Abstract
Przedmiotem zgłoszenia jest sposób oczyszczania jednoniciowego DNA o długości do 100 zasad znajdujący zastosowanie w przemyśle biotechnologicznym w procedurach oczyszczania DNA. Sposób oczyszczania jednoniciowego DNA o długości do 100 zasad charakteryzuje tym, że próbkę DNA zawieszoną w buforze TBE, PBS, TRIS, TE lub wodzie miesza się z 3 M roztworem octanu sodu o pH 5,3 +/- 0,1 w stosunku objętościowym od 10:1 - 10:2, do próbki dodaje się izopropanol w ilości od 0,33 do 1,25 objętości otrzymanej wcześniej mieszaniny, następnie mieszaninę nanosi się na kolumienkę ze złożem krzemionkowym, kolumienkę wiruje się lub podłącza do pompy próżniowej, celem przesączenia przez kolumienkę jej zawartości, w następnym etapie kolumienkę przemywa się za pomocą roztworu etanolu o stężeniu 75% - 98% w sumarycznej ilości około 500 - 1200 µl i ponownie wiruje lub podłącza do pompy próżniowej na 1 - 2 minuty, celem przesączenia przez kolumienkę jej zawartości, po czym w ostatnim etapie ssDNA związane ze złożem kolumienki eluuje się poprzez naniesienie na jej powierzchnię od 10 - 100 µl wody, a następnie kolumienkę wiruje się.
