Abstract
A method of isolating a cytoplasmic fraction from an oocytes or an embryonic cell without impairing the capacity of the oocytes or embryonic cell to be fertilized includes the step of releasing a cytoplasmic fraction from the oocytes or embryonic cell which is about 5% of the volume of the oocytes or embryonic cell. The method can further comprise the steps of inserting a releasing means into the oocytes of embryonic cell, then drawing a cytoplasmic fraction into the releasing means and withdrawing the releasing means so that the fraction is isolated in the releasing means. The releasing means may be an injection pipette. Also described is a method for detecting a nucleotide sequence, polymorphism or mutation in the mitochondrial genome of the mitochondria located in the oocytes or embryonic cell without impairing its ability to be fertilized. This method comprises the steps of isolating a cytoplasmic fraction that includes mitochondria according to the above method and then analyzing the nucleotide sequence of the mitochondrial genome for the presence of a nucleotide sequence, polymorphism or mutation in the mitochondrial genome.
