Abstract

Por primera vez, la presente invencion proporciona pANCA aislado, esencialmente puro, y/o producido recombinantemente asociado con colitis ulcerativa (UCpANCA) y material de UCpANCA) así como polinucleotidos que codifican el UCpANCA, material de UCpANCA y polipéptidos de UCpANCA de la presente invencion. Usando la técnica de presentacion del bateriofago, los repertorios de gen de inmunoglobulina derivados para los fines de inmunoglobulina utilizados por los pacientes de UC seropositivo de pANCA se han aislado, amplificado, recombinado al azar, para regenera las bibliotecas del vector de expresion fagemid que codifica los homologos ADN del repertorio del gen. Se proporcionan métodos para enriquecer estas bibliotecas para crear bibliotecas de homologos de ADN que codifican VH- y VL- que tienen inmunorreactividad del antígeno de UCpANCA. Se proporcionan también los métodos para seleccionar las bibliotecas de la presente invencion para UCpANCA y material de UCpANCA. Estas bibliotecas pueden encapsularse en partículas del bateriofago o en las células.
