Abstract
Det er beskrevet en fremgangsmåte for å bestemme om en bestemt base er tilstede ved en spesifisert posisjon i en nukleinsyresekvens. Fremgangsmåten omfatter trinnene (a) tilveiebringing av en enkelttrådet prøve av sekvensen som skal bli undersøkt ("prøvesekvens") i immobilisert form på en fast bærer, (b) hybridisering av en primer til nevnte prøvesekvens slik at basen i 3' enden av primeren blir hybridisert til den basen i prøvesekvensen som er rett ved siden av den spesifiserte posisjonen på 3' siden derav, (c) behandling av primeren under ekstensjonsbetingelser med en merket del som har evne til å tilveiebringe en ekstensjonsenhet av primeren i en posisjon som tilsvarer nevnte spesifiserte posisjon i prøvesekvensen dersom nevnte bestemte base er tilstede i den posisjonen, i det ekstensjonsbetingelsene er slik at en hvilke som helst ekstensjon av primeren ikke fortsetter utover nevnte spesifiserte posisjon av prøvesekvensen på annen måte enn ved ekstensjon med merket del, (d) vasking av bæreren for å fjerne ureagert merket del og andre reagenser, og (e) testing for innkorporering av merket del i primeren.
