Abstract
For the prepn. of antitransferrin human serum, 1 vol. of urine, contg. 50 mg of transferrin is mixed with a corresp. quantity of mono-specific antitransferrin specific serum. The mixt. is allowed to stand at 37 deg.C for 1 hr. prior to centrifuging. The ppte. obtd. is washed with 0.05% to Na-dodecyl-sulphate soln. and taken up into a least vol. of 0.05 M Na-veronal buffer contg. 0.5% Na-dodecyl-sulphate at 30 deg.C and pH. The liq. is passed through an extrafine Bio-Gel P-1000 chromatographic column at 30 deg.C and at a rate of 10 mls/hr. The second drop is collected, purified, concentrated and used to immunise animals with the urine to prepare the serum.
