Способ дифференциации типичных и атипичных штаммов Yersinia pestis основного подвида средневекового биовара, предусматривающий выделение ДНК штамма, провдение ПЦР с гибридизационно-флуоресцентным учетом результатов с использованием олигонуклеотидных праймеров и зондов на мишени "Med24" и "pCKF" следующего состава: праймеры прямой Med24 RealTime-S GCCAGTGTGTGTCTAAA, обратный Med24 RealTime-As CAACATTCGTCGCAAAG и зонд Med24 Zond FAM-ACATTGTGCTGGACTCACAGCCCC-BHQ1, праймеры прямой pCKF RealTime-S AACCGCCTAAGCACTTTAT, обратный pCKF RealTime-As CGTCAGGAACTCAACGAA и зонд pCKF Zond FAM-ATCAGAGAGCATTTGAGCGGTTG-BHQ1, и разделение типичных и атипичных штаммов средневекового биовара по отсутствию гибридизационно-флуоресцентного сигнала по обеим ДНК мишеням у типичных штаммов и по его наличию по обеим ДНК мишеням у атипичных штаммов.A method for differentiating typical and atypical strains of Yersinia pestis from the main subspecies of medieval biovar, which involves isolating the strain DNA, performing PCR with hybridization-fluorescence results using oligonucleotide primers and probes on the Med24 and pCKF targets of the following composition: direct Med24 RealTime primers GCCAGTGTGTGTCTAAA, reverse Med24 RealTime-As CAACATTCGTCGCAAAG and probe Med24 Zond FAM-ACATTGTGCTGGACTCACAGCCCC-BHQ1, primers direct pCKF RealTime-S AACCGCCTAAGCACTTTAT, reverse pCKF RealTime-As CGTCAGGAACTCAACGAA and probe pCKF Zond FAM-ATCAGAGAGCATTTGAGCGGTTG-BHQ1, and separation of typical and atypical strains ednevekovogo biovar by the absence of hybridization-fluorescence signal on both DNA targets at typical strains and its presence on both target DNA in atypical strains.
