Abstract

In recovering proteins, microorganism and/or enzymes are first treated at elevated temps. in an aq. medium to form a hydrolysate. The prod. is then concentrated by ultrafiltration, opt. after removal of solid material. The process gives excellent recovery of protein in virtually quantitative yield from hydrolysates contg. proteins, denatured proteins, protein hydrolysis prods. such as oligopeptides and mixts. of these with carbohydrates, fatty acids, etc. The process used less energy than evapn. processes. The process can be used for treating hydrolysates obtd. by hydrolysing active sludges from waste water treatment plants in the presence of NH3.

The invention relates to a method of recovering proteins by treating microorganisms and / or enzymes at an elevated temperature in an aqueous medium to form a hydrolyzate.

Such a method is known, inter alia, from Netherlands Patent Application 7707081, not pre-published. In this patent application it is described that it is possible to hydrolyze microorganisms in the form of active sludge from a purification installation at elevated temperature in the presence of NH 2.

The hydrolyzate formed during such a treatment contains, besides proteins, carbohydrates and hydrolysis products thereof, large amounts of water. For an economic application of especially the proteins from the hydrolyzate it is therefore necessary to completely or largely remove this water.

In this connection, the term protein is understood to include not only protein itself, but also denatured protein, hydrolysis products of protein, such as oligopeptides, and mixtures of the above products with carbohydrates, fatty acids and the like.

A possible solution to the above problem would be to evaporate the water completely. However, the amount of energy required for this is so great that such a method is not attractive.

Surprisingly, it has now been found that it is possible to recover protein, as defined above, with high purity and substantially quantitatively from the hydrolyzate.

According to the invention, a protein recovery method is characterized in that the hydrolyzate is concentrated by ultrafiltration after separation of any solid present.

In the process according to the invention it is preferred to hydrolyze from 2 waste water purification plant in the presence of NH 2.

Depending on the desired application, the concentrate obtained after the ultrafiltration can be further processed in many ways.

If a completely dry product is desired, it is possible to add the concentrate 5 to a drying installation, e.g. to supply a spray dryer. Another possibility is to coagulate the protein in the concentrate, and subsequently separate and dry it.

It is particularly surprising that it is possible to separate the hydrolysed protein almost quantitatively in this manner, since 10 ultracentrifuge measurements Rebben indicated that the molecular weight (Mw) of the hydrolysed protein is approximately 5,000, while it is well known that ultrafiltration in is in fact hardly suitable for concentrating such low-molecular products.
