Abstract
Przedmiotem zgłoszenia są nowe trójskładnikowe kompleksy fenamatów o wzorze ogólnym [Ag(L)(fen)<sub>2</sub>]•nH<sub>2</sub>O, w którym Ag oznacza jon srebra, L oznacza ligand podstawowy w postaci anionu kwasu N-fenyloantranilowego C<sub>13</sub>H<sub>10</sub>NO<sub>2</sub> albo anionu kwasu mefenamowego C<sub>15</sub>H<sub>14</sub>NO<sub>2</sub> albo anionu kwasu niflumowego C<sub>13</sub>H<sub>8</sub>F<sub>3</sub>N<sub>2</sub>O<sub>2</sub>, fen oznacza koligand w postaci 1,10-fenantroliny C<sub>12</sub>H<sub>8</sub>N<sub>2</sub>, zaś n wynosi co najwyżej 5. Zgłoszenie obejmuje także sposób otrzymywania trójskładnikowych kompleksów fenamatów, który prowadzi się tak, że przygotowuje się wodny roztwór formy jonowej liganda podstawowego, tak, że do naważki kwasu N-fenyloantranilowego albo kwasu mefenamowego albo kwasu niflumowego dodaje się wodę destylowaną, przy czym na 1 g kwasu stosuje się od 280 do 380 cm<sup>3</sup> wody i tworzy się zawiesinę wodną, do której wprowadza się roztwór NaOH w stosunku molowym kwasu do NaOH 1:1-1,1. Mieszaninę umieszcza się w łaźni wodnej ultradźwiękowej i przetrzymuje się w niej aż do całkowitego roztworzenia zawiesiny, po czym roztwór przenosi się ilościowo do kolby, uzupełnia się wodą do objętości kolby i miesza się dokładnie. Następnie przygotowuje się dwuskładnikowy kompleks 1,10-fenantroliny oraz jonów srebra(I), przy czym do kolby okrągłodennej z czujnikiem temperatury umieszczonej w płaszczu grzewczym z mieszadłem magnetycznym wprowadza się wodny roztwór azotanu(V) srebra i ogrzewa się go do temperatury z przedziału od 58°C do 62°C, a następnie dodaje się do niego naważkę 1,10-fenantroliny, przy czym stosunek jonów srebra(I) do 1,10-fenantroliny stosuje się 1:2-2,1. Zawartość kolby miesza się z szybkością od 100 do 110 obr./min przez czas od 10 do 12 minut, następnie do kolby wprowadza się, ogrzany do temperatury z zakresu od 60°C do 62°C, wodny roztwór formy jonowej liganda podstawowego. Stosunek jonów srebra(I) do anionu kwasu stosuje się 1:1-1,1. Zawartość kolby miesza się z prędkością od 100 do 110 obr./min w czasie od 15 do 16 minut w temperaturze od 60°C do 62°C. Mieszaninę reakcyjną pozostawia się w ciemnym miejscu w temperaturze pokojowej na 24 godziny, po czym z mieszaniny wyizolowuje się osad, który następnie przemywa się od 4 do 5 razy wodą i pozostawia się w ciemnym miejscu w temperaturze pokojowej do wysuszenia do stałej masy. Przedmiotem zgłoszenia jest także zastosowanie trójskładnikowych kompleksów fenamatów jako środków o działaniu bakteriobójczym lub bakteriostatycznym względem szczepów bakterii Gram-ujemnych, zwłaszcza Escherichia coli ATCC 10536 oraz Klebsiella pneumoniae ATCC 13883 oraz Gram-dodatnich, zwłaszcza Staphylococcus aureus ATCC 6538, Staphylococcus epidermidis ATCC 12228, Staphylococcus epidermidis ATCC 35984, Listeria monocytogenes SLR 2249.The subject of the application are new three-component complexes of fenamates with the general formula [Ag(L)(phen)<sub>2</sub>]•nH<sub>2</sub>O, in which Ag is a silver ion, L is a basic ligand in the form of the N-phenylantranilic acid anion C<sub>13</sub>H<sub>10</sub>NO<sub>2</sub> or the mefenamic acid anion C<sub>15</sub>H<sub >14</sub>NO<sub>2</sub> or the niflumic acid anion C<sub>13</sub>H<sub>8</sub>F<sub>3</sub>N<sub> 2</sub>O<sub>2</sub>, phen means coligand in the form of 1,10-phenanthroline C<sub>12</sub>H<sub>8</sub>N<sub>2</ sub>, and n is at most 5. The application also covers a method for obtaining three-component complexes of fenamates, which is carried out in such a way as to prepare an aqueous solution of the ionic form of the basic ligand, so that an amount of N-phenylanthranilic acid, mefenamic acid or niflumic acid is added to distilled water is used, with 280 to 380 cm<sup>3</sup> of water per 1 g of acid and an aqueous suspension is created into which NaOH solution is introduced in a molar ratio of acid to NaOH of 1:1-1, 1. The mixture is placed in an ultrasonic water bath and kept in it until the suspension is completely dissolved, then the solution is quantitatively transferred to a flask, topped up with water to the volume of the flask and mixed thoroughly. Then, a two-component complex of 1,10-phenanthroline and silver(I) ions is prepared. An aqueous solution of silver nitrate(V) is introduced into a round-bottomed flask with a temperature sensor placed in a heating jacket with a magnetic stirrer and heated to a temperature ranging from 58°C to 62°C, and then an amount of 1,10-phenanthroline is added to it, with the ratio of silver(I) ions to 1,10-phenanthroline being 1:2-2.1. The contents of the flask are mixed at a speed of 100 to 110 rpm for 10 to 12 minutes, then an aqueous solution of the ionic form of the basic ligand, heated to a temperature ranging from 60°C to 62°C, is introduced into the flask. The ratio of silver(I) ions to acid anion is 1:1-1.1. The contents of the flask are stirred at a speed of 100 to 110 rpm for 15 to 16 minutes at a temperature of 60°C to 62°C. The reaction mixture is left in a dark place at room temperature for 24 hours, after which a precipitate is isolated from the mixture, which is then washed 4 to 5 times with water and left in a dark place at room temperature to dry to constant weight. The subject of the application is also the use of three-component complexes of fenamates as agents with bactericidal or bacteriostatic activity against strains of Gram-negative bacteria, especially Escherichia coli ATCC 10536 and Klebsiella pneumoniae ATCC 13883, and Gram-positive bacteria, especially Staphylococcus aureus ATCC 6538, Staphylococcus epidermidis ATCC 12228, Staphylococcus epidermidis ATCC 35984, Listeria monocytogenes SLR 2249.
