Abstract

Foreliggende oppfinnelse til- veiebringer for første gang isolert, i det vesentlige rent og/eller rekombin- ant fremstilt pANCA forbundet med ul- cerøs kolitt (UCpANCA og UCpANCA- materiale) så vel som polynukleotider som koder for UCpANCA, UCpANCA-materi- ale og UCpANCA-polypeptider ifølge foreliggende oppfinnelse. Ved å benytte fagfremvis- ningsteknikken har immunoglobulingen- repertoarer anriket på immunoglobulin- gener som benyttes av pANCA-seroposi- tive UC-pasienter, blitt isolert, amplifisert, tilfeldig rekombinert for dannelse av fagmidekspresjonsvektor- biblioteker som koder for DNA-homologer fra genrepertoaret. Fremgangsmåter til- veiebringes for anrikning av disse biblioteker slik at det dannes biblio- teker av VH- og VL-kodende DNA-homologer med immunoreaktivitet overfor UCpANCA- antigen. Videre tilveiebringes frem- gangsmåter for gjennomsøking av biblio- tekene ifølge foreliggende oppfinnelse for UCpANCA og UCpANCA-materiale. Disse biblioteker kan innkapsles i fagpartik- ler eller i celler. Fremgangsmåter og analysesett tilveiebringes også for analyse av UCpANCA i en prøve og isolering av UCpANCA-antigen.The present invention provides for the first time isolated, substantially pure and / or recombinantly produced pANCA associated with ulcerative colitis (UCpANCA and UCpANCA material) as well as polynucleotides encoding UCpANCA, UCpANCA material and UCpANCA polypeptides of the present invention. Using the phage display technique, immunoglobulin gene repertories enriched with immunoglobulin genes used by pANCA seropositive UC patients have been isolated, amplified, randomly recombined to form phagemid expression vector libraries encoding DNA homologs from gene homologs. Methods are provided for enriching these libraries to form libraries of VH and VL coding DNA homologs with immunoreactivity to UCpANCA antigen. Further, methods for scanning the libraries of the present invention for UCpANCA and UCpANCA material are provided. These libraries can be encapsulated in phage particles or cells. Methods and assays are also provided for analysis of UCpANCA in a sample and isolation of UCpANCA antigen.
