Abstract

Przedmiotem zgłoszenia są sekwencje starterów pozwalające na analizę poziomu ekspresji genów w korzeniach jęczmienia oraz sposób identyfikacji genotypów jęczmienia o podwyższonej tolerancji na kadm. Bardziej szczegółowo zgłoszenie dotyczy starterów do identyfikacji genów o identyfikatorach HORVU.MOREX.r2.7HG0595000, HORVU.MOREX.r2.2HG0141160, HORVU.MOREX.r2.6HG0487860, HORVU.MOREX.r2.5HG0366120, HORVU.MOREX.r2.7HG0607750, kodujących odpowiednio czynnik transkrypcyjny szoku cieplnego, białko szoku cieplnego, białko oporności na choroby, białko lektyny podobne do konkanawaliny A oraz izomerazę glukozo-6-fosforanową. Dla każdego z genów zaprojektowano startery pozwalające na określenie ich poziomu ekspresji w korzeniach jęczmienia z użyciem zaproponowanego systemu badania poziomu ekspresji genów. Prezentowane zgłoszenie pozwoli na selekcję odmian roślin pod kątem tolerancji na stres kadmu, przez co możliwe będzie wykorzystanie w hodowli takich form, które nie będą akumulowały w swoich tkankach metali ciężkich.The subject of the application are primer sequences enabling the analysis of gene expression levels in barley roots and a method of identifying barley genotypes with increased tolerance to cadmium. More specifically, the application concerns primers for identifying genes with the identifiers HORVU.MOREX.r2.7HG0595000, HORVU.MOREX.r2.2HG0141160, HORVU.MOREX.r2.6HG0487860, HORVU.MOREX.r2.5HG0366120, HORVU.MOREX.r2.7HG06077 50, encoding heat shock transcription factor, heat shock protein, disease resistance protein, concanavalin A-like lectin protein, and glucose-6-phosphate isomerase, respectively. Primers were designed for each gene to determine their expression level in barley roots using the proposed gene expression level testing system. The presented application will allow the selection of plant varieties in terms of tolerance to cadmium stress, which will make it possible to use in breeding forms that will not accumulate heavy metals in their tissues.
