Description
The utility model relates to the field of microbiology, namely, equipment and devices for analyzing microorganisms, in particular, equipment used during clinical laboratory diagnostics.
п.).Currently, a significant number of different methods of analysis of microorganisms are known, for the implementation of which devices and devices are required. Typically, in the analysis, containers are used for sample preparation, cultivation of microorganisms, their storage, introduction of labels or other components (tubes, Petri dishes, etc.), devices for analysis, for example, a set of glass and cover glasses and analytical instruments ( microscope, fluorimeter, etc.).
56-61).A known complex for the analysis of microorganisms using an automated microscope, in which a preparation containing microorganisms, is applied to a coverslip before analysis and covered with a layer of agar gel (A.V. Chernyadyev, A.G. Zolotarev, A.L. Kovtun. Automated morphometry of bacteria , possibilities and limitations of application. - Biotechnology, 3, 1996, p.56-61).
The disadvantage of the above complex is the use of expensive equipment, as well as the inability to quickly prepare a series of samples and their analysis, which is usually required in clinical laboratories.
Closest to the claimed technical solution is the complex used for the analysis of microorganisms in clinical laboratories in accordance with the order of the Ministry of Health of the USSR No. 535 of April 22, 1985. The complex includes tubes for storing samples, a set of subject (75 × 50 mm) and coverslips (22 × 22 mm) of borosilicate glass (for example, Ted Pella, Inc (USA) Cat. No. 26005 and Cat. No. 260140) and a light microscope.
Микроскопию проводят при опущенном конденсоре сначала при малом увеличении (объектив ×8), затем при большом (объектив ×40).For analysis, a sample from a test tube is placed on a glass slide in a drop of physiological saline and the coverslip is carefully covered so that the liquid is free of air bubbles. A correctly made drop fills the entire space between the coverslip and the slide, but the liquid does not protrude beyond the edges of the coverslip. Microscopy is carried out with the condenser lowered, first at low magnification (lens × 8), then at large (lens × 40).
The disadvantage of this device is the need for a separate preparation of each analyzed sample, a large number of consumables is required (the consumption of one slide and one coverslip is assumed for one study), the need to place a microscope near the sample preparation site, which significantly increases the time during serial analyzes.
The challenge facing the authors was to create a device for microbiological research, which allows to reduce the analysis time, simplify the procedure for conducting research and increase their reliability by reducing the role of the subjective factor.
The specified technical result is achieved by using a microbiological multi-well strip as a container for sample preparation, and a transparent two-layer plastic comb cartridge, the number of teeth of which corresponds to the number of holes of the strip, and the teeth of the comb are at a distance from corresponding to the distance between the holes of the strip. In addition, each of the teeth has a lateral slot 0.05-0.2 mm wide and 6-25 mm long. The specified size limits of the slots allow fluid to fill the entire tooth chamber. The sizes are established by the authors empirically.
A standard strip (for example, according to TU 9393-009-16548645-2005) contains 8 isolated holes, but it is possible to use strips with a different number of holes.
In fig. 1 shows a General view of the device - front view (a) and side view (b).
(7).The device consists of a strip (1) with holes (2) and a transparent two-layer plastic comb cartridge cartridge (3). The comb cartridge (3) is made of two plates of a polymer optically transparent material, which are interconnected in three places in such a way that each tooth (4) of the comb has an inner chamber (5), 0.1 mm thick, with side slots (6 ) The number of teeth (4) of the comb cartridge (3) corresponds to the number of holes (2) of the strip (1), moreover, the teeth of the comb are at a distance corresponding to the distance between the holes of the strip. On the comb housing opposite each tooth there is a place for inscription (7).
The device is used as follows. Analyzed liquid samples are placed in the wells (2) of the strip (1), and then the teeth (4) of the comb cartridge (3) are pre-marked in the appropriate places (7). Under the action of capillary forces for 10-15 seconds, the liquid from the holes through the slots (6) completely fills the microchambers (5) of the cartridge. Excess fluid is removed by touching the tips of the teeth on the filter paper. Then the cartridge is placed in a transparent plastic envelope and microscopic using a light microscope in a dark field at a magnification of 400-600 times.
The device allows simultaneous testing for the presence of microorganisms in the culture medium, as well as determining the sensitivity of microorganisms to a large number of chemotherapeutic agents. At the same time, the test time is significantly shortened due to the simultaneous preparation and viewing of 8 samples at once, and the reliability of the results also increases, since the stage of manual preparation of the preparation is not ruled out.
