Abstract
La présente invention concerne un procédé pour préparer un vaccin pour lutter contre la theilériose. Ce procédé consiste à isoler des cellules lymphoïdes infectées avec des schizontes de Theileria annulata présentes chez un animal infecté, à mettre en culture les cellules infectées dans un milieu de croissance, à soumettre ces cellules en culture à l'étape de propagation in vitro par un repiquage en série, à développer ces cultures clonées à partir d'une seule cellule en limitant la technique de dilution et en repiquant à nouveau celles-ci, à établir un caryotype des cellules dans le milieu, afin de déterminer le caractère diploïde, à obtenir des cellules viables avec des taux d'infectiosité maximum, à déterminer le nombre de masse des macroschizontes, à soumettre un certain nombre de cellules viables avec le caractère diploïde à l'étape de cryoconservation dans de l'azote liquide, en présence d'un agent de cryoprotection dans le milieu de croissance, afin d'obtenir une suspension de cellules viables, à tester ces cultures aux différents repiquages et à divers niveaux de dosage dans des groupes de veaux, afin de déterminer leur pouvoir pathogène/pouvoir antigénique, à déterminer le niveau de repiquage et la dose pour la sécurité et l'efficacité d'un vaccin candidat, son utilisation sûre chez des veaux nouveau-nés et chez des vaches pleines, le temps mis pour induire l'immunité, les réponses sérologiques et l'effet sur des paramètres hématologiques, puis enfin à tester son efficacité dans des conditions à champ large sur la population susceptible d'être touchée.
