Abstract

For the prepn. of a Rubella antigen, a strain of the virus is cultivated at 37 deg.C on Vero films in Eagle medium, contg. 5% inactive calf serum until a maximum cytopathological effect is reached. Following this the cellular monolayer is rinsed twice with a seline phosphate buffer of pH 7.2-7.4. The sauce buffer is added in a quantity of 1/50-1/100 of the original vol. of the culture. The culture is frozen at -70 to -75 deg.C for 1-5 mins. and defrosted suddenly at 37 deg.C. The sample is treated by 24 KHz ultrasound for three cycles of 30 secs. and centrifuged. 10% gelatine sorbitol soln. is added to the supernatant and the prepn. is lyophilised.
