Abstract

Pyrazine-2-carboxylic acid derivs. (I), their salts and pharmaceutical compns. contng. them. X = alkoxy, piperidino, morpholino, oxazolidino Y = H, 1-5C alkoxy As hypoglycaemic agents. Also reduce blood free fatty acid and triglyceride levels. (a) A suspn. of pyrazinecarboxylic acid (16 g) in 22% HCl-MeOH (100 ml) is refluxed for 1 hr. to give the Me ester (9 g), m.p. 58.5-60.5 deg. (ether). (b) This (13.8 g) is refluxed for 30 mins. with anhyd. morpholine (17.4 ml) and the product purified on silica gel in CHCl3 to give 2-morpholinocarbonyl-pyrazine (17.4 g.), b.p.125-9 deg./0.02 mm.; 4-methiodide m.p.196-7 deg. dec.
