Przedmiotem zgłoszenia jest sposób wytwarzania kwasu hialuronowego z wykorzystaniem hodowli kultur bakterii zdolnych do produkcji kwasu hialuronowego, obejmujący kolejne etapy: przygotowania inokulum, zaszczepienia fermentora i hodowli bakterii oraz czynności pofermentacyjnych (ang. downstream) charakteryzuje się tym, że: podłoże hodowlane poddaje się zaszczepieniu kulturą bakterii zdolną do produkcji kwasu hialuronowego, następnie zaszczepione podłoże hodowlane namnaża się na wytrząsarce przy obrotach w zakresie od 100 do 250 obr/min, w temperaturze od 30 do 37°C w czasie od 10 do 24 godz.; następnie fermentor wypełnia się podłożem hodowlanym, po czym podłoże hodowlane szczepi się inokulum w objętości od 5 do 20% obj. fermentora, a następnie prowadzi się hodowlę w temp. od 30 do 37°C, przy stężeniu tlenu od 10 do 40%, w czasie od 12 do 24 godz.; a następnie zawiesinę poddaje się wirowaniu na wirówce ciągłej przy obrotach od 10000 do 15000 obr/min, po czym komórki bakteryjne zawiesza się w wodzie dejonizowanej z dodatkiem od 0,05 do 0,5% wag. detergentów przy zastosowaniu ultradźwięków w czasie od 0,5 do 2 godz., po czym ponownie wiruje się, a uzyskany supernatant filtruje się na membranach do filtracji stycznej o wielkości porów w zakresie od 2 do 10 kDa, po czym filtrat schładza się do temperatury od 4 do 10°C, a następnie dodaje się rozpuszczalnik organiczny o temperaturze od -20 do -10°C w objętości od 100 do 300% obj. w stosunku do filtratu, po czym mieszaninę filtratu i rozpuszczalnika organicznego pozostawia się w temperaturze od -20 do -10°C na czas od 12 do 24 godz., a następnie zawiesinę kwasu hialuronowego odwirowuje się przy obrotach od 5000 do 10000 obr/min, po czym otrzymany osad kwasu hialuronowego rozpuszcza się w wodzie (123) o temperaturze od 40 do 70°C oraz filtruje, a następnie filtrat schładza się do temperatury od 4 do 10°C oraz dodaje się rozpuszczalnik organiczny o temperaturze od -20 do -10°C w objętości od 100 do 300% obj. w stosunku do filtratu, a następnie mieszaninę filtratu i rozpuszczalnika organicznego wytrąca się w temperaturze od -20 do -10°C w czasie od 12 do 24 godz., po czym osad kwasu hialuronowego suszy się w temperaturze od 20 do 40°C pod ciśnieniem od 5 do 300 mbar.The subject of the application is a method for producing hyaluronic acid using cultures of bacteria capable of producing hyaluronic acid, comprising the following stages: preparation of the inoculum, fermentation of the fermenter and bacterial culture, as well as downstream operations characterized in that: the culture medium is seeded with a culture bacteria capable of producing hyaluronic acid, then the inoculated culture medium is propagated on a shaker at rotations in the range from 100 to 250 rpm, at a temperature of 30 to 37 ° C for 10 to 24 hours; the fermentor is then filled with culture medium, after which the culture medium is inoculated with an inoculum volume of 5 to 20% by volume. fermentor, followed by culturing at a temperature of 30 to 37 ° C, an oxygen concentration of 10 to 40%, for 12 to 24 hours; and then the suspension is centrifuged in a continuous centrifuge at 10,000 to 15,000 rpm, after which the bacterial cells are suspended in deionized water with an addition of 0.05 to 0.5 wt. detergents using ultrasound for 0.5 to 2 hours, then centrifuged again, and the resulting supernatant is filtered on tangential filtration membranes with a pore size in the range of 2 to 10 kDa, after which the filtrate is cooled to a temperature of 4 to 10 ° C, followed by the addition of an organic solvent at a temperature from -20 to -10 ° C in a volume of 100 to 300% vol. relative to the filtrate, after which the mixture of the filtrate and the organic solvent is left at a temperature from -20 to -10 ° C for 12 to 24 hours, and then the hyaluronic acid suspension is centrifuged at 5,000 to 10,000 rpm, then the obtained hyaluronic acid residue is dissolved in water (123) at a temperature of 40 to 70 ° C and filtered, and then the filtrate is cooled to a temperature of 4 to 10 ° C and an organic solvent at a temperature of -20 to -10 is added ° C in a volume of 100 to 300% vol. relative to the filtrate, and then the mixture of filtrate and organic solvent precipitates at a temperature of -20 to -10 ° C for 12 to 24 hours, after which the hyaluronic acid precipitate is dried at a temperature of 20 to 40 ° C under pressure from 5 to 300 mbar.
