Poligraf. UP PRL. Naklad 120 egz.Cena 100 zl PLThe subject of the invention is a method of producing raw melanins from post-rectification spirit, intended for pharmaceutical, fodder or food additives. Melamine compounds have the ability to protect animal organisms against ultraviolet and penetrating radiation, and above all, they can inhibit the growth of tumors. So far, no known method of biomass production is known. rich in melanin from the byproduct of the spirit industry - the "foreruns" fraction. Known from the publication in the journal "Fiermientnaja i Spirtowaja Promyszlennost" from 1975, number 8 pages 25-27 the method of cultivating yeast - not containing melanins with the use of correctional alcohol, it consisted of adding of spirit to a decoction of 0.5% vol. and yeast multiplication at a temperature of 33-35 ° C at a pH of 4.5 to 5.5. The yield of biomass was 16.5 g / dm3 and the yield in relation to the porectflkate did not exceed 66%, while the spirit used was the second source of carbon in the substrate next to the assimilated components of the decoction. In turn, there are various synthetic substrates for yeast cultivation with the use of raw materials not conventional - alcohol. An example of a nutrient medium is described in the Polish patent specification 91042. Moreover, the commonly known methods of culturing microorganisms consist in the multiplication of a specific strain in optimal conditions, i.e. a precisely defined temperature, pH, concentration of the substrate being the source of energy, specified time, aeration and even the speed of the stirrer rotation. is to develop a method of producing melanins using the "foreruns" fraction as the basic raw material, which is a by-product of the spirit industry. The essence of the invention is that, unlike known methods of microbial cultivation, the temperature and pH are simultaneously and continuously lowered during the multiplication process. The method according to the invention consists in the fact that the synthetic substrate enriched with porectification spirit with a concentration of 0.01-0.1% by volume is inoculated with black yeast Torula jaapi in an amount not lower than 1 g / dm3 of the substrate at an initial temperature of 34 ° C, which During cultivation, it drops to a temperature of 28 ° C, and at the same time the pH changes smoothly from 4.4 to 3.8, cultivating cultures for no more than 45 hours, with a simultaneous smooth increase in aeration to 200% of the initial value, and then it separates in the known method of multiplied melanin biomase from the substrate.2 114875 The advantage of the method according to the invention is the possibility of using a by-product of the spirit industry - the "forerun" fraction, which so far has found its main application in the production of denatured alcohol. The method according to the invention is explained in more detail by an example that does not limit its scope. Example. Synthetic substrate with the following composition: (NH4) 2SO4 - 2.0g, (NH4) 2HP04 - 1.1 g, CO (NH2) 2 - 0.3g, K2HP04 - 0.22g, NaCl - 0.05g, • 7H20 - 0 , 2g, CaCl2-6H20 - 0.05g, H3P04 85% - 0.6cm, FeSO-7H20 - 0.001 g and "Difco" yeast extract - 0.3 g and tap water up to 1 dm3 volume are inoculated with Torulajaapi yeast in the amount of 2 g of dry matter per 1 dm3 of nutrient solution in a fermenter, vol. 3dm3 by "Biotec". It introduces post-correction spirit - 3 cm3, regulates the pH to 4.4 with NH4OH and H * P04 and the temperature of 34 ° C, and the aeration - 4.5 dm3 (min) of 3dm3 of nutrient solution. The cultivation is carried out for 45 hours, gradually lowering the temperature to 28 ° C, and the pH to 3.8. At the same time, the aeration is increased to 9 dm3 (min) 3dm3 of the substrate and after 45 hours the yield of raw melanin biomass 17g of dry substance from 1dm3 of nutrient solution with 51% efficiency in relation to the spirit used. Next, the biomase is centrifuged in a known manner to obtain a raw melanin preparation. Patent claim A method of producing raw melanins from post-rectification spirit in a synthetic medium using "black yeast **, characterized by the fact that the synthetic medium is enriched with correction spirit at a concentration of 0.01-0, 1% by volume of yeast is inoculated with Torulajaapi in an amount of not less than 1 g / dm3 of the substrate at an initial temperature of 34 ° C, which drops smoothly during cultivation to 28 ° C, and at the same time the pH smoothly changes from 4.4 to 3.8 cultivating for a period not longer than 45 hours, with a simultaneous smooth increase in aeration to 200% of the initial value, after which the multiplied melanin biomase is separated from the substrate in a known manner.
