Abstract
IgE-생성 증식 세포를 비-거부 동물에 이식시켜, 동물이 이식 조직 세포에 의해 야기되는 IgE-매개된 알레르기 반응을 유발하는 능려을 갖도륵 하여 제조된 동물 모델, 및 이러한 동물 모델을 제조하는 방법 및 IgE-매개된 질환에 대한 예방제 및 치료제를 스크리닝하는 방법이 기술되어 있다. IgE-관련 질환에 대한 예방제 및 치료제의 스크리닝을, IgE-매개된 알레르기 반응이 반복된 항원 도전에 의해 반복적으로 유발되는 본 발명에 따르는 동물 모델을 사용함으로서 효과적이고 편리하게 수행할 수 있다.

제1도는 항-트리니트로페닐(TNP)ㆍIgE 생성 하이브리도마 IGELa2가 이식되거나 이식되지 않은 각각의 마우스로부터 수집한 혈액중 항-TNPㆍIgE수준의 시간 경과를 그래프로 나타낸 것이다. 제1도에서, A는 비이식된 그룰의 결과를 나타내고, B는 100개 IGELa2-이식된 그룹의 결과를 나타내고, C는 107개 IGELa2-이식된 그룹의 결과를 나타낸다.
제2도는 항-TNPㆍIgE-생성 하이브리도마IGELa2가 이식되거나 이식되지 않은 항원-도전된 마우스에서 귀의 팽창 결과를 나타낸다. 제2도에서,- □-는 대조 그룹(n=7)을 나타내고, - 0 -는 100개 IGELa2-이식된 그룹(n=7)을 나타내고, - ●-는 107개 IGELa2-이식된 그룹(n=6또는 7)을 나타낸다.
제3도는 항-TNP IgE-생성 하이브리도마 IGELa가 이식되거나 이식되지 않은 1차 항원-도전된 마우스에서 귀의 팽창 결과를 나타낸다. 제3도에서,- 0 -는 0.4% 피크릴 클로라이드(PC)-도전된 비이식된 그룹(n=12)을 나타내고, - ●-는 4%PC-도전된 비이식된 그룹(n=12)을 나타내며, - □-는 0.4%PC-도전된 IGELa2-이식된 그룹(n=12)을 나타내고, - ■-는 4%PC-도전된 IGELa2-이식된 그룹(n=11)을 나타낸다.
제4도는 항-TNP IgE-생성 하이브리도마 IGELa2가 이식되거나 이식되지 않은 2차 항원-도전된 마우스에서 귀의 팽창 결과를 나타낸다. 제3도의 실험에서 사용된 마우스를 6일 후 다시 재도전시킨다. 제4도에서, - 0 -는 IGELa2-이식된 2회 도전된 그룹(n=6)을 나타내고, - □-는 비이식된 2회 도전된 그룹(n=6)을 나타내며, -●-는 IGELa2-이식된 1회 도전된 그룹을 나타내고. - ■-는 비이식된 1회 도전된 그룹을 나타낸다.

Claims (11)
- IgE-생성 증식 세포(vegetative cell)를 비-거부 동물에 이식시켜 이식 세포에 의해 유발되는 IgE-매개된 알레르기 반응을 유도시킴으로써 제조된 동물 모델.
- 제1항에 있어서, 동물이 마우스, 래트, 기니아 피그 및 래비트로 이루어진 그룹 중에서 선택된 동물 모델.
- 제1항에 있어서, IgE-생성 증식 세포가 IGELa2(ATCC 번호 TIB142), IGELb2(ATCC 번호 TIB141), SE-1.3(ATCC 번호 HBl37) 및 U266Bl(ATCC 번호 HB196)로 이루어진 그룹 중에서 선택된 동물 모델.
- IgE-생성 증식 세포를 비-거부 동물에 이식시켜, 동물이 이식 조직 세포에 의해 유발되는 IgE-매개된 알레르기 반응을 유도시키는 능력을 갖도록 하는 단계를 포함하여, 제1항에 따르는 동물 모델을 제조하는 방법.
- 제4항에 있어서, 동물이 마우스, 래트, 기니아 피그 및 래비트로 이루어진 그룽 중에서 선택되는 방법.
