Abstract
Oppfinnelsen angår nye immunogene midler som kan indusere spesifikk B-celleimmunitet rettet mot en enkelt epitop tilstede i et selv-antigen. Det immunogene middelet er et kimært bindingsprotein som bindes spesifikt til en første reseptor, hvor nevnte første reseptor er en som binder en andre reseptor som er tilstede i et antigen til et pattedyr, hvor nevnte kimære bindingsprotein omfatter: (a) En B-celleepitop i form av et bindingssete som spesifikt binder den første reseptoren og som konkurrerer med den andre reseptoren for binding til den første reseptoren, (b) en plattformproteinstruktur som er autolog i nevnte pattedyr og (c) i det minste én toleransebrytende aminosyresekvens som er heterolog i nevnte pattedyr og som binder til et MHC klasse IImolekyl i nevnte pattedyr. I foretrukne utførelsesformer er det kimære bindingsproteinet i form av et antiidiotypisk antistoff med en introdusert toleransebrytende aminosyresekvens. Oppfinnelsen angår videre fremgangsmåter for å fremstille immunogenene og fremgangsmåter for å anvende immunogenene i terapi. Oppfinnelsen angår også proteiner, nukleinsyrefragmenter, rekombinant modifiserte vertsceller og virus som er anvendelige i utøvelsen av 25 fremgangsmåtene ifølge oppfinnelsen.
