Abstract
Przedmiotem zgłoszenia jest sposób wydzielania lizozymu z białka jaja, szczególnie kurzego, który ma zastosowanie w przemyśle spożywczym jako konserwant i w przemyśle farmaceutycznym jako środek bakteriostatyczny. W sposobie otrzymywania lizozymu z białka jaja, białko jaja mieszane jest z roztworem soli fizjologicznej (0.89% NaCl; 0.15 M NaCl), przy czym stosunek soli do wsadu wynosi od 1:1 do 1:6, którą to mieszaninę poddaje się filtracji, przy zastosowaniu membrany mikrofiltracyjnej o porowatości od 0.14 µm do 1.45 µm, a następnie, uzyskany permeat, stanowiący od 50% do 80% nadawy mieszany jest znanymi środkami z żywicą jonowymienną, gdzie w trakcie mieszania, następuje adsorpcja lizozymu, przy czym czas adsorpcji nie może być mniejszy od 0.5 h i większy od 4 h. Po procesie sorpcji żywicę z zaadsorbowanym enzymem oddziela się od roztworu białka, przy czym proces desorpcji umożliwiający zerwanie połączenia żywica - enzym realizowany jest z zastosowaniem roztworów 0.5 ÷ 2.5 M NaCl, a desorbowany roztwór jest odsalany przy pomocy membran ultrafiltracyjnych o granicznym punkcie odcięcia molekularnego 5 ÷ 30 kDa, przy ciśnieniu transmembranowym w zakresie 0.05 ÷ 0.5 MPa a proces ultrafiltracji realizowany jest z diafiltracją, przy czym proces diafiltracji jest prowadzony do momentu zagęszczenia nadawy do 50% wyjściowej objętości, i tak uzyskany retentat jest suszony znanymi metodami lub uzupełniany wodą lub buforem.The subject of the application is a method for the secretion of lysozyme from egg protein, especially chicken, which is used in the food industry as a preservative and in the pharmaceutical industry as a bacteriostatic agent. In the method of obtaining lysozyme from egg protein, egg protein is mixed with physiological saline (0.89% NaCl; 0.15 M NaCl), with a salt to feed ratio of 1: 1 to 1: 6, which is filtered, using a microfiltration membrane with a porosity of 0.14 µm to 1.45 µm, and then, the obtained permeate, constituting from 50% to 80% of the feed is mixed by known means with an ion exchange resin, where during mixing, lysozyme adsorption occurs, while the adsorption time cannot be less than 0.5 h and greater than 4 h. After the sorption process, the resin with the adsorbed enzyme is separated from the protein solution, with the desorption process allowing the resin to break off - the enzyme is carried out using 0.5 ÷ 2.5 M NaCl solutions, and the desorbed solution is desalted ultrafiltration membranes with a molecular cut-off limit point 5 ÷ 30 kDa, at transmembrane pressure in the range of 0.05 ÷ 0 .5 MPa and the ultrafiltration process is carried out with diafiltration, whereby the diafiltration process is carried out until the feed is concentrated to 50% of the initial volume, and the retentate thus obtained is dried by known methods or supplemented with water or buffer.
