Claims (1)
- Способ оценки эффективности лечения лепры с помощью полимеразной цепной реакции (ПЦР), основанный на определении жизнеспособности Mycobacterium leprae, включающий применение ОТ-ПЦР с внешними и внутренними праймерами: 5'-GCATGTCTTGTGGTGGAAAGC-3' и 5'-CACCCCACCAACAAGCTGAT-3' и с использованием флуоресцентного зонда (FAM)-5'-CATCCTGCACCGCA-3'-(RTQ1), отличающийся тем, что ДНК/РНК выделяют из биоптатов и скарификатов кожи с предварительной их гомогенизацией, реакцию проводят в «Real time» при режиме амплификации: 95°С - 5 мин, 60°С - 50 сек - 1 цикл; 95°С - 15 сек, 62°С - 40 сек - 40 циклов, в Трис HCl буфере с KCl, MgCl2 твином и глицеролом и при снижении бактериоскопического индекса (БИН) и процента положительных образцов, полученных через 6 и 9 месяцев после начала лечения, определяют эффективность проводимой антимикобактериальной терапии.
