Opis wynalazku
Przedmiotem wynalazku są nowe, disulfonamidowe pochodne 5-anilino-7-fenylo-pirazolo[4,3-e][1,2,4]triazyny o wzorze ogólnym 1, sposób ich wytwarzania oraz ich zastosowania związane z aktywnością przeciwnowotworową, cytotoksyczną i/lub cytostatyczną, w szczególności względem komórek nowotworowych, zwłaszcza komórek raka sutka, jak również kompozycje farmaceutyczne je zawierające.
Ze stanu techniki znane są sulfonamidowe pochodne układu 5-anilino-1,3-dimetylo-1/-/-pirazolo[4,3-e][1,2,4]triazyny wykazujące działanie przeciwnowotworowe [M. Mojzych, V. Subertova, A. Bielawska, K. Bielawski, V. Bazgier, K. Berka, T. Gucky, E. Fornal, V. Krystof, Eur. J. Med. Chem., 78, 217 (2014)], jak również znane są sposoby ich syntezy [Pat RP-219242-(2014)].
W patencie nr PL-198965 oraz w literaturze (M. Mojzych, A. Rykowski; Heterocycles, 2004, 63, 1829-1838) opisana jest synteza 5-anilino-1-fenylo-3-metylo-1/-/-pirazolo[4,3-e][1,2,4]triazyny będącej związkiem wyjściowym do syntezy nowych disulfonamidów opisanych wzorem ogólnym 1, natomiast w patencie nr PL-219242 opisana jest synteza i aktywność przeciwnowotworową sulfonamidów pochodnych 5-anilino-1,3-dimetylo-1/-/-pirazolo[4,3-e][1,2,4]triazyny. Pochodne te mają odmienną strukturę od pochodnych będących przedmiotem niniejszego zgłoszenia patentowego.
Celem niniejszego wynalazku było dostarczenie nowych związków nadających się do leczenia nowotworu, zwłaszcza raka sutka, w szczególności związków o działaniu cytotoksycznym, cytostatycznym i antyproliferacyjnym. Celem niniejszego wynalazku było także dostarczenie sposobu wytwarzania takich związków oraz kompozycji farmaceutycznych do leczenia nowotworu, zwłaszcza raka sutka.
Cele te zrealizowano przy pomocy rozwiązań przedstawionych w załączonych zastrzeżeniach patentowych.
Przedmiotem niniejszego wynalazku są nowe disulfonamidowe pochodne 5-anilino-1-fenylo-3-metylo-1 /-/-pirazolo[4,3-e][1,2,4]-triazyny o wzorze ogólnym 1:
w którym R oznacza podstawnik, będący jedną z wymienionych grup: A/-metylopiperazynową, pirolidynową, morfolinową, piperydynową, piperazynową, homopiperazynową, 2-morfolinoetyloaminową, 2-aminoetyloaminową, 2-aminoetanolową, 2-amino-2-metylo-1,3-propanodiolową, 2-amino-2-metylo-1-propanolową, (S)-(+)-1-amino-2-propanolową, (R)-(-)-1-amino-2-propanolową, (S)-(+)-2-amino-1-propanolową, (R)-(-)-2-amino-1-propanolową, (S)-(+)-2-amino-3-metylo-1-butanolową, (R)-(-)-2-amino-3-metylo-1 -butanolową, (S)-(+)-3-amino-1,2-propanediolową, (R)-(-)-2-amino-1,2-propanediolową, (S)-(+)-leucinolową, (R)-(-)-leucinolową, S-(+)-2-amino-1-butanolową, R-(-)-2-amino-1-butanolową, estru metylowego L-proliny i estru metylowego c/s-4-hydroksy-L-proliny.
Przedmiotem niniejszego wynalazku jest również sposób wytwarzania nowych disulfonamidowych pochodnych 5-anilino-1-fenylo-3-metylo-1/-/-pirazolo[4,3-e][1,2,4]-triazyny o wzorze ogólnym 1, innym niż sposoby wytwarzania znane dotychczas w stanie techniki.
Sposób wytwarzania disulfonamidowych pochodnych 5-anilino-1-fenylo-3-metylo-pirazolo[4,3-e][1,2,4]triazyny o wzorze ogólnym 1 według niniejszego wynalazku polega na wprowadzeniu do 5-anilino-1-fenylo-3-metylo-pirazolo[4,3-e][1,2,4]triazyny dwóch identycznych chiralnych podstawników sulfonamidowych. W pierwszym etapie syntezy znaną 5-anilino-1-fenylo-3-metylo1 /-/-pirazolo[4,3-e][1,2,4]triazynę przekształca się w odpowiednią 4-[1 -(4-(chlorosulfonylo)fenylo)-3
PL 240 932 B1 metylo-1 H-pirazolo[4,3-e][1,2,4]-triazyn-5-ylamino]benzeno-1-chlorosulfonu poprzez reakcję z kwasem chlorosulfonowym. W kolejnym etapie tak otrzymaną pochodną dichlorosulfonową poddaje się reakcji z odpowiednią aminą lub enancjomerem chiralnego aminoalkoholu bądź estru aminokwasu. Odpowiednia aminopochodna wybierana jest z grupy: N-metylopiperazyna, pirolidynowa, morfolina, piperydyna, piperazyna, homopiperazyna, 2-morfolinoetyloamina, 2-aminoetyloamina, 2-aminoetanolowa, 2-amino-2metylo-1,3-propanodiol, 2-amino-2-metylo-1-propanol, (S)-(+)-1-amino-2-propanol, (R)-(-)-1-amino-2propanol, (S)-(+)-2-amino-1-propanol, (R)-(-)-2-amino-1-propanol, (S)-(+)-2-amino-3-metylo-1-butanol, (R)-(-)-2-amino-3-metylo-1-butanol, (S)-(+)-3-amino-1,2-propanediol, (R)-(-)-2-amino-1,2-propanediol, (S)-(+)-leucinol, (R)-(-)-leucinol, S-(+)-2-amino-1-butanol, R-(-)-2-amino-1-butanol, ester metylowy L-proliny, i ester metylowy cis-4-hydroksy-L-proliny, w zależności od żądanego produktu końcowego sposobu wytwarzania według wynalazku. W wyniku tej reakcji otrzymuje się odpowiednią disulfonamidawą pochodną o ogólnym wzorze 1.
Otrzymaną tak nową pochodną disulfonamidową 5-anilino-1-fenylo-3-metylo-1 H-pirazolo-[4,3-e][1,2,4]triazyny o wzorze ogólnym 1 według wynalazku, można korzystnie oczyszczać chromatograficznie, na przykład z zastosowaniem chromatografii cieczowej kolumnowej.
Przykładowy sposób wytwarzania reprezentatywnej disulfonamidowej pochodnej 5-anilino-1-fenylo-3-metylo-1H-pirazolo[4,3-e][1,2,4]triazyny o wzorze ogólnym 1 według wynalazku przedstawiono w przykładzie 1. Wzory stosowanych grup aminowych (podstawnik R w ogólnym wzorze 1) obecnych w nowych disulfonamidowych pochodnych 5-anilino-1-fenylo-3-metylo-1 H-pirazolo[4,3-e][1,2,4]triazyny o wzorze ogólnym 1 według wynalazku przedstawiono w załączonej Tabeli 1. W Tabeli 1 podano także odpowiednie temperatury topnienia i wydajności otrzymywania związków według wynalazku (związki numer 1 do 25).
Przedmiotem wynalazku jest ponadto wykorzystanie nowych pochodnych o wzorze ogólnym 1, otrzymanych według niniejszego wynalazku, do zastosowania jako lek do leczenia nowotworu, zwłaszcza do leczenia raka sutka.
Przedmiotem wynalazku jest również wykorzystanie nowych pochodnych o wzorze ogólnym 1, otrzymanych według niniejszego wynalazku, do zastosowania jako związki o działaniu cytotoksyczn ym, cytostatycznym i antyproliferacyjnym.
W wyniku przeprowadzonych prób i badań okazało się, że związki według wynalazku, o ogólnym wzorze 1, wykazują działania przeciwnowotworowe, cytotoksyczne, cytostatyczne i antyproliferacyjne względem komórek nowotworowych, w szczególności komórek raka sutka, i mogą być zatem stosowane jako leki, w szczególności leki do leczenia nowotworów, zwłaszcza raka sutka. Dzięki swoim właściwościom związki według wynalazku mogą być także stosowane jako środki cytostatyczne i/lub cytotoksyczne. Wyniki badań biologicznych dla związków według wynalazku przedstawiono w przykładach, w szczególności w Tabeli 2 (związki numer 1 do 25).
Przedmiotem wynalazku jest także kompozycja farmaceutyczna, charakteryzująca się tym, że jako substancję aktywną zawiera nowe disulfonamidowe pochodne 5-anilino-1fenylo-3-metylo-1Hpirazolo[4,3-e][1,2,4]-triazyny o wzorze ogólnym 1, otrzymywane według wynalazku. Substancjami dodatkowymi są farmaceutycznie akceptowalne nośniki, rozcieńczalniki lub zaróbka.
Disulfonamidowe pochodne 5-anilino-1-fenylo-3-metylo-pirazolo[4,3-e][1,2,4]-triazyny o wzorze ogólnym 1 według wynalazku można stosować w zastosowaniach według wynalazku w. postaci kompozycji farmaceutycznych. Kompozycja farmaceutyczna według wynalazku zawiera, oprócz związku według wynalazku jako składnika aktywnego, farmaceutycznie akceptowalny nośnik, rozcieńczalnik lub zaróbkę. Kompozycje farmaceutyczne według wynalazku zawierające jako składnik aktywny co najmniej jeden związek według wynalazku można stosować do leczenia nowotworów, zwłaszcza raka sutka.

Oznaczanie aktywności przeciwnowotworowej nowozsyntetyzowanych disulfonamidowych pochodnych 5-anilino-1-fenylo-3-metylo-1 H-pirazolo[4,3-e][1,2,4]-triazyny o wzorze ogólnym 1 według wynalazku przeprowadzono w warunkach in vitro.
Badania aktywności przeciwnowotworowej przeprowadzono zarówno z wykorzystaniem komórek estrogenozależnego raka sutka MCF-7 jak i komórek estrogenoniezależnego raka sutka MDA-MB-231. W skrócie, na obu liniach komórkowych przeprowadzono pomiar działania cytotoksycznego i cytostatycznego disulfonamidowych pochodnych 5-anilino-1-fenylo-3-metylo-1 H-pirazolo(4,3-e][1,2,4]triazyny o wzorze ogólnym 1 według wynalazku, a także oceniono ich wpływ, na biosyntezę DNA. Pomiar działania cytotoksycznego i cytostatycznego chiralnych pochodnych disulfonamidowych 5-anilino-1-fenylo3-metylo-1 H-pirazolo[4,3-e][1,2,4]triazyny o wzorze ogólnym 1 według wynalazku wykonano metodą Carmichael’a, określając żywotność komórek przy użyciu soli tetrazolowej (MTT) w badaniu MTT. Jest to badanie kolorymetryczne, wykorzystujące aktywność oksydoredukcyjną mitochondriów komórek. W żywych komórkach, pod wpływem mitochondrialnej dehydrogenazy, zastosowany barwnik ulega przekształceniu z żółtej soli tetrazolowej do purpurowego formazanu. Przemiana ta nie zachodzi w martwych komórkach. Żywotność komórek inkubowanych w obecności badanych disulfonamidowych pochodnych 5-anilino-1-fenylo-3-metylo-1 H-pirazolo[4,3-e][1,2,4]triazyny o wzorze ogólnym 1 według wynalazku przedstawiano jako procent wartości kontrolnych. Badanie wpływu pochodnych disulfonamidowych 5-anilno-1-fenylo-3-metylo-1 H-pirazolo[4,3-e][1,2,4]triazyny o wzorze ogólnym 1 według wynalazku na proces biosyntezy DNA przeprowadzono na liniach komórkowych raka sutka, poprzez pomiar wbudowywania [3H]-tymidyny do DNA badanych komórek nowotworowych. Dokładny, przykładowy opis oznaczania aktywności przeciwnowotworowej nowozsyntetyzowanych pochodnych disulfonamidowych 5-anilino-1-fenylo-3-metylo-1 H-pirazolo[4,3-e][1,2,4]triazyny o wzorze ogólnym 1 według wynalazku w warunkach in vitro przedstawiono w przykładzie 2. W Tabeli 2 zestawiono wyniki badania aktywności cytotoksycznej, cytostatycznej i antyproliferacyjnej pochodnych disulfonamidowych 5-anilino-1-fenylo-3-metylo-1 H -pirazolo[4,3-e][1,2,4]triazyny o wzorze ogólnym 1 według wynalazku.
Przeprowadzone badania wykazały, że pochodne 5-anilino-1-fenylo-3-metylo-1 H-pirazolo[4,3-e][1,2,4]triazyny o wzorze ogólnym 1 według wynalazku wykazują zarówno działanie cytostatyczne, jak i działanie cytostatyczne. Przeprowadzone badania wykazały także, iż pochodne disulfonamidowe 5-anilino-1-fenylo-3-metylo-1 H-pirazolo[4,3-e][1,2,4]triazyny o wzorze ogólnym 1 według wynalazku wykazują aktywność przeciwnowotworową, zarówno w przypadku komórek nowotworowych estrogenozależnego raka sutka (MCF-7), jak i w przypadku komórek nowotworowych estrogenoniezależnego raka sutka (MDA-MB-231). Przeprowadzone badania pokazują zatem, że pochodne disulfonamidowe 5-anilino-1-fenylo-3-metylo-1 H-pirazolo[4,3-e][1,2,4]triazyny o wzorze ogólnym 1 według wynalazku można stosować jako leki, zwłaszcza leki przeciwnowotworowe, do leczenia nowotworów, zwłaszcza raka sutka, zarówno estrogenozależnego jak i estrogenoniezależnego. Przeprowadzone badania pokazują również, że pochodne disulfonamidowe 5-anilino-1-fenylo-3-metylo-1 H-pirazolo[4,3-e][1,2,4]triazyny o wzorze ogólnym 1 według wynalazku mogą być stosowane jak środki cytostatyczne, a także jako środki cytotoksyczne.
Niniejszy wynalazek zostanie poniżej zilustrowany za pomocą przykładów wykonania, które nie mają jednak w jakikolwiek sposób ograniczać zakresu ochrony wynalazku, jak zdefiniowano w zastrzeżeniach patentowych. O ile nie wskazano inaczej, stosowano znane i/lub komercyjnie dostępne urządzenia, metody, parametry, warunki reakcji, odczynnik i zestawy, takie jakie powszechnie stosuje się w dziedzinie, do której należy niniejszy wynalazek, i jakie są zalecane przez wytwórców odpowiednich odczynników i zestawów.

Sposób wytwarzania 4-[1-(4-(chlorosulfonylo)fenylo)-3-metylo-1 H-pirazolo[4,3-e][1,2,4]-triazyn-5-ylamino]benzeno-1 -chlorosulfonu
Do kolby zawierającej 3 ml kwasu chlorosulfonowego ochłodzonego w mieszaninie wody z lodem do temperatury 0-5°C dodano 5-anilino-1-fenylo-3-metylo-1 H-pirazolo[4,3-e][1,2,4]triazynę (604 mg, 2 mmola). Początkowo reakcję prowadzono w temperaturze 0-5°C, a następnie w temperaturze pokojowej przez 2 godziny. Po zakończeniu reakcji mieszaninę reakcyjną wylano na wodę z lodem i ekstrahowano chlorkiem metylenu (4 x 50 ml). Połączone ekstrakty suszono nad bezwodnym MgSO4. Po odparowaniu chlorku metylenu na wyparce surowy produkt oczyszczono na kolumnie chromatograficznej, stosując chlorek metylenu jako eluent. Produkt otrzymuje się w postaci żółtego osadu.

Sposób wytwarzania disulfonamidów
Dichlorosulfon (100 mg, 0,28 mmol) rozpuszczono w bezwodnym acetonitrylu (5 ml) i dodano 3,5-krotny nadmiar odpowiedniej aminy lub chiralnego aminoalkoholu, lub pochodnych L-proliny (1 mmol). Reakcję prowadzono w temperaturze pokojowej do całkowitego zaniku substratu (kontrola TLC). Po zakończeniu reakcji rozpuszczalnik odparowano, a surowy produkt reakcji oczyszczono na kolumnie chromatograficznej, stosując jako eluent mieszaninę CH2Cl2/EtOH (25 : 1). Otrzymane w ten sposób sulfonamidy przedstawia Tabela 1.

Komórki estrogenozależnego raka sutka MCF-7 (ATCC, Manassas, VA, USA) hodowano w pożywce DMEM (Dulbecco's Minimal Essential Medium) z dodatkiem 10% płodowej surowicy bydlęcej (FBS), 2 mmol/dm3 glutaminy, 50 U/cm3 penicyliny, 50 μg/cm3 streptomycyny w temperaturze 37°C, w atmosferze zawierającej 5% CO2. Następnie komórki liczono w hemocytometrze i przenoszono w ilości 1 x 106 komórek na „oczko” w 1 ml pożywki hodowlanej z butelek „Costar” (Sigma, USA), w których prowadzono hodowlę, do 6-oczkowych płytek „Nunc” (Wiesbaden, Niemcy). Do przenoszenia komórek wykorzystano bezwapniowy bufor fosforanowy z dodatkiem 0,05% trypsyny i 0,02% EDTA. Po 48 godzinach pożywkę hodowlaną wymieniono na pożywkę DMEM, która nie zawierała czerwieni fenolowej i płodowej surowicy bydlęcej (FBS). Tak przygotowane komórki inkubowano przez 24 godziny z badanymi związkami i stosowano do dalszych doświadczeń.
Komórki estrogenoniezależnego raka sutka MDA-MB-231 (ATCC, Manassas, VA, USA) hodowano w pożywce DMEM (Dulbecco's Minimal Essential Medium) z dodatkiem 10% płodowej surowicy bydlęcej (FBS), 2 mmol/dm3 glutaminy, 50 U/cm3 penicyliny, 50 μg/cm3 streptomycyny w temperaturze 37°C, w atmosferze zawierającej 5% CO2. Następnie komórki liczono w hemocytometrze i przenoszono w ilości 1 x 106 komórek na „oczko” w 1 ml pożywki hodowlanej z butelek „Costar” (Sigma, USA), w których prowadzono hodowlę, do 6-oczkowych płytek „Nunc” (Wiesbaden, Niemcy). Do przenoszenia komórek wykorzystano bezwapniowy bufor fosforanowy z dodatkiem 0,05% trypsyny i 0,02% EDTA. Po 48 godzinach pożywkę hodowlaną wymieniono na pożywkę DMEM, która nie zawierała czerwieni fenolowej i płodowej surowicy bydlęcej (FBS). Tak przygotowane komórki inkubowano przez 24 godziny z badanymi związkami i użyto do dalszych doświadczeń.
Pomiar cytotoksyczności badanych związków wykonano metodą Carmichael'a (J. Carmichael et al, Cancer Res., 1987, 47, 943-946), określając żywotność komórek przy użyciu soli tetrazolowej (MTT) w teście MTT. W żywych komórkach, pod wpływem mitochondrialnych dehydrogenaz, zastosowany barwnik ulega przemianie do purpurowego formazanu. Przemiana ta nie zachodzi w martwych komórkach. Komórki inkubowano 24 godziny w inkubatorze zapewniającym odpowiednie warunki, tj. w temperaturze 37°C, w atmosferze zawierającej 5% CO2 wraz z pożywką zawierającą różne stężenia badanych związków. Po 24-godzinnej inkubacji komórki płukano trzy razy (3 x 1 ml) PBS. Następnie dodano 1 ml PBS oraz 50 μl MTT o stężeniu 5 mg/cm3 PBS i kontynuowano inkubację przez 4 godziny. Po upływie wymaganego czasu podłoże usunięto, a do komórek dodano 1 ml 0,1 M kwasu solnego w absolutnym izopropanolu i komórki odstawiono na 10 minut. W tak przygotowanymi lizacie komórek mierzono absorbancję przy długości fali 630 nm. Wynik absorbancji uzyskany w hodowlach komórek kontrolnych przyjęto za 100%. Natomiast żywotność komórek inkubowanych w obecności badanych związków przedstawiono jako procent wartości kontrolnych. Poziom istotności p wyznaczono nieparametrycznym testem U Manna-Whitney’a za pomocą programu SigmaStat. Wyniki przy poziomie istotności p < 0,05 uznano za istotne statystycznie. Wartości IC50 (ang. inhibitory concentration 50 - stężenie powodujące 50-procentową redukcję przeżywalności komórek) dla linii MCF-7 i MDA-MB-231 wyznaczono na podstawę analizy zależności zastosowanych stężeń badanych związków i stopnia przeżywalności komórek.
Ponadto oceniono wpływ związków według wynalazku na proces biosyntezy DNA poprzez pomiar wbudowywania [3H]-tymidyny do DNA badanych komórek. Komórki inkubowano 24 godziny w inkubatorze zapewniającym odpowiednie warunki, tj. w temperaturze 37°C, w atmosferze zawierającej 5% CO2 wraz z pożywką zawierającą różne stężenia badanych związków. The subject of the invention is new disulfonamide derivatives of 5-anilino-7-phenyl-pyrazolo [4,3-e] [1,2,4] triazines of the general formula 1, their preparation process and their applications related to antitumor, cytotoxic and / or cytostatic, in particular towards neoplastic cells, especially breast cancer cells, as well as pharmaceutical compositions containing them.
Sulfonamide derivatives of the 5-anilino-1,3-dimethyl-1H-pyrazolo [4,3-e] [1,2,4] triazine system showing antitumor activity are known from the prior art [M. Mojzych, V. Subertova, A. Bielawska, K. Bielawski, V. Bazgier, K. Berka, T. Gucky, E. Fornal, V. Krystof, Eur. J. Med. Chem., 78, 217 (2014)], as well as methods of their synthesis are known [Pat RP-219242- (2014)].
In the patent no PL-198965 and in the literature (M. Mojzych, A. Rykowski; Heterocycles, 2004, 63, 1829-1838) the synthesis of 5-anilino-1-phenyl-3-methyl-1 / - / - pyrazolo [ 4,3-e] [1,2,4] triazine being a starting compound for the synthesis of new disulfonamides described by the general formula 1, while patent No. PL-219242 describes the synthesis and antitumor activity of 5-anilino-1,3-dimethyl sulfonamides -1 / - / - pyrazolo [4,3-e] [1,2,4] triazines. These derivatives have a structure different from the derivatives that are the subject of the present patent application.
The object of the present invention was to provide new compounds suitable for the treatment of cancer, especially breast cancer, in particular compounds with cytotoxic, cytostatic and antiproliferative activity. It was also an object of the present invention to provide a method for the preparation of such compounds and pharmaceutical compositions for the treatment of cancer, especially breast cancer.
These aims have been achieved with the solutions set out in the appended claims.
The present invention relates to new disulfonamide derivatives of 5-anilino-1-phenyl-3-methyl-1H-pyrazolo [4,3-e] [1,2,4] -triazine of the general formula 1:

As a result of the tests and studies carried out, it has been found that the compounds according to the invention, of the general formula I, have antitumor, cytotoxic, cytostatic and antiproliferative effects on neoplastic cells, in particular breast cancer cells, and can therefore be used as medicaments, in particular medicaments for the treatment of cancers, especially breast cancer. Due to their properties, the compounds according to the invention can also be used as cytostatic and / or cytotoxic agents. The biological test results for the compounds according to the invention are shown in the examples, in particular in Table 2 (compounds Nos. 1 to 25).

The tests carried out thus show that the 5-anilino-1-phenyl-3-methyl-1H-pyrazolo [4,3-e] [1,2,4] triazine disulfonamide derivatives of the general formula 1 according to the invention can be used as medicaments, especially anti-tumor drugs, for the treatment of tumors, especially breast cancer, both estrogen-dependent and estrogen-dependent. The tests carried out also show that the 5-anilino-1-phenyl-3-methyl-1H-pyrazolo [4,3-e] [1,2,4] triazine disulfonamide derivatives of the general formula 1 according to the invention can be used as agents cytostatic, and also as cytotoxic agents.

The cytotoxicity of the test compounds was measured by the method of Carmichael (J. Cells were incubated for 24 hours in an incubator that provided the appropriate conditions, ie at 37 ° C, in an atmosphere containing 5% CO2, along with a medium containing various concentrations of test compounds. After 24-hour incubation, cells were washed three times (3 x 1 ml) with PBS. Then 1 ml of PBS and 50 μl of MTT at a concentration of 5 mg / cm 3 PBS were added and the incubation continued for 4 hours. Cells were incubated for 24 hours in an incubator that provided the appropriate conditions, ie at 37 ° C, in an atmosphere containing 5% CO2, along with a medium containing various concentrations of test compounds. Test compounds were added to the culture medium and incubated for 24 hours. After this time, the medium was replaced with fresh medium and 0.5 μCi [ 3 H] -thymidine (specific activity 6.7 Ci / mmol) was added to each hole and the cells were further incubated for 4 hours in an incubator with 5% CO2 at temperature 37 ° C.
