(57) Sammendrag
Det beskrives anvendelse av endoparasitiske depsipeptider for fremstilling av medikamenter for prevensjon av vertikale endoparasittinfeksjoner.
Den foreliggende oppfinnelse angår anvendelse av endoparasittisidet depsipeptider for fremstilling av legemidler for inhibering av den vertikal infeksjon med endoparasitter.
Det sykliske depsipeptid PFI 022 og dens virkning mot endoparasitter er kjent fra EP-A 382 173. Ytterligere sykliske depsipeptider og deres endoparasitiske virkning er gjenstand for søknadene EP-A 626 376; EP-A 626 375 og EP-A 644 883.
Endoparasitiske midler som inneholder praziquantel eller epsiprantel og sykliske depsipeptider er beskrevet i EP 662 326.
EP 1142577 A2 omtaler middel uten depsipeptid, men med selamectin.
Det er kjent at disse midler etter oral, parenteral eller transdermal applikering virker mot endoparasitter hos det angjeldende dyr. Til nå var det imidlertid ikke kjent at disse aktive bestanddeler også kan forhindre vertikale infeksjoner hos etterkommerne av dyrene.
Foreliggende oppfinnelse angår derfor:
Anvendelsen av endoparasitiske depsipeptider for fremstilling av legemidler for å forhindre vertikal infeksjon av endoparasitter.
Depsipeptider ligner peptidene og skiller seg fra disse ved at en eller flere aaminosyrebyggeklosser er erstattet med α-hydroksykarboksylsyrebyggeklosser. I henhold til oppfinnelsen anvendes det fortrinnsvis sykliske depsipeptider med 18 til 24 ringatomer, og særlig med 24 ringatomer.
Til depsipeptidene med 18 ringatomer hører forbindelsen med den generelle formel (I);
der

Aktiv bestanddeler kan etter struktur foreligge i stereoisomere former eller som stereoisomere blandinger, for eksempel som enantiomerer eller racemater. Både stereoisomere blandingene og de rene stereoisomerer kan anvendes ifølge oppfinnelsen.
Videre kan eventuelt anvendes: salter av aktiv bestanddelen med farmasøytisk godtagbare syrer eller baser og også solvater og særlig hydrater av de aktive bestanddeler eller deres salter.
Ifølge oppfinnelsen kan det bekjempes patogene endoparasitter som forekommer oss mennesker og i dyrehold og dyreoppdrett ved nytte-, foredlings-, dyrehave-, arbeids-, forsøk- eller hobbydyr. De er derved virksomme mot alle eller enkelte utviklingsstadier av skadedyrene samt mot de resistende og normalt sensible arter. Ved bekjempelse av de patogene endoparasitter kan sykdommer, dødsfall og y tel se sreduksj on er (f. eks. ved produksjon av kjøtt, melk, ull, hud, egg, osv.) reduseres slik at det ved anvendelse av aktiv bestanddelen er mulig med et mer økonomisk og enkelt dyrehold. Til laboratorie og - forsøksdyr hører mus, rotter, marsvin, gullhamstre, hunder og katter.
Til hobbydyrene hører hunder og katter.
Anvendelsen kan skje både profilaktisk og også terapeutisk. Helt foretrukket er anvendelsen på hunder.
Ved oppfinnelsen anvendelse viser depsipeptidene seg som godt fordragelig og dette gjelder både moderdyr og avkommet.
Vertikal infeksjon betyr at det skjer en overføring fra moderdyr til avkommet og særlig prenatalt, det vil altså si i drektighetsperioden, og/eller galaktogent, også via melken. Av særlig betydning er forhindring av den prenatal e infeksjon.
‘'Forhindring av prenatal og galaktogeninfeksj on” står ikke bare for en fullstendig forhindring av infeksjon men også en ufullstendig forhindring der infeksjonsrisikoen kun forringes og/eller infeksjonen svekkes. Det tilstrebes en minst 90% virksomhet.
Anvendelsen av aktive bestanddeler skjer direkte eller i form av egnete tilberedninger, fortrinnsvis enteralt, parenteralt eller derm alt (transdermalt) hos moderdyret.
Den enteral e anvendelse av denne aktive bestanddel skjer for eksempel oralt i form av pulver, stikkpillere, tabletter, kapsler, næring, drikke, granulater, dren, bolik, medikert for eller drikkevann. Den dermale anvendelse skjer for eksempel i form av dypping, spraying, bading, vasking, påhelling (pouring-on) og innpudring. Den parenterale anvendelse skjer for eksempel i form av injeksjon (intramuskulært, subkutant, intravenøst eller intraperitonealt) eller ved implantater.
Egnede tilberedning er:
Oppløsninger som inj eksj on sl øsninger, orale oppløsninger, konsentrater for oral administrering etter fortynning, oppløsning for bruk på huden eller i kroppshulrom, påhellingsformuleringer, geler;
Emulsjoner og suspensjoner for oral eller dermal anvendelse samt injeksjon; halvfaste tilberedning;
Formuleringer der den aktive bestanddel er forarbeidet i et salvegrunnlag eller i en oljei-vann eller vann-i-olje emulsjoner;
Faste tilberedninger som pulver, forblandinger eller konsentrater, granulater, pellets, tabletter, boli, kapsler; aerosol er og inhalater, aktive bestanddel sholdi ge legemer.
Inj eksj onsoppløsninger kan administreres intravenøst, intramuskulært eller subkutant.
Inj eksj onsoppløsninger fremstilles ved at den aktive bestanddel oppløses i et egnet oppløsningsmiddel og eventuelt tilsettes eventuelle tilsetninger som oppløsningsformidlere, syrer, baser, buffersalter, antioksidanter, konserveringsmidler. Oppløsningene steriliseres filtrert og fylles.
Som oppløsningsmiddel skal nevnes: fysiologisk godtakbare oppløsningsmidler som vann, alkoholer som etanol, butanol, benzylalkohol, glyserin, hydrokarboner, propylenglykol, polyetylenglykoler, N-metylpyrrolidon samt blandinger av disse.
Akliv bestanddelene kan eventuelt også oppløses i fysiologiske godtakbare vegetabilske eller syntetiske oljer som er egnet for injeksjon.
Som oppløsningsformidler skal nevnes: oppløsningsmidler som fremmer oppløsning av aktiv bestanddelene i hovedoppløsningsmidler eller forhindrer utfelling. Eksempler er polyvinylpyrrolidon, polyetoksylerte ricinusolje eller polyetoksylert sorbitanester.
Konserveringsmidler er: benzylalkohol, triklorbutanol, p-hydroksybenzosyreester og nbutanol.
Orale oppløsninger anvendes direkte. Konsentrater anvendes etter forutgående fortynnelser til den ønskede anvendelseskonsentrasj on på oral måte. Orale oppløsninger og konsentrater fremstilles som beskrevet ovenfor ved injeksjonsoppløsninger hvorved man kan gi avkall på og arbeide sterilt.
Oppløsninger for anvendelse på huden kan dryppes på, strykes på, gnies inn, sprøytes på eller påføres ved dypping (dypping, bading eller vasking). Disse oppløsninger fremstilles som beskrevet ovenfor ved inj eksj onsoppløsningene.
Det kan derfor være fordelaktig å tilføye forty kningsmidler ved fremstillingen.
Fortykningsmidler er for eksempel uorganiske fortykningsmidler som bentonitter, kolloide kiselsyre, aluminiummonostearat, organiske fortykningsmidler som cellulosederivater, polyvinylalkoholer og deres kopolymerer, akryl ater og metakrylater.
Gel bringes på huden eller strykes på eller bringes inn i kroppshulrom. Gel fremstilles ved oppløsninger som i sin tur fremstilles som beskrevet ovenfor for
inj eksj onsoppløsninger, tilsettes så mye forty kningsmiddel at det oppstår en klar masse med salvelignende konsistens.
Påhellingsformulering helles på begrensede områder av huden eller sprøytes på slik hvorved aktiv bestanddelen enten trenger gjennom huden og virker systemisk, eller fordeler seg på kroppsoverflaten.
Påhellingsformuleringer fremstilles ved at en den aktive bestanddel oppløses i egnede, hudforenlige oppløsningsmidler eller oppløsningsmiddelblandinger, eller suspenderes eller emulgeres i slike. Eventuelt settes det til ytterligere hjelpestoffer som fargestoffer, resorbsj onsfremmende stoffer, antioksidanter, lysbeskyttelsesmidler eller vedheftingsmidler.
Som oppløsni ngsmi dl er skal nevnes vann, alkanoer, glykol er, polyetyl englykol er, polypropylenglykoler, glyserin, aromatiske alkoholer som benzylalkohol, fenyletanol, fenoksyetanol, estere som eddiksyre, butyl acetat, benzylbenzoat, etere som alkylenglykolalkyletere, som dipropylenglykolmonometyleter og
di etyl engl ykolmonobuty 1 eter, ketoner, som aceton og metyletylketon, aromatiske og/eller alifatiske hydrokarboner, vegetabilske eller syntetiske oljer, DMF, dimetylacetamid, N-m etylpyrroli don og 2-dimetyl -4-oksymetylen- 1 ,3 -di oksolan.
Fargestoffer er alle fargestoffer som er til att for anvendelse på dyr og som kan være oppløst eller suspendert.
Desorpsjonsfremmende stoffer er for eksempel DMSO, utspredende oljer som i sopropylmyri stat, di propyl engl y kolpel argonat, silikonoljer, fettsyreestere, triglyserider og fettalkoholer.
Antioksidanter er sulfitter eller metabisulfitter som kaliummetabi sulfat, askorbinsyre, butylhydroksytoluen, butyl hy droksy ani sol og tocoferol.
Lysbeskyttelsesmidler er for eksempel stoffer fra klassen benzofenoner eller novantisolsyre.
Heftemidler er for eksempel cellulosederivater, sti vel sesderi vater, polyakrylater og naturlige polymerer som alginater eller gelatin.
Emulsjoner kan anvendes oralt, dermalt eller som injeksjon.
Emulsjoner er enten av typen vann-i-olje eller av typen olje-i-vann.
Disse fremstilles ved at den aktive bestanddel enten oppløses i den hydrofobe eller i den hydrofile fase og denne så homogeniseres ved hjelp av egnede emul gator er og eventuelt ytterligere hjelpestoffer som fargestoffer, resorpsj onsfremmende stoffer, konserveringsmidler, antioksidanter, lysbeskyttelsesmidler eller viskositetsøkende stoffer, med oppløsningsmidlet i den andre fase.

Fettsyreestere som etylstearat, di-n-butyryladipat, heksyllaurat og dipropylenglykol pelargonat, estere av en forgrenet fettsyre med midlere kjedelengde med mettede fettalkoholer med kjedelengde C16-C18, i sopropylmyristat, isopropylpalmitat, kapryl/kaprinsyrestere med mettede fettalkoholer med kjedelengde C12-C18, isopropylstearat, oleyloleat, decyloleat, etyloleat, melkesyreetyl, vokslignende fettsyreestere, som kunstig andestj ertkj ertelfett, dibutylftalat, diisopropyladipat, eller esterblandinger av slike også videre.
Fettalkoholer som isotridecylalkohol, 2-oktyldodecanol, cetyl stearylalkohol og oleyl alkohol.
Fettsyrer som for eksempel oljesyrer eller deres blandinger.
Som hydrofil fase kan nevnes:
Vann, alkoholer som propylenglykol, glyserin, sorbitol og blandinger derav.
Som em ul gator er kan nevnes: ikke-jonigenerte ten si der som pol yoksyl erte ritsinusoljer, polyoksylert sorbitanmonooleat, sorbitanmonostearat, glyserinmonostearat, polyoksyetylstearat og alkylfenolpolyglykoletere;
Amfolytiske tensider som di-Na-N-lauryl-p-iminodipropionat eller lecitin;
Anionaktive tensider som Na-laurylsulfat, fettalkoholetersulfater, mono/dialkylpolyglykoleterortofosforsyreestermonoetanolaminsalt;
Kationaktive tensider som cetyltrimetylammoniumklorid.
Som ytterligere hjelpestoffer kan nevnes: viskositetsøkende og emulsjonsstabiliserende stoffer som karboksym etyl cellulose, metylcellulose, og andre cellulose- og
stivel se sderi vater, polyakrylater, alginater, gelatiner, gummi arabikum, polyvinylpyrrolidon, polyvinylalkohol, kopolymerer av metylvinyleter og maleinsyreanhydrid, polyetylenglykoler, vokser og kolloid kiselsyre eller blandinger av de angitte stoffer.
Suspensjonene kan anvendes oralt, derm alt eller som injeksjon. De fremstilles i det den aktive bestanddel suspenderes i en bærervæske, eventuelt under tilsetning av ytterligere hjelpestoffer som fomettere, fargestoffer, resorpsj onsfremmede stoffer, konserveringsmidler, antioksidanter eller lysbeskyttelsesmidler.
Som bærervæsker kan nevnes alle homogene oppløsningsmidler eller oppløsningsmiddelblandinger.
Som fornettere (dispergeringsmidler) kan nevnes de med ovenfor angitte tensider.
Som ytterligere hjelpestoffer nevnes de ovenfor angitte.
Halvfaste preparater kan avgis oralt eller derm alt. De skiller seg fra de ovenfor angitte suspensjoner og emulsjoner kun ved den høyere viskositet.
For fremstilling av faste preparater blir den aktive bestanddel blandet med egnete bærere og eventuelt under tilsetning av hjelpestoffer og brakt til ønsket form.
Som bærere kan nevnes alle fysiologisk godtakbare, faste inerte stoffer. Som slike tjener uorganiske og organiske stoffer. Uorganiske stoffer er for eksempel koksalt, karbonater som kalsiumkarbonat, hydrogenkarbonater, aluminiumoksider, kiselsyrer, leire, felt eller kolloidsilitsiumdioksyd eller fosfater.
Organi ske stoffer er for eksempel sukker-, cellulose-, nærings- eller formidler som melkepulver, dyremel, kornmel eller produkter derav eller stivelse.
Hjelpestoffer er konserveringsmidler, antioksidanter, fargestoffer som allerede angitt ovenfor.
Ytterligere egnet hjelpestoffer er smøre- og glidemidler som for eksempel magnesiumstearat, stearinsyre, talkum, bentonitt, nedbrytningsfremmende stoffer som stivelse eller fornettet polyvinylpyrrolidon, bindemidler som stivelse, gelatin eller lineær polyvinylpyrrolidon, samt tørkebindemidler som mikrokrystallinsk cellulose.
Foretrukne applikasj onsformer er oral applikering, for eksempel ved egnete tabletter, eller transdermal applikering, for eksempel ved en egnet spot-on formulering,
Oppfinnelsens midler kan i tillegg inneholde synergi ster eller andre aktive bestanddeler, for eksempel, slike som virker mot patogene endoparasitter. Slike aktive bestanddeler er for eksempel L-2,3,5,6— tetrahydro-6-fenylimidazotiazol, benzimidazolkarbamater som febantel, videre pyrantel, epsiprantel eller makrosykliske laktoner som for eksempel avermectin, ivermectin eller selamectin. Særlig foretrukket er praziquantel som kombinasj onspartner, særlig sammen med emodepsid.
Praziquantel har lenge vært kjent som aktiv bestanddel mot endoparasitter (se for eksempel US 4001 411). Den fordelaktige kombinasjonen av depsipeptider med praziquantel eller epsiprantel er beskrevet i EP-A-662 326, og det henvises spesielt til dette dokument.
Kombinasjonen av depsipeptider med piperaziner er beskrevet i EP-A-1 189615, og det henvises spesielt til dette dokument.
Transdermalt applikerbare midler inneholdende sykliske depsipeptider for bekjempelse av endoparasitter er beskrevet i søkers egen tyske søknad med aktnummer 10358525.7.
Anvendel sesferdige tilberedninger inneholder de aktive bestanddelene, generelt i konsentrasjoner fra 0,01-25 vekt-% og særlig 0,1-20 vekt-%,
De sykliske depsipeptider anvendes vanligvis i mengde fra 0,1 til 8 vekt-% og særlig 1 til 6 vekt-%.
Praziquantel anvendes vanligvis i mengder på ltil 25 vekt-%, og særlig fra 5 til 15 vekt-%, spesielt 6 til 14 vekt-%.
Middelet fremstilles ved blanding av komponentene i angitte mengder i egnede apparater. Fortrinnsvis går man dermed frem slik at de flytende komponenter blandes hvoretter de faste komponenter tilsettes og det fremstilles en homogen oppløsning.
Generelt har det vist seg fordelaktig å administrere mengder av oppfinnelsens blandinger på rundt 0,1 til 20 mg aktiv bestanddel pr. kg. kroppsvekt pr. dag for å oppnå virksomme resultater. Foretrukket er 0,5 til 10 mg pr. kg. aktiv bestanddel pr. kg. kroppsvekt.
De følgende eksempler skal forklare oppfinnelsen.
Biologisk eksempel
Undersøkelsene skjedde på atten beagle hunn hunder som var infisert med Toxocara canis. Av disse tjente seks dyr som ubehandlet kontrollgruppe. To forskjellige behandlingsskjemaer ble anvendt. I behandlingsgruppe (TI), ble seks hunder fra den 42. drektighetsdag til og med fødselen, og behandlingsgruppe 2 (2T) ble seks hunder fra 42. drektighetsdag hver tredje dag til fødsel av valpene, behandlet med i hvert tilfelle 1 mg emodepsid/kg av kroppsvekt oralt eller som tablettformulering. Bedømmelsen av virkningen av behandlingen skjedde ved å fastslå tre målstørrelser hos valpene: antall somatiske larver av T. canis i fordøyelsesprøver fra lever, lunger, nyre, mave-tarmkanal inkludert innhold og muskulatur (euthanasia av valpene direkte etter fødselen), antall uskilte egg pr. gram avføring og antall intestinale stadier av T canis etter tarmutvasking (euthanasia av valper den 35. livsdag).
Den prosentuale virkning av emodepsid i TI utgjorde for målstørrelsen antall utskilte pr. gram avføring 99,9%, for målstørrelsen antall somatiske larver i
fordøyel sesorganene 96,4% og for målstørrelsen antall intestinale stadier i tarmen 99,9% . I T2 ble det for målstørrelsen antall utskilte pr. gram avføring likeledes oppnådd en virkning på 99,9%. For målstørrelsen antall somatiske larver ble det med behandlingsskjema 2 oppnådd en prostentual virkning av emodepsid på 93,8% og for målstørrelsen antall intestinale stadier i tarmen på 98,7%. I begge behandlingsskj emaer lå den prosentuale virkning av emodepsid for de enkelte målstørrelser samt også middelverdien for disse målstørrelsene i hver behandlingsgruppe (Tl= 98,7%;
T2= 97,5%) tydelig over de av anførte International Cooperation on Harmonisation of Technical Requirements for Registration of Veterinary Medicinal Products (VICH), guideline 7, krevede, prosentuale minste virkninger på 90% for antelmintisk virksomme stoffer. Med begge behandlingsskjemaer ble det oppnådd en tydelig reduksjon av den prenatal e I canis infeksjon hos valpene.
Verken ved mor eller valper kunne det observeres tegn på dårlig fordragelighet.
