Abstract
1. Новый природный гликопептидный антибиотик ИНА 5812 с мол. массой 1917,8 Да, характерным УФ-спектром с λ(метанол) при 237, 262 и 360 нм и ИК-спектром в области: ν: 3301; 3062; 2927; 1645; 1538; 1382; 1231; 1072; 1003 и 702 см, содержащий специфический набор аминокислот - β-аланин, глицин, орнитин, тирозин, α,β-диаминомасляная кислота, аланин, пролин, лейцин, аспарагиновая кислота, серии, три небелковые аминокислоты и нейтральный сахар арабинозу и имеющий высокие показатели противобактериальной и химиотерапевтической активности при лечении стафилококкового сепсиса у белых мышей в опытах in vivo.2. Штамм Streptomyces roseoflavus ИНА-Ас-5812 - продуцент антибиотика ИНА 5812.3. Способ получения антибиотика ИНА 5812, предусматривающий культивирование продуцента Streptomyces roseoflavus, сорбцию из нативного раствора на Амберлите XAD-2 (Serva, Германия) с последующей его десорбцией смесью ацетон-н.-бутанол-вода и выделением антибиотика экстракцией н.-бутанолом, сорбцией экстракта на DEAE-Сефадексе, концентрацией, осаждением из 50% водно-спиртового раствора ацетоном и очисткой препарата-сырца на колонке с обращенной фазой С-18 в виде белого порошка, хорошо растворимого в воде и метаноле.1. Strain Streptomyces roseoflavus INA-Ac-5812 - producer of the antibiotic INA 5812.3. A method of producing an INA 5812 antibiotic, which involves culturing the producer Streptomyces roseoflavus, sorption from a native solution on Amberlite XAD-2 (Serva, Germany), followed by its desorption with acetone-n-butanol-water mixture and isolation of the antibiotic by extraction of n-butanol, sorption of the extract on DEAE-Sephadex, concentration, precipitation of acetone from a 50% aqueous-alcoholic solution and purification of the crude preparation on a C-18 reverse phase column in the form of a white powder, readily soluble in water and methanol.
