Abstract
Deproteinizator przepływowy, zawierający przewód doprowadzający próbkę ciekłą zawierającą białko, komorę deproteinizacji oraz przewód wyprowadzający ciekłą próbkę zdeproteinizowaną, charakteryzuje się tym, że działa w trybie strąceniowym i jest wyposażony w układ wibracyjny zapewniający możliwość wytrząsania zawartości komory deproteinizacji (2), która ma objętość 1,2 - 1,8 cm<sup>3</sup>, korzystnie 1,5 cm<sup>3</sup>, i kształt graniastosłupa lub odwróconego naboju zorientowanego w osi pionowej, którego wysokość jest większa niż przekątna podstawy, dzięki czemu możliwe jest łatwe samorzutne rozdzielenie frakcji odbiałczonej od frakcji zawierającej osad białkowy, przy czym deproteinizator jest połączony z układem analitycznym za pomocą elastycznych przewodów umożliwiających wprowadzanie go w wibracje, a także zawiera układ odprowadzający próbkę zdeproteinizowaną, układ odprowadzający strącony osad zawierający białko, układ czyszczenia komory deproteinizacji oraz układ odpowietrzania, przy czym wszystkie kanały doprowadzające i odprowadzające wyposażone są w sterowane elektronicznie zawory, a także, korzystnie, układ optyczny pozwalający na pomiar mętności deproteinizowanej próbki. Sposób deproteinizacji ciekłych próbek biologicznych wykorzystujący reakcję denaturacji białek za pomocą czynnika wytrącającego, charakteryzuje się tym, że reakcję denaturacji białka zawartego w próbce biologicznej inicjuje się wprowadzeniem czynnika strąceniowego i prowadzi się w deproteinizatorze przepływowym opisanym powyżej. Deproteinizator przepływowy umożliwiający odbiałczenie próbek biologicznych w trybie przepływowym, pozwala na skrócenie czasu analizy i uproszczenie procedury przygotowania próbki w stosunku do odbiałczania przeprowadzanego w trybie manualnym, a także ograniczenie kosztów analizy i eksploatacji układu w stosunku do odbiałczania w trybie przepływowym za pomocą membran.The flow-through deproteinizer, comprising a protein-containing liquid sample supply line, a deproteinisation chamber and a deproteinisation liquid sample discharge line, is characterized in that it operates in a precipitation mode and is equipped with a vibrating system for shaking the content of the deproteinisation chamber (2), which has a volume of 1, 2 - 1.8 cm<sup>3</sup>, preferably 1.5 cm<sup>3</sup>, and the shape of a prism or inverted cartridge oriented in the vertical axis, the height of which is greater than the diagonal of the base, so that it is possible to easily spontaneously separate the deproteinized fraction from the fraction containing the protein sludge, the deproteinizer is connected to the analytical system by means of flexible hoses enabling it to vibrate, and also includes a system for removing the deproteinized sample, a system for removing the precipitated sludge containing protein, a system for cleaning the deproteinization chamber and the venting system, wherein all the inlet and outlet channels are equipped with electronically controlled valves and, preferably, an optical system for measuring the turbidity of the deproteinized sample. A method for deproteinizing liquid biological samples using a protein denaturation reaction with a precipitating agent is characterized in that the denaturation reaction of a protein contained in a biological sample is initiated by the introduction of a precipitating agent and carried out in the flow deproteinizer described above. The flow deproteinizer, which enables the deproteination of biological samples in the flow mode, shortens the analysis time and simplifies the sample preparation procedure in relation to manual deproteinization, as well as reduces the costs of analysis and operation of the system in relation to the flow deproteinization using membranes.
