. Acemine 4.3 wt.
The examples presented in table. 1 and 2, obtained under conditions similar to those described above.
Analysis of tabular data shows:
an increase in the concentration of CATAPOL is accompanied by a certain decrease in the reparative activity of the composition without increasing its antimicrobial effect,
a decrease in the concentration of CATAPOL to 0.2 wt. accompanied by a significant decrease in the antimicrobial effect of the composition,
only in a narrow range of the stated concentrations of KATAPOL while maintaining therapeutically active concentrations of 1-methyluracil and atsemin it is possible to create a self-sterilizing composition with reparative activity that can be stored at 10 30 o C for 12 months without changing its properties.
The content of CATAPOL in the composition is determined in accordance with the provisional pharmacopeia article (VFS 42-2034-90), approved by the Pharmacopoeia Committee on 12/27/1990.
