Abstract

La presente divulgación proporciona guías para su uso en sistemas CRISPR de tipo V en los que dichas guías contienen bases ribonucleotídicas y al menos una base desoxirribonucleotídica. Asimismo, se describen guías Cas12 de CRISPR que contienen al menos una base desoxirribonucleotídica, así como complejos nucleproteicos de proteínas CRISPR-Cas12 de tipo V y dichas guías. Asimismo, se divulgan métodos para fabricar y utilizar los polinucleótidos y guías que contienen desoxirribonucleótidos, y para preparar y utilizar los complejos nucleoproteicos. Asimismo, se divulgan métodos para la manipulación genética de células que utilizan complejos de guía Cas12 chAR(D)N/nucleoproteína para producir células que expresan CAR; y el uso de dichas células que expresan CAR en terapia celular adaptativa.
