Claims (20)
1. Способ получения или повышения стойкости, по меньшей мере, к одному биотическому или абиотическому стрессовому фактору в растениях, включающий следующие стадии
a) повышение количества протеина или функции, по меньшей мере, одного протеина Вах ингибитора -1 (BI1) в, по меньшей мере, одной растительной ткани с условием, что экспрессию в эпидермисе листьев оставляют в основном неизменной или снижают, и
b) выбор растений, при которых по сравнению с исходными растениями имеется или повышена стойкость к, по меньшей мере, одному биотическому или абиотическому стрессовому фактору.
2. Способ по п.1, при котором стрессовым фактором является растительный патоген.
3. Способ по п.1, при котором стрессовым фактором является сапрофитный или гемибиотрофный патоген.
4. Способ по п.1, при котором BI-1 протеин включает, по меньшей мере, одну последовательность, которая имеет гомологию, по меньшей мере, в 50% к, по меньшей мере, одному консенсусному BI1-мотиву, выбранному из группы, включающей
a) H(L/I)KXVY,
b) AXGA(Y/F)XH,
c) NIGG,
d) P(V/P)(Y/F)E(E/Q)(R/Q)KR,
e) (E/Q)G(A/S)S(V/I)GPL,
f) DP(S/G)(L/I)(1/L),
g) V(G/А)Т(А/S)(L-/I)AF(А/G)CF(S/T),
h) YL(Y/F)LGG,
i) L(L/V)SS(G/W)L(S/T)(I/M)L(L/M)W, и
j) DTGX(I/V)(I/V)E.
5. Способ по п.1, при котором BI-протеин кодируют полипептидом, который включает последовательность, выбранную из группы, состоящей из
a) последовательностей согласно SEQ ID NO: 2, 4, 6, 8, 10, 12, 14, 16, 18, 20, 22, 24, 26, 28, 30, 32 или 38, и
b) последовательностей, которые имеют идентичность, по меньшей мере, 50% к одной из последовательностей согласно SEQ ID NO: 2, 4, 6, 8, 10, 12, 14, 16, 18, 20, 22, 24, 26, 28, 30, 32 или 38,
c) последовательностей, которые включают, по меньшей мере, одну частичную последовательность, по меньшей мере, 10 связанных аминокислотных остатков одной из последовательностей согласно SEQ ID NO: 2, 4, 6, 8, 10, 12, 14, 16, 18, 20, 22, 24, 26, 28, 30, 32 или 38.
6. Способ по п.1, при котором повышение количества протеина или функции, по меньшей мере, одного BI1-протеина осуществляют рекомбинантной экспрессией названного BI1-протеина под контролем корень-, клубень- или мезофиллспецифичного промотора.
7. Способ по п.1, включающий
(a) стабильную трансформацию растительной клетки рекомбинантной экспрессионной кассеты, содержащей кодирующую BI1-протеин последовательность нуклеиновых кислот в функциональной связи с тканеспецифичным промотором, причем промотор в основном не имеет активности в эпидермисе листьев и причем промотор является гетерологичным по отношению к названной, кодирующей BI-протеин последовательности нуклеиновых кислот,
(b) регенерацию растения из растительной клетки и
(c) экспрессию названной кодирующей BI-протеин последовательности нуклеиновых кислот в количестве и на время, достаточные для получения или повышения стойкости к стрессу и/или патогену в названном растении.
8. Способ по п.1, при котором растения выбирают из однодольных и двудольных растений.
9. Способ по п.1, при котором растения выбирают из группы, включающей однодольные растения, выбранные из группы, содержащей пшеницу, овес, просо, ячмень, рожь, кукурузу, рис, гречиху, сорго, тритикале, полбу, лен или сахарный тростник.
10. Способ по п.1, при котором повышают экспрессию Вах ингибитора-1 (BI-1) в мезофилле.
11. Способ по одному из пп.1-10, при котором растение имеет MIo-стойкий фенотип, или экспрессию или функцию MIo, RacB и/или NaOx ингибируют, по меньшей мере, в эпидермисе или снижают по сравнению с контрольным растением и/или экспрессию или функцию PEN2, SNAP34 и/или PEN1, по меньшей мере, в эпидермисе повышают по сравнению с контрольным растением.
12. Полипептидная последовательность, кодирующая BI1-протеин, включающая, по меньшей мере, одну последовательность, выбранную из группы, включающей
a) последовательности согласно SEQ ID NO: 12, 14, 16, 18, 20, 22, 24, 28, 30, 32 или 38,
b) последовательности, которые имеют гомологию, по меньшей мере, 90%, предпочтительно, по меньшей мере, 95%, особенно предпочтительно, по меньшей мере, 98% к одной из последовательностей согласно SEQ ID NO: 12, 14, 16, 18, 20, 22, 24, 28, 30, 32 или 38, и
c) последовательности, которые включают, по меньшей мере, 10, предпочтительно, по меньшей мере, 20, особенно предпочтительно, по меньшей мере, 30 связанных аминокислот одной из последовательностей согласно SEQ ID NO: 12, 14, 16, 18, 20, 22, 24, 28, 30, 32 или 38.
13. Последовательность нуклеиновых кислот, кодирующая полипептидную последовательность по п.12.
14. Рекомбинантная экспрессионная кассета, содержащая кодирующую BI1-протеин последовательность нуклеиновых кислот в функциональной связи с тканеспецифичным промотором, причем промотор в основном не имеет активности в эпидермисе листьев и причем промотор является гетерологичным по отношению к названной кодирующей BI1-протеин последовательности нуклеиновых кислот.
15. Рекомбинантная экспрессионная кассета по п.14, при которой
a) BI1-протеин определен согласно одному из пп.4, 5 или 11, и/или
b) тканеспецифичный промотор выбран из группы, включающей корень-, клубень- или мезофиллспецифичные промоторы.
16. Рекомбинантный вектор, содержащий экспрессионную кассету согласно п.14 или 15.
17. Рекомбинантный организм, содержащий, по меньшей мере, одну экспрессионную кассету согласно п.14 или 15 и/или, по меньшей мере, один вектор согласно п.16.
18. Рекомбинантный организм по п.17, выбранный из группы, включающей бактерии, дрожжи, нечеловеческие животные и растения.
19. Рекомбинантный организм по п.17, выбранный из группы, включающей пшеницу, овес, просо, ячмень, рожь, кукурузу, рис, гречиху, сорго, тритикале, полбу, лен, сахарный тростник, рапс, кресс, Arabidopsis, сорта капусты, сою, люцерну, горох, бобовые растения, земляной орех, картофель, табак, томаты, баклажаны, перец, подсолнечник, тагетес, салат, календулу, дыню, тыкву и цукини.
20. Рекомбинантный организм по одному из пп.17-19, при котором организм является растением, которое дополнительно имеет MIo-стойкий фенотип.
