Description
Denne oppfinnelse vedrører en rekke cykloalkylsubstituerte glutaramidderivater som er diuretica og som har anvendelse på et stort terapeutisk område som omfatter behandling av forskjellige kardiovaskulære forstyrrelser så som hypertensjon, hjertesvikt og renal svikt.
Forbindelsene er nemmere av sink-avhengig, nøytral endopeptidase É.C.3.4.24.11. Dette enzym deltar i nedbrytelsen av forskjellige peptidhormoner inklusive ANF ("artrial natriuretic factor") som utskilles av hjertet og som har potent vasodilatorisk, diuretisk og natriuretisk aktivitet. Således kan forbindelsene i oppfinnelsen ved å hemme den nøytrale endopeptidase E.C.3.4.24.11, forsterke den biologiske virkning av ANF, og spesielt er forbindelsene diuretica som har anvendelse ved behandling av forskjellige forstyrrelser inklusive hypertensjon, hjertesvikt, angina, renal svikt, premenstrual syndrom, cyklisk oedema, Menieres's sykdom, hyperaldosteronisme (primær og sekundær), pulmonær oedema, ascites og hypercalciuremi. I tillegg har forbindelsene på grunn av sin evne til å forsterke virknings av ANF anvendelse i behandlingen av glaucoma. Som et ytterligere resultat av sin evne til å hemme den nøytrale endopeptidase E.C.3.4.24.11kan forbindelsene i oppfinnelsen ha virkning på andre terapeutiske områder inklusive f.eks. ved behandling av astma, betennelser, smerte, epilepsi, følelsesforstyrrelser, dementia og geriatrisk forvirring, fedme og fordøyelsesforstyrrelser (spesielt diarrhoea og irretabelt tarmsyndrom), modulasjonen av gastrisk syre-sekresjon og behandlingen av hyperreninaemia og leukemi.

I ovenfornevnte definisjon kan, hvis ikke annet er sagt, alkylgrupper som har tre eller flere karbonatomer, være rette og forgrenede. Uttrykket aryl betyr når det er brukt her, en aromatisk hydrokarbongruppe så som fenyl, naftyl eller bifenyl som alternativt kan være substituert f.eks. med en eller flere OH, CN, CF3, C1-C4-alkyl, C1_C4-alkoksy, halo, karbamoyl, aminosulfonyl, amino, mono eller di (C;l_C4-alkyl)amino eller (C1-C4-alkanoyl)aminogrupper. Halo betyr fluor, klor, brom eller jod.
Uttrykket heterocyklyl betyr en 5- eller 6-leddet nitrogen-, oksygen- eller svovelinneholdende heterocyklisk gruppe som, hvis ikke annet er sagt, kan være mettet eller umettet, og .som kan valgfritt inkludere ytterligere et oksygen eller en til tre nitrogenatomer i ringen, og som valgfritt kan være benzosammensmeltet eller substituert med f.eks. en eller flere halo, C1-C4-alkyl, hydroksy, karbamoyl, benzyl, okso, amino eller mono eller di-(C1-C4-alkyl)amino eller (C1-C4-alkanoyl)aminogrupper. Forbindelsene med formel (I) kan inneholde flere asym-metriske sentra, og således kan de eksistere som enantiomere og diastereomere.Oppfinnelsen omfatter både blandinger og de enkelte isomere for seg. Substituentene R<2>, R<3>og C02R<4>kan ha cis- eller transgeometri i forhold til amidtil-knytningen.
De farmasøytisk akseptable salter av forbindelsene med formel (I) som inneholder et surt senter, er de som dannes med baser som danner ikke-toksiske salter. Eksempler på det er alkalimetallsalter så som natrium-, kalium- eller kalsiumsalter eller salter med aminer så som dietylamin. Forbindelser som har et basisk senter kan også danne syreaddisjonsalter med farmasøytisk akseptable syrer. Eksemplene omfatter hydroklorid-, hydrobromid-, sulfat-elier bisulfat-, fosfat- eller hydrogenfosfat-, acetat-, citrat, fumarat-, glukonat-, laktat-, maleat-, suksinat- og tartratsalter.
Uttrykket bioforløper i ovenfornevnte definisjon betyr et farmasøytisk akseptabelt biologisk degraderbart derivat av forbindelsen med formel (I) som ved administrasjon til et dyr eller menneske konverteres i kroppen for å gi en forbindelse med formel (I).
En foretrukket gruppe av forbindelser med formel (I) er de hvor A er (CH2)4, R<1>er H, og B er (CH2)2/ dvs- forbindelse med formel (II) nedenfor hvor R,R<2>,R<3>,R<4>, Y ogR<5>er som tidligere definert for formel (I):
Også foretrukket er de forbindelser med formelene (I) og (II) hvor R og R<4>begge er H (tovalente syrer), samt biolabile mono- og diesterderivater derav hvor en av eller både R og R<4>er en biolabil esterdannende gruppe.
Uttrykket biolabil esterdannende gruppe forstås av en fagmann som en gruppe som fremskaffer en ester som lett kan spaltes i hovedbestanddelen for å frigjøre den tilsvarende divalente syre med formel (I) hvor R og R<4>begge er H. Et antall slike estergrupper er velkjente f.eks. på penicillin-området eller når det gjelder ACE-hemmende antihypertensive midler.
Vedrørende forbindelsene med formelene (I) og (II) er slike biolabile legemiddelegnede estere særlig fordelaktige når det gjelder å tilveiebringe forbindelser med formel (I) som passer for oral administrasjon. Egnetheten hos enhver enkelt esterdannende gruppe kan fastsettes ved konvensjonelle dyreforsøk eller in vitro enzymhydrolyseforsøk. Således, hvis man ønsker optimal effekt, bør esteren bare hydrolyseres etter adsorbsjon, følgelig bør esteren være motstandsdyktig mot hydrolyse før adsorbsjonen av fordøy-elsesenzymer, men bør lett hydrolyseres av f.eks. lever-enzymer. På denne måte frigjøres den aktive divalente syre inn i blodstrømmen etter oral adsorbsjon.
I tillegg til lavere alkylestere (især etyl) og benzylestere omfatter passende biolabile estere alkanoyloksyalkylestere inklusive alkyl-, cykloalkyl- og arylsubstituerte derivater derav, aryloksyalkylestere, aroyloksyalkylestere, aral-kyloksyalkylestere, arylestere, aralkylestere og haloalkyl-estere hvor nevnte alkanoyl- eller alkylgrupper har fra 1 til 8 karbonatomer og er forgrenet eller rette kjeder, og nevnte arylgrupper er fenyl, naftyl eller indanyl valgfritt substituert med en eller flere C1-C4-alkyl eller C^- C^-alkoksygrupper eller haloatomer.
Således omfatter eksempler på R og R<4>når de er biolabile esterdannende grupper andre enn etyl og benzyl: 1-(2,2-dietylbutyryloksy)etyl, 2-etylpropionyloksymetyl, 1-(2-etylpropionyloksy)etyl, 1-(2,4-dimetylbenzoyloksy)etyl, tf-benzoyloksybenzyl, 1-(benzoyloksy)etyl, 2-metyl-l-propionyloksypropyl, 2,4,6-trimetylbenzoyloksymetyl, 1-(2,4,6-triemetylbenzoyloksy)etyl, pivaloyloksymetyl, fenetyl, fenpropyl, 2,2,2-trifluoretyl, 1- eller 2-naftyl, 2,4-dimetylfenyl, 4-t-butylfenyl, 5-(4-metyl-l,3-dioksalynyl-2-onyl)metyl og 5-indanyl.
Særlig foretrukne biolabile esterdannende grupper er etyl, benzyl, 2,4-dimetylfenyl, 4-t-butylfenyl og 5-indanyl.
Forbindelser med formelene (I) og (II) hvor en av eller både R ogR<4>er C^-Cg-alkyl, særlig etyl eller benzyl, er også aktive i kraft av sin hydrolyse in vivo, og er i tillegg verdifulle mellomprodukter for å fremstille de divalente syrer hvor R og R<4>begge er H.
I en ytterligere gruppe av foretrukne forbindelser med formel II er R, R<2>ogR<4>hver H. R<3>er fortrinnsvis H eller C4_Cg-alkyl især n-butyl. Særlig foretrukket er de forbindelser hvor karboksylgrupper C002R<4>er knyttet til 3-eller 4-posisjonen i cykloheksanringen, især de forbindelser som har cis-stereokjemi i forhold til amidgruppen.

Forbindelsene med formel (I) fremstilles ved en rekke forskjellige fremgangsmåter i henhold til oppfinnelsen: (a) I en fremgangsmåte fremstilles forbindelsene med formel (I) hvorR<5>erR<6>CONR<9>ved en fremgangsmåte som innebærer å acylere et amin med formel:

Omsetningen av forbindelsene med formel (III) og (IV) oppnås idet man bruker konvensjonelle amidkoblingsteknikker. Således oppnås i en fremgangsmåte omsetningen med reaktant-ene oppløst i en organisk syre f.eks. diklormetan, idet man bruker et karbodiimidkondenserende middel f.eks. l-etyl-3-(3-dimetylaminopropyl)-karbodiimid eller N,N'-dicykloheksyl-karbodiimid, fortrinnsvis i nærvær av 1-hydroksybenzotriazol og en organisk base så som N-metylmorfolin. Reaksjonen er vanligvis ferdig etter en tidsperiode på 10 til 24 timer ved værelsestemperatur, og produktet isoleres deretter ved konvensjonelle metoder, dvs. ved å vaske med vann eller filtrere for å fjerne biproduktet urinstoff og inndunste løsningsmiddelet. Produktet kan renses ytterligere ved krystallisering eller kroraatografi hvis nødvendig. Forbindelsene med formel (V) omfatter forbindelser med formel (I) hvor R og R<4>er C1-C6-alkyl eller benzyl.
Diesterene med formel (V) kan videre omsettes for å gi monoesteren eller disyrens derivater med formel (I) hvor en av eller både R og R<4>er H. Betingelsene som man bruker, vil avhenge av de nøyaktige egenskaper hos gruppene R<15>og R<16>som er tilstede i forbindelsen med formel (V), og et antall varianter er mulige. Således vil f.eks. når R<15>og R<16>er benzyl, hydrogenering av produktet gi den divalente syre med formel (I) hvor R og R<4>er begge H. Alternativt hvisR<15>er benzyl ogR1<6>er alkyl, vil hydrogenering gi et monoesterprodukt. Dette kan deretter hydrolyseres, hvis ønsket, for igjen å gi disyreproduktet. Når en av R<15>og R<16>er t-butyl, gir behandling av forbindelsen med formel (V) med trifluoreddiksyre den tilsvarende syre. Diester-produktet hvor R<15>og R<16>er benzyl eller lavere alkyl, kan også behandles med trimetylsilyljodid for å gi dikarboksyl-syren. Hvis man bruker en annen karboksylsyrebeskyttende gruppe forR<15>ellerR1<6>, må man bruke helt passende betingelser for å fjerne den i det avsluttende trinn for å få esteren eller disyren med formel (I). I det tilfellet hvor ringen A eller substituenten R^ er umettet, kan man oppnå deblokkering ved ikke-reduktive metoder, således f.eks. hvis enten R eller R<4>er benzyl, kan de fjernes ved behandling med trimetylsilyljodid.
Til slutt fjernes enhver beskyttende gruppe som kan være tilstede i R<6>', ved metoder som passer til den spesielle gruppe som er brukt. Således f.eks. hvis en aminogruppe er tilstede i R<6>, kan denne beskyttes som benzyloksy-karbonylaminogruppe, idet benzyloksykarbonylgruppen fjernes i det avsluttende trinn ved katalytisk hydrogenering. Forbindelser med formel (I) hvor en av eller både R ogR<4>er biolabile esterdannende grupper, fremstilles idet man følger lignende fremgangsmåter, utgående med en forbindelse med formel (III) hvorR1<5>og/ellerlR<16>er biolabile ester-dannende grupper.
I hvert tilfelle kan produktet erholdes som den frie karboksylsyre, eller den kan nøytraliseres med en passende base og isoleres i saltform.
Utgangsmaterialet cykloalkyl-substituerte glutarsyremono-estere med formel (III) kan fremstilles ved forskjellige fremgangsmåter som beskrevet i vår europeiske patentsøknad nr. 0274234.
Syrene med formel (IV) er vanligvis kjente forbindelser som enten er kommersielt tilgjengelige, eller de kan fremstilles ved å følge tidligere litteratur. (b) I en ytterligere fremgangsmåte kan forbindelser med formel (I) hvor R<5>er R<6>S02NR<9->, fremstilles ved en helt analog fremgangsmåte ved å omsette et sulfonylhalid med formel R<6>S02-hal med et amin med formel (III). (c) Forbindelser med formel (I) hvorR<5>erR<7>NR<9>CO-fremstilles ved å omsette en forbindelse med formel:
hvor A, B, Y,R<1>,R2,R3,R15 og R16 er som tidligere definert, med et amin med formel R<7>R<9>NH, etterfulgt av fjerning av enhver beskyttende gruppe som måtte være tilstede, og hydrolyse av esterproduktet for å få karboksylsyrene hvor R og R<4>er H.
Reaksjonen mellom forbindelsen med formel (IX) og aminet kan oppnås ved å bruke amidkoblingsteknikker som er beskrevet under fremgangsmåte (a). De påfølgende trinn er også tidligere beskrevet. Forbindelsene med formel (IX) fremstilles ved å følge fremgangsmåtene som er beskrevet i vår europeiske patentsøknad nr. 0274234, og man får den tilsvarende benzylester (hvor R<5>er C6H5CH2OCO-), katalys-tisk hydrogenering gir karboksylsyren med formel (IX). Aminene med formel R<7>R<9>NH får man vanligvis fra naturlig forekommende aminosyrer med passende beskyttelse av reaktive sidekjeder.
(d) I en ytterligere fremgangsmåte fremstilles forbindelser med formel (I) hvor R<5>er R<6>CONR<9>eller hvorR<5>er R<6>C02-,R<6>C0-,R<6>SOq-,R7NR<9>S02- ellerR<7>0C0, idet man følger den syntetiske fremgangsmåte som er beskrevet i vår europeiske patentsøknad nr. 0274234, dvs. idet man bruker følgende f remaancrsmåte:
hvor A, B, Y,R<1>,R2,R3,R15 ogR<16>er som tidligere definert, og R<5>' er som definert for R<5>med enhver reaktiv gruppe deri valgfritt beskyttet.
De beskyttende teknikker og koblingsteknikker som er nødvendige, er beskrevet tidligere. Forbindelsene med formelene (VI) og (VII) kan fremstilles ved å følge de generelle fremgangsmåter som er beskrevet i ovenfornevnte europeiske patentsøknad.
Som nevnt tidligere er oppfinnelsens forbindelser kraftige hemmere av den nøytrale endopeptidase (E. C. 3 . 4 . 24 .11),. Dette enzym er medvirkende i nedbrytningen av et antall peptidhormoner, og den medvirker især i nedbrytningen av ANF. Dette hormon består av en familie beslektede natriuretiske peptider som utskilles av hjertet, av hvilke den vesentligste sirkulerende form i mennesker er kjent som det 2 8 aminosyrers store peptid som omtales som oc-hANP (se f.eks. G. A. Sagnella og G. A. MacGreggor, Nature, 1984, 309. 666 og S. A. Atlas og andre, Nature, 1984, 309, 717-725. Således kan oppfinnelsens forbindelser, ved å hindre nedbrytning av ANF, forsterke med endopeptidase E.C.3.4.24.11 sin biologiske virkning, og forbindelsene er således diuretica og natriuretiske midler for bruk i et antall forstyrrelser som nevnt tidligere.
Virkning mot nøytral endopeptidase E.C.3.4.24.11 fastsettes ved å bruke en fremgangsmåte som er basert på prøvemetoden som er beskrevet av J. T. Gafford, R. A. Skidgel, E. G. Erdos og L. B. Hersh, Biochemistry. 1983, 32., 3265-3271. Metoden innebærer å bestemme konsentrasjonen av forbindelsen som er nødvendig for å redusere til 50% hastigheten for frigjørelsen av radioaktivt merket hippursyre fra hippuryl-L-fenylalanyl-L-arginin av et nøytralt endopeptidasepreparat fra rottenyrer.
Forbindelsenes aktivitet som diuretica bestemmes ved å måle deres evne til å øke urinutskillelsen og natriumionutskill- eisen i bevisste mus som er injisert med salin. I dette forsøket akklimatiseres hannmus (Charles River CD1, 22-28
g), og de får faste over natten i metabolske bur. Musene doseres intravenøst via haleåren med forsøksforbindelsen som
er oppløst i et volum saltløsning som tilsvarer 2,5% av kroppsvekten. Urinprøver oppsamles hver time i 2 timer i forhåndsveide rør, og konsentrasjonen av elektrolytt bestemmes. Urinvolumet og natriumionkonsentrasjonen fra forsøksdyrene sammenlignet med en kontrollgruppe som bare fikk salin.
Administrasjonen til mennesker ved helbredende eller forebyggende behandling av hypertensjon, hjerteinfarkt eller renal insuffisiens vil orale doser av forbindelsene vanligvis være i området fra 4 til 800 mg daglig for en voksen pasient (70 kg). For en typisk voksen pasient inneholder således enkelttabletter eller kapsler fra 2 til 400 mg aktiv forbindelse i en passende farmasøytisk akseptabel form eller bærerstoff for å gis enkeltvis eller i flere doser en eller flere ganger om dagen. Dosering for intravenøs administrasjon vil vanligvis være innen området 1 til 400 mg pr. enkeltdose etter behov. I praksis vil legen bestemme den nødvendige dose som vil være mest passende for hver enkelt pasient, og den vil variere med alder, vekt og mottagelighet hos den enkelte pasient. Ovenfornevnte doseringer er eksempler på det gjennomsnittlige tilfellet, men det kan naturligvis være enkelte tilfeller hvor høyere eller lavere doseringer er påkrevet, og disse er innenfor denne oppfinnelses omfang.
For bruk til mennesker kan forbindelsene med formel (I) administreres alene, men vil vanligvis bli administrert i blanding med et farmasøytisk bærerstoff valgt med hensyn på hvilken måte man har tenkt å administrere stoffet på og normal farmasøytisk praksis. F.eks. kan de administreres oralt i form av tabletter som inneholder slike hjelpestoffer som stivelse eller laktose, eller i kapsler eller ovuler enten alene eller i blanding med hjelpestoffer, eller i form av eleksirer eller suspensjoner som inneholder aroma- eller fargestoffer. De kan injiseres parenteralt f.eks. intra-venøst, intramuskulært eller subkutant. For parenteral administrasjon brukes de best i form av en steril vanndig oppløsning som kan inneholde andre stoffer, f.eks. nok salt eller glukose for å gjøre oppløsningen isotonisk med blod.
Forbindelsene kan administreres alene, men kan også administreres sammen med slike midler som legen forordner for å optimere kontrollen av blodtrykket eller for å behandle hjerteinfarkt, renal insuffisiens eller andre forstyrrelser i hver enkelt pasient i henhold til etablert medisinsk praksis. Således kan forbindelsene koadministr-eres med et stort utvalg kardiovaskulære midler, f.eks. med en ACE-hemmer så som captopril eller enalapril for å gjøre kontrollen av blodtrykket lettere ved behandling av hypertensjon; eller med digitalis eller et annet hjerte-stimulerende middel eller med en ACE-hemmer ved behandling av hjerteinfarkt. Andre muligheter omfatter koadministrer-ing med en kalsiumantagonist (f.eks. nifedipin, amlodipin eller diltiazem), en betablokkerer (f.eks. atenolol) eller en alfablokkerer (f.eks. prazosin eller doxazosin) ettersom legen bestemmer hva som er passende ved behandling av den spesielle pasient eller aktuelle tilfelle.
I tillegg til ovennevnte kan forbindelsene også administreres sammen med eksogent ANF eller et derivat derav eller beslektet peptid eller peptidfragment som har diuretisk/natriuretisk aktivitet eller med andre ANF-genbeslektede peptider (f.eks. som beskrevet av D. L. Vesely et al., Biochem. Biophys. Res. Comm., 1987, 143, 186).
Således tilveiebringer oppfinnelsen i et videre trekk en farmasøytisk blanding som omfatter en forbindelse med formel (I) eller et farmasøytisk akseptabelt salt derav eller bioforløper for det sammen med et farmasøytisk akseptabelt fortynningsstoff eller bærerstoff.
Oppfinnelsen omfatter også en forbindelse med formel (I) eller et farmasøytisk akseptabelt salt derav eller biofor-løper for det for bruk i medisin især for bruk som et diuretica ved behandling av hypertensjon, hjerteinfarkt eller renal insuffisiens hos mennesker.
Oppfinnelsen omfatter videre bruk av en forbindelse med formel (I) ved fremstilling av et legemiddel for behandling av hypertensjon, hjertesvikt, angina, renal insuffisiens, premenstrual sydrom, cyklisk oedema, Menieres sykdom, hyperaldosteronisme, pulmonær oedema, ascites, hypercalciuremi, glaucoma, astma, betennelse, smerte, epilepsi, følelsesforstyrrelser, dementia og geriatrisk forvirring, fedme, fordøyelsesforstyrrelser (inkl. diarrhoea), hyperreninaemia, leukemia og behandling av magesyresekresjon.
Fremstillingen av oppfinnelsens forbindelser vil nå bli nærmere illustrert ved henvisning til følgende eksperiment-elle eksempler. Forbindelsenes renhet ble rutinemessig kontrollert tynnskiktskromatografi idet man brukte Merck Kieselgel 60 F254~<p>later. 1H-Kjernemagnetiske resonans-spektra ble nedtegnet med et Nicolet QE-3 00 spektrometer, og de var i alle tilfeller i samsvar med de foreslåtte strukturer.
EKSEMPEL 1
2- ( N-^- acetvl- N-^- benzvloksvkarbonyl- L- lysyl- aminometyl) - 3-( cis- 4- etoksykarbonyl- cykloheksvl) karbamoyl) cyklopentyl ^ propansyre- t- butylester
1-hydroksybenztriazol (207 mg, 1,53 mmol) og N-metylmorfolin (235 mg, 2,36 mmol) ble tilsatt til en omrørt oppløsning avN<2->acetyl-N<6->benzyloksykarbonyl-L-lysin (456 mg, 1,41 mmol) i tørt diklormetan ved 0°C, etterfulgt av l-etyl-3-(3-dimetylaminopropyl)karbodiimid (361 mg). Løsningen ble omrørt ved 0°C i 20 minutter, og 3- £l-((cis-4-etoksy-karbonylcykloheksyl)-karbamoyl)cyklopentyl3 -2-(aminometyl)-propansyre-t-butylester (500 mg, 1,18 mmol) i diklormetan (10 ml) ble tilsatt i en omgang, og reaksjonsblandingen fikk oppvarmes til værelsestemperatur og ble omrørt i 16 timer. Løsningen ble konsentrert under våkum til et volum på 10 ml og fordelt mellom etylacetat og vann. Den organiske fase ble vasket med vann (2 x 50 ml), 2M saltsyre (50 ml), 2 x 25 ml), natriumbikarbonatløsning (2 x 25 ml) og saltlake og deretter tørket (magnesiumsulfat), og løsningsmiddelet ble inndampet. Residiet ble kromatografert på silikagel idet man eluerte med etylacetat for å få tittelforbindelsen (610 g, 71%). Funnet: C,63,34; H, 8, 61; -N, 7,45.

Følgende forbindelser ble fremstilt ved den generelle metode fra eksempel 1 idet man brukte som utgangsmateriale enten 3- £l-((cis-4-etoksykarbonyl-cykloheksyl)karbamoyl)cyklopentyl i -2-(aminometyl)propansyre-t-butylester (se preparat 1) eller 3-etoksykarbonylisomeren (se preparat 2) og omsatte med den passende syre med formel IV istedenfor N<2->acetyl-N<6->benzyloksykarbonyl-L-lysin.

Følgende forbindelser ble,fremstilt idet man fulgte den generelle fremgangsmåte eksempel 1 og brukte som utgangsmateriale 3- {l-((cis-3-etoksykarbonyl-cykloheksyl)-karbamoyl)cyklopentyl3 -2-amino-propansyre og omsatte med den passende syre med formel IV.

Følgende forbindelser ble fremstilt idet man fulgte den generelle fremgangsmåte fra eksempel 1 og brukte det passende amin med formel (III) og koblet med N<2->acetyl-N<6->benzyloksykrbonyl-L-lysin.

N T4- £l-( cis- 3- t- butoksykarbonvlcykloheksyl)- karbamoyl)-cyklopentyll - 3-( t- butoksykarbonyl)- butanoylTl fenvlalanin- t-butvlester
1-hydroksybenztriazol (76 mmol) og N-metylmorfolin (0,5 ml, 4,5 mmol) ble tilsatt til en omrørt oppløsning av 3- £l-((cis-3-t-butoksykarbonylcykloheksyl)-karbamoyl)cyklopentyl j-2-(karboksymetyl)propansyre-t-butylester (219,6 mg, 0,457 mmol) i tørt diklormetan (20 ml) ved 0°C etterfulgt av 1-etyl-3-(dimetylaminopropyl)karbodimid (94 mg). Løsningen ble omrørt ved 0°C i 20 minutter, og fenylalanin-t-butylester (128 mg, 0,5 mmol) i tørt diklormetan (5 ml) ble tilsatt i en omgang, og reaksjonen fikk oppvarmes til værelsestemperatur og ble omrørt i 16 timer. Løsningen ble konsentrert under våkum til et volum på 10 ml og fordelt mellom etylacetat og vann. Den organiske fase ble vasket med vann (2 x 25 ml), 2 M saltsyre (2 x 10 ml), natrium-bikarbonatløsning (2 x 10 ml) og saltlake og tørket (MgS04), og løsningsmiddelet ble inndampet hvilket ga tittelforbindelsen som en olje (325 mg, 100%).
Funnet: C,66,59; H,8,92; N,4,25. C39<H>50<N>2<O>8. H20 krever C,66,63; H,8,89; N,3,49%.
EKSEMPLENE 3 5- 38
Følgende forbindelser ble fremstilt idet man fulgte fremgangsmåten fra eksempel 34 og brukte som utgangsmateriale den passende 3- [1-(cis-3-alkoksykarbonyl-cykloheksyl)karbamoyl)cyklopentyl3 -2-karboksy-metyl-propansyre-t-butylester og omsatte med det passende amin med formelR7R<9>NH.
EKSEMPEL 39
2-( N—- acetyl- N—- benzyloksykarbonyl- L- lysyl- aminometyl)- 3-£l-(( cis- 4- etoksykarbonylcykloheksyl) karbamoyl) cyklopentyl 3 propansyre
T-butylesteren fra eksempel 1 (571 mg, 0,783/ mmol) ble oppløst i en blanding av trifluoreddiksyre (2 ml) og diklormetan (1 ml). Løsningen ble holdt på 4°C over natten, deretter konsentrert til tørrhet under våkum, og residiet ble behandlet azeotropt seks ganger med diklormetan. Det resulterende råprodukt ble deretter opptatt i etylacetat og vasket med vann inntil vaskevannet var nøytralt. Den organiske fase ble tørket (magnesiumsulfat) og inndampet under våkum hvilket ga tittelforbindelsen (442 mg, 84%) som et hvitt skum. En løsning av produktet fra eksempel 77 (200 mg, 0,31 mmol) i en blanding av etanol (27 ml) og vann (3 ml) ble redusert på 10% palladium på trekull (20 mg) under et trykk på 50 p.s.i. (3,36 bar) hydrogen i 2 1/2 time. Løsningen ble filtrert, og løsningsmiddelet inndampet under våkum, og residiet ble azeotropisk behandlet med diklormetan (6 x) hvilket ga tittelforbindelsen (161 mg, 100%) som et hvitt fast stoff, smp. 161-163°C. Funnet: C,56,80; H,8,49; N,9,22.<C>25<H>42N407.H20 krever C,56,80; H,8,39; N,10,60%.

2- ( N-^- metansulf onyl- L- lysyl- aminometyl)- 3- £l-( ( cis- 4-karboksy- cis- 3- butylcykloheksyl) karbamoyl) cyklopentyl A propansyre
1. 3-( 1- karboksycyklopentyl)- 2- aminopropansyre t- butvlester
3- (1-karboksycyklopentyl)-2-(dibenzylaminometyl)propansyre-t-butylesterhydroklorid (2,0 g; 41 mmol) i etanol (160 ml) og trietylamin (20 ml) ble hydrogenert over palladium (fra 20% Pd(OH)2/C; 20 g) ved 60 p.s.i. (4,1 bar). Etter atten timer ble blandingen filtrert gjennom arbicel, løsnings-middelet ble inndampet, og residiet tørket azeotropisk med toluen. Det ønskede primære aminosyretrietylaminsalt, som inneholdt en molekvivalent trietylaminhydroklorid, ble således erholdt som et hvitt fast stoff (16,21 g).
2. 2-( N^- benzvloksykarbonvl- N^- t- butoksykarbonyl- L- lysyl-aminometyl)- 3-( 1-( 1- karboksycvklopentyl) propansyre- t-butvlester ( diastereoisomer)
Ovenfornevnte produkt 8,2 4 g; 2 0,1 mmol) og N<2->benzyloksykarbonyl-N<6->t-butoksykarbonyl-L-lysin 4-nitrofenylester (9,50 g, 18,9 mmol) ble oppløst i tørt metylenklorid (70 ml). Løsningen ble omrørt, etter avkjøling til 10°C ble det tilsatt trietylamin (2,64 ml, 18,9 mmol). Etter en halv time ble blandingen homogen og fikk stå ved værelsestemperatur over natten. Løsningen ble deretter vasket med IM sitronsyre etterfulgt av vann, tørket over magnesiumsulfat og inndampet. Residiet ble renset ved kromatografi på kiselgel (500 g) idet mån eluerte med økende mengder etylacetat i heksan (2:1 til 4:1) og til slutt med etylacetat, heksan, eddiksyre (4:1:0,05). Den ønskede blanding av diastereoisomerene ble således erholdt som en fargeløs gummi (10,5 g). Isomerene ble deretter adskilt ved kromatografi på kiselgel (1 kg) idet man eluerte med en blanding av toluen, isopropanol og dietylamin (10:2:1). Dietylaminsaltet av den ønskede, ikke-polare diastereoisomer ble erholdt som et orangefarget skum (3,01 g), som ble oppløst i etylacetat og vasket med IM sitronsyre og saltlake. Tørket over magnesiumsulfat og inndamping ga den frie syre som et gult skum (2,81 g).
Funnet: C,62,89; H,8,29; N,6,69. C33<H>51<N>3°9 krever C,62,54; H,8,ll; N,6,63%. Ytterligere kromatografi av en liten prøve på kiselgel med økende mengder etylacetat i heksan (2:3 til 17:3) ga et kremfarget pulver (a)<2>^- 2,8°, (C0 365"3'6° (c = °'5'etanol).
3. 2- ( N-^- benzyloksykarbonyl- N-^- t- butoksykarbonyl- L- lysyl-aminometyl)- 3- £l-(( cis- 4- etoksykarbonyl- cis- 3- butyl-cykloheksyl)- karbamoyl) cyklopentyl^ propansyre- t- butylester
Kobling av ovenfornevnte produkt fra trinn 2 (650 mg; 1,03 mmol) med c-4-amino-c-2-butyl-r-l-cykloheksan karboksylsyre-etylesterhydroklorid (271 mg; 1,03 mmol) som beskrevet i eksempel 1 etterfulgt av kromatografi på kiselgel idet man eluerte med økende mengder etylacetat i heksan (7:3 til 4:6) ga det ønskede produkt som et lyst skum (63 0 mg; 73%). Funnet: C,64,37; H,8,98; N,6,81. C46H74<N>4O10(0,75 H20) krever C,64,50; H,8,88; N,6,54%.
4. 2-( N^- metansulf onyl- L- lysyl- aminometyl) - 3- fl-(( cis- 4-karboksy- cis- 3- butvlcykloheksyl) karbamoyl) cyklopentyl^ propansyre
i) Ovenfornevnte produkt fra trinn 3 (62 0 mg; 0,735 mmol) i etanol (18 ml) og vann (2 ml) ble hydrogenert over 5% palladium på kull (200 mg) ved 50 p.s.i. (3,5 bar). Etter 3 timer ble blandingen filtrert gjennom arbicel og inndampet til tørrhet hvilket ga et hvitt skum (520 mg; 95%).
ii) Metansulfonylklorid (0,11 ml; 1,41 mmol) ble tilsatt dråpevis til en iskald omrørt løsning av ovenfornevnte produkt (500 mg; 0,67 mmol) og N-metylmorfolin (0,16 ml; 1,4 mmol) i tørt metylenklorid (15 ml). Etter tre timer ble det
tilsatt mer metansulfonylklorid (0,03 ml) og N-metylmorfolin (0,04 ml) og blandingen ble holdt på 0°C over natten. Blandingen ble deretter vasket etter hverandre med vann, mettet vanndig natriumkarbonat og vann, tørket over magnesiumsulfat og inndampet for å gi råproduktet som ble kromatografert på kiselgel. Eluering med økende mengder etylacetat i heksan (4:6 til 1:9) ga det ønskede metansulf onylderivat som et fargeløst skum (460 mg; 87%) . Rf. 0,15 (etylacetat, heksan, 1:1).
iii) Behandling av ovennevnte produkt (440 mg; 0,56 mmol) med trifluoreddiksyre som beskrevet i eksempel 39, etterfulgt av hydrolyse med IN natriumhydroksyd (4,5 ml) ved 50-55°C i 65 timer og adsorbsjon på ionebytterharpiks 50W-X8 idet man eluerte med 10% vanndig pyridin ga den ønskede divalente syre som et skum. Triturering med acetonitril ga et hvitt pulver (245 mg; 73%).
Funnet: C,54,92; H,8,29; N,9,12. C28<H>50<N>4O8S (0,5 H20) krever C,54,97; H,8,40; N,9,16%.

Denne forbindelse ble fremstilt idet man fulgte fremgangsmåten fra eksempel 135 men brukte c-4-amino-c-2-(3-metylbutyl)-r-l-cykloheksan karboksylsyre i trinn 3. Produktet ble erholdt som et kremfarget pulver.
Funnet: C,56,07; H,8,22; N,8,95. C29H52N4<0>8S krever C,56,47; H,8,50; N,9,08%.
Fremstilling 1
3- fl- f( cis- 4- etoksykarbonvl- cykloheksyl) karbamoyl) cyklopentyl 3- 2-( aminometyl) propansyre- t- butylester (a) En løsning av 2-(bromometyl)propansyre-t-butylester (20 g, 90,5 mmol) i tørt acetonitril (360 ml), avkjølt til 0°C, ble behandlet med fast kaliumkarbonat (15,63 g, 113 mmol), etterfulgt av en løsning av dibenzylamin (17,83 g, 90,5 mmol, i tørt acetonitril (600 ml) hvilket fikk temperaturen til å stige til 10°C. Reaksjonsblandingen ble omrørt ved 0°C i en halv time, etterfulgt av en time ved værelsestemperatur, og deretter ble den fordelt mellom vann og dietyleter. Eterfasen ble vasket igjen med vann, tørket (natriumsulfat) og inndampet for å gi råproduktet (31 g) som ble filtrert gjennom et lag kiselgel, idet man eluerte med (heksan/CH2Cl2(1:1), for å gi 2-(dibenzylaminometyl)propen-syre-t-butylester (23,8 g, 78%) som et fast stoff, smp. 62-63°C. Funnet: C,78,09; H,8,20; N,4,18. C22<H>27N02krever C,78,3; N,8,06; N,4,15%. (b) Til en omrørt løsning av diisopropylamin (14,98 g, 20,75 ml, 148 mmol i tørt tetrahydrofuran (250 ml), avkjølt til -3 0°C under nitrogen, ble det tilsatt dråpevis n-butyl litium (59,3 ml 2,5 M løsning, 148 mmol), idet man holdt temperaturen under -2 0°C. Reaksjonsblandingen ble omrørt ved -2 0°C i 1 time, deretter avkjølt til -3 0°C, og cyklo-pentankarboksylsyre (8,05 g, 7,65 ml, 70,6 mmol) ble tilsatt dråpevis i en liten mengde tørt tetrahydrofuran. Reaksjonsblandingen ble omrørt ved 0°C i 2 timer, og i løpet av denne tiden dannet det seg en hvit felling. Løsningen ble
deretter avkjølt til -70°C, og en løsning av 2-(di-benzylaminometyl) -propensyre-t-buty lester (23,8 g, 70,6 mmol) i tørt tetrahydrofuran (35 ml) ble tilsatt dråpevis. Blandingen fikk stå over natten (under -40°C), og deretter ble den helt inn i iskald saltsyre (4,2 ekv., slutt pH = 2), og produktet ble ekstrahert inn i dietyleter. Eterskiktet ble vasket med saltsyre, vann (2 x), tørket (natriumsulfat)
og inndampet for å gi råprodukte (40 g). Dette ble kromatografert over kiselgel (500 g) idet man eluerte med diklormetan som inneholdt metanol (0 til 5%) hvilket ga etter inndamping av den rene fraksjon, 3-(1-karboksy-cyklopentyl) -2-(dibenzylaminometyl)propansyre-t-butylester som en olje (18,3 g, 57%). (c) T-butylesteren fra trinn (b) ovenfor (11,97 g, 26,54 mmol) i tørt diklormetan (200 ml), under nitrogen, ble behandlet med l-etyl-3-(3-dimetylaminopropyl)karbodiimid-hydroklorid (10,19 g, 53 mmol), 1-hydroksybenztriazol (3,94 g, 29 mmol), trietylamin (16,08 g, 22,15 ml, 159 mmol) og cis-4-aminocykloheksan-karboksylsyreetylesterhydroklorid (6,04 g, 20 mmol). Etter omrøring over natten ved værelsestemperatur ble løsningsmiddelet dampet vekk, og residiet fordelt mellom etylacetat og vann. Den organiske fase ble vasket med fortynnet saltsyre (2 x), fortynnet vanndig natriumbikarbonat, vann, tørket (natriumsulfat) og inndampet for å gi råproduktekt (15,5 g). Dette ble kromatografert på kiselgel (400 g) idet man eluerte med 1-10% dietyleter i diklormetan. Inndamping av den ønskede fraksjon ga 3- £l-((cis-4-etoksykarbonyl-cykloheksyl)-karbamoyl)cyklopentyl^ -2-(dibenzylaminometyl)propansyre-t-butylester som en olje (11,43 g, 71%). Funnet: C,73,38; H,8,87; N,4,42. C37<H>52<N>2°5krever: C,73,48; H,8,67; N,4,68%.
(d) Produktet fra trinn (c) ovenfor (4,6 g, 7,62 mmol) i etanol (75 ml) ble hydrogenert i en atmosfære av hydrogen (50 p.s.i., 3,46 bar) over palladium på trekull (200 mg) ved værelsestemperatur over natten. Reaksjonsblandingen ble filtrert gjennom et Solkaflok lag, og filtratet inndampet til tørrhet for å gi råproduktet som en olje. Dette ble oppløst i dietyleter og ekstrahert inn i fortynnet saltsyre (pH 2) (2 x).
