Abstract
Description
The invention relates to medicine, namely to the pharmacy of immunoactive drugs, and can be used in the manufacture of immunity modulators used in various diseases.
491 494). In modern medicine, in the treatment of various diseases of the body’s immune system, thymic polypeptide hormones are used. The technology for the manufacture and modification of preparations includes the preparation of them from the calf’s goiter gland using an acetic reaction followed by gel filtration. This preparation refers to polypeptides with low molecular weight (Morozov V.G. Havinson V.Kh. Isolation from the thymus and study of the nature of the factor stimulating immunogenesis. Reports of the USSR Academy of Sciences, 1977, v. 233, No. 3, p. 491 494).
The known method due to the fact that the preparation is based on extraction using acids and ion exchange chromatography does not allow extracting a drug with high biological activity from the feedstock, which necessitates the use of sufficiently large doses of drugs.
4, с. 610 614). There is also known a method for modifying biologically active substances, based on the principle of mechanical dispersion of high molecular weight substances, in particular, at temperatures of 80-150 K, in a steel drum of a laboratory micro-mill with steel balls up to 12 min (A. Dubinskaya et al. Changing the structure of collagen under the influence of mechanical dispersion, Biophysics, 1980, v. XXV,
However, the existing modification method induces polymorphic transitions with an inactive reaction form in immunotropic preparations, protein denaturation, inactivation of enzymatic activity and instability as a result of hydrolytic decomposition.
The aim of the invention is to provide immunotropic drugs with greater biological activity and stability.
This goal is achieved by the fact that in the known method of modifying immunotropic drugs, including mechanochemical dispersion of 20 mg of the drug in a sealed vessel of molybdenum glass having a volume of 20 cm 3 in air with 30% relative humidity and a temperature of +25 o C using 7- mi glass balls of molybdenum glass weighing 7 g and a diameter of 6 mm for 2 5 min with a vibration amplitude of 9 mm and a frequency of 60 Hz.
The principal difference of the proposed modification method is that the main mechanochemical changes under the claimed dispersion regimes cause a specific modification of the molecular bonds of the thymus hormone polypeptides with the formation of free radicals, and since the dispersion was carried out in the presence of air with 30% relative humidity, the bulk of the free radicals were radicals peroxide type, which are formed during the oxidation of primary free radical mechanical degradation products Air islorodom.
It is known that immunization of an organism is accompanied, at certain stages, by the activation of peroxidation processes (Lukashova R.G. et al. The influence of thymus hormones on the activation of lipid peroxidation in rat lymphoid organs during the development of a humoral immune response. Report of the Ukrainian Academy of Sciences. Ser. B, 1989, N 9, p. 63 67). Therefore, the introduction into the body of previously mechanochemically modified polypeptide hormones of the thymus leads to the activation processes of immunological reactions. In this case, a stable topochemical form of the drug is formed with significantly higher biologically active properties, without contamination by wear products of glass products grinding it, since modifiable natural peptide hormones are significantly softer than glass.
Example. Carried out mechanochemical vibrational modification of 20 mg of the official drug "Timalin" (series 250690, 1992, St. Petersburg), placed in a sealed vessel of molybdenum glass, having a volume of 20 cm 3 in air at a temperature of +25 o C and 30 % relative humidity using 7 glass balls weighing 7 g of molybdenum glass with a diameter of 6 mm for 2 and 5 minutes with a vibration amplitude of 9 mm and a frequency of 60 Hz in a laboratory micro-mill.
(Bach J.F. Dardenne M. Bach M.A. Demonstration and characteeization of a circulating thymic hormones in mouse and in man. Transplant. Proc. 1973, v. 1, N 1, p. 99 104). 71 73). The biological activity of the preparations after mechanochemical modification, as well as the content of substances with thymosin-like serum activity (TCA) in the blood serum of thymectomized mice was determined in the test (Bach JF Dardenne M. Bach MA Demonstration and characteeization of a circulating thymic hormones in mouse and in man. Transplant. Proc. 1973, v. 1,
The studies were conducted on 100 male mice of the CBA line weighing 16–18 g, obtained from the Stolbovaya nursery of the Russian Academy of Medical Sciences.
1 и 2. The results of the studies are shown in table. 1 and 2.
The data presented in table 1 directly characterize the properties of thymalin in the in vitro system and indicate that the modified preparations have a higher biological activity and, accordingly, a higher peroxidation rate (according to the mechanoluminescence parameter) in comparison with the official control drug obtained in a known manner.
An analysis of the data presented in table 2 indicates that only mechanochemically modified preparations were able to increase the level of thymosin-like serum activity in thymectomized mice to a level higher than the thymic serum activity of intact mice. The introduction of modified drugs also restored the mechanoluminescence of splenocytes from thimectomized mice to the values observed in intact mice. At the same time, the introduction of official thymalin only partially restored the studied parameters. The noted properties of mechanochemically modified preparations and their storage in lyophilized form in glowing glass ampoules in the dark at a temperature of +4 o C are stored for more than 6 months.
существенное сокращение расхода исходного лекарственного сырья, уменьшение доза используемого препарата при одновременном уменьшении вероятности побочных явлений;
надежное сохранение полученного уровня свойств при общепринятых условиях хранения.the use of the proposed method for the modification of immunotropic drugs provides, in comparison with existing methods, the following advantages:
a significant reduction in the consumption of the initial medicinal raw materials, a reduction in the dose of the drug used, while reducing the likelihood of side effects;
reliable preservation of the obtained level of properties under generally accepted storage conditions.
