The present invention relates to a blood collecting tube suitable for use with liquid separating agents ;used: for separating components in liquids consisting of more than one component of different specific gravities.
The blood collecting tube of the invention is suitable for use with a liquid separating agent used to fractionate a specific component from other components using the specific gravity difference between the specific component and other components of a liquid to be fractionated and by being interposed between those of said components which is flowable during centrifugal separation, and is not flowable and stable after the operation of centrifugal separation is completed. A suitable liquid separating agent is described in our New Zealand patent specification Number 19 4,868.
The present invention provides a blood collecting tube containing therein a blood coagulation-promoting medium,. characterized in that said blood coagulation-promoting medium comprises a carrier having a specific gravity higher than that of serum, and a powdered blood coagulation-promoting agent supported on the carrier.
In the accompanying drawings Fig. 1 is a perspective view illustrating the blood collecting tube of the invention encapsulating therein a liquid separating agent as a blood serum separator. Fig. 2 is a sectional view of the blood collecting tube shown in Fig. 1 after centrifugal separation.
The blood collecting tube 2, which is suitably formed of glass, is shown in the drawings containing about 1.7 ml of a liquid separating agent composition 1. The tube has a capacity of about 10cc. A polyester unwoven cloth 3 coated with 1 - 3 mg of diatomaceous earth (e.g., Caper Flattery Sand, trade name WG-200, of Kyoritsu Ceramic Materials Co.,
Ltd.), is positioned at a slant in the blood collecting tube.
The tube can be stopped with a butyl rubber plug 4.

In use the tube is placed under reduced pressure and a blood sample is placed in the blood collecting tube and allowed to stand for 7 to 8 minutes. As a result, the diatomaceous earth disperses in the blood upon the introduction of the blood, and it accelerates blood coagulation together with the unwoven cloth 3. Adequate coagulation is thus attained in a short time. Each blood collecting tube is then placed in a centrifuge for 10 minutes at 700 - 1,000 G, and the liquid separating agent compositions become stably distributed between the serum and the clot layers. This state is shown in Fig. 2. Because the liquid separating agent is thixotropic and has a specific gravity between that of the blood serum 5 and that of the blood clots 6, it stays between the blood serum 5 and blood clot 6, forming a gel that separates these two layers. Since the diatomaceous earth and the unwoven cloth 3 have higher specific gravities, they are not included in the layer of blood serum 5. Thus, blood serum 5 obtained is of high purity with no entrainment of fibrin. This blood serum 5 is readily collected from the blood collecting tube by decantation or by suction.

Claims (4)
1. A blood collecting tube containing therein a blood coagulation-promoting medium, characterized in that said blood coagulation-promoting medium comprises a carrier having a specific gravity higher than that of serum, and a powdered blood coagulation-promoting agent supported on the carrier.
2. A blood collecting tube as claimed in Claim 1 wherein said powdered blood coagulation-promoting agent comprises diatomaceous earth.
3. A blood collecting tube as claimed in either of Claims 1 pr 2 wherein said carrier comprises a polyester mflwoven cloth.
