Abstract

Przedmiotem zgłoszenia jest sposób otrzymywania biologicznego implantu do regeneracji tkanki chrzęstnej, obejmujący etapy: izolację chondrocytów z tkanki chrzęstnej, hodowlę wyizolowanych chondrocytów, umieszczania i prowadzenia hodowli chondrocytów na powierzchni membrany opartej na pochodnej kwasu hialuronowego. Sposób ten charakteryzuje się tym, że w kolejnym etapie prowadzi się wizualizację chondrocytów za pomocą roztworu trypsyny o stężeniu 0.25% i barwnika fluorescencyjnego silnie wiążącego się do DNA oraz przeprowadza się ocenę zagęszczenia chondrocytów na powierzchni membrany opartej na pochodnej kwasu hialuronowego poprzez zastosowanie trawienia enzymatycznego.The subject of the application is a method of obtaining a biological implant for the regeneration of cartilage tissue, comprising the steps of: isolating chondrocytes from cartilage tissue, culturing isolated chondrocytes, placing and culturing chondrocytes on the surface of a membrane based on a hyaluronic acid derivative. This method is characterized in that in the next step chondrocytes are visualized with a trypsin solution with a concentration of 0.25% and a fluorescent dye strongly binding to DNA, and the chondrocyte density on the surface of the membrane based on hyaluronic acid derivative is assessed by enzymatic digestion.
