Abstract
Procedeul de preparare a antigenului treponemic, conform inventiei, consta în aceea ca tulpina de Treponema pallidum înregistrata în colectia Institutului Cantacuzino la nr.13156 se inoculeaza intratesticular la iepuri, dupa 10 zile se recolteaza tesutul infectat si se suspenda în tampon fosfat salin ce contine 100 u.i. heparina/ml, se lasa 45 min la 37 degree C, dupa care se decanteaza, se centrifugheaza si se filtreaza prin filtru de sticla cu porozitate medie, se trateaza apoi cu amestec constituit din sarea metil-glucaminica a acidului N,N'-diacetil-3,5-diamino- 2,4,6-triizodbenzoic si polidextran cu greutate moleculara 400.000 în raport de 7:3 si se centrifugheaza, iar la supernatantul obtinut se adauga formaldehida pîna la obtinerea concentratiei de 1%, se lasa 7 zile la 4 degree C, se centrifugheaza si sedimentul este spalat de 3 ori cu tampon fosfat salin, prin centrifugare se aduce la concentratia finala de 2.10 exponent 7 germeni/ml în tampon fosfat salin si apoi se adauga 0,75% glicocol, 0,5% glutamat de sodiu si 0,02% tiomersal, se repartizeaza în fiole, se liofilizeaza prin congelare la -58 degree C si apoi uscare pîna la temperatura de 30 degree C pe o perioada de 48 h, fiolele se închid în atmosfera de argon.The process for preparing the treponemic antigen according to the invention consists in that the strain of Treponema pallidum, registered in the Cantacuzino Institute at No. 13156, is inoculated intra-articularly in rabbits, after 10 days the infected tissue is harvested and suspended in saline phosphate buffer containing 100 IU heparin / ml, is left for 45 min at 37 ° C, then decanted, centrifuged and filtered through a medium-porosity glass filter, then treated with a mixture of N, N'-diacetyl- -3,5-diamino-2,4,6-triisobenzoic acid and polydextran with a molecular weight of 400,000 in a 7: 3 ratio and centrifuged and formaldehyde was added to the obtained supernatant to a 1% concentration, left 7 days at 4 ° C, centrifuged and the sediment washed 3 times with saline phosphate buffer, centrifugation at a final concentration of 2.10 g / mL in saline phosphate buffer and then 0.75% glycol, 0.5% sodium glutamate and 0.02% thiomersal are dispensed into vials, lyophilized by freezing at -58 ° C and then dried to 30 ° C for 48 hours and the vials are closed under argon.
