Abstract
Способ позвол ет повысить контрастность, результативность исследовани ..Usage: in clinical microbiology and can be used for staining flagella in proteas in determining their topology. The essence of the invention: sequentially fix the preparation with an aqueous solution containing 1% acetic acid and 2% formalin, etch with an aqueous solution containing 1% phenol and 5% tannin, without heating, followed by impregnation, which is carried out in two stages: at the first stage, the preparation impregnated with a 25% solution of silver nitrate for 10 minutes and oxidized with a 0.5% solution of formalin, and in the second step, it is passed through a mixture consisting of a 25% solution of silver ammonia and 40% sodium hydroxide taken in a ratio of 100: 1, and then reduced with a 1% formalin solution. The method allows to increase the contrast, research efficiency ..
Description
The invention relates to clinical microbiology and can be used for staining flagellum proteins.
The aim of the invention is to increase the contrast and efficiency of the method.
The method is carried out as follows.
A suspension of the growing protea culture is placed on a previously prepared clean, defatted glass slide and the preparation is dried at a temperature of 20 ± 5 ° С, and then fixed with an aqueous solution containing 1% acetic acid and 2% formalin for 10 min. The solution is drained, the preparation is washed with running water for 15 minutes (no direct flow of water to the preparation is allowed). After this, the drug is etched with an aqueous solution containing 1% phenol and 5% tannin for
15 minutes, washed with running water for 20 minutes and dried at room temperature.
Impregnation of the drug is carried out in two stages. At the first stage of coloring, a 25% solution of silver nitrate is layered on the preparation and kept in it for 10 min at room temperature. The solution is drained and layered with a 0.5% formalin solution for one and a half minutes.
In a second step, after removal of the formalin solution, a silver ammonia mixture is layered on the preparation for 1.5 minutes. This mixture is prepared as follows: a 25% silver nitrate solution and a 40% sodium hydroxide solution are taken in a ratio of 100: 1, and the resulting mixture is brought to a state of slight opalescence and a 25-27% ammonia solution. Ополаскивают дистиллированной водой, высушивают при комнатной температуре, с последующим заключением препарата в канадский бальзам под покровное стекло.silver cat is drained from the preparation and a 1% formalin solution is layered for 1 min. Rinse with distilled water, dry at room temperature, followed by the conclusion of the drug in a Canadian balm under a coverslip.
Выполнение способа обеспечивает повышение контрастности окраски жгутиков проте и повышает результативность метода .The implementation of the method provides an increase in the contrast of the color of the flagella prote and increases the effectiveness of the method.
On the stained preparation, dark brown prote cells of different sizes and their flagella located peritrichously are clearly visible. Moreover, regardless of the size of the cells, the flagella are stained in contrast, they were absent in single cells.
The achieved clarity of revealing the morphological picture of the protein in a series of experiments made it possible to study it at different stages of swarming, to reveal the effect of antibiotics and other bactericidal agents on the change in the nature of flagging, as well as the dependence of flagging on the stages of swarming and cell size.
