Claims (10)
- 시료중에 존재하는 바이러스 유전자를 정성·정량하는 방법에 있어서, 시료에 폴리에틸렌 글리콜을 함유하는 수용액 또는 염수용액으로된 침전액을 가하고, 와동한 후에 실온에서 방치한 후에 원심분리하여 얻은 침강물을 현탁액으로 현탁하고, DNA 프로우브와 하이브리드 형성시켜 얻어진 결과에 부착율을 곱하여 정성·정량하는 것을 특징으로 하는 바이러스 유전자의 정성·정량방법.
- 제1항에 있어서, 전기시료가 혈청, 오줌 또는 침인 것이 특징인 방법.
- 제1항에 있어서, 전기시료에 전기 침전액을 가할 때의 폴리에틸렌 글리콜의 최종농도가 5중량% 이상으로 되게하는 것을 특징으로 하는 방법.
- 제1항에 있어서, 전기 추출액과 시료를 합한 후에 방치한 시간이 10분 이하인 것이 특징인 방법.
- 제1항에 있어서, 전기 침전액과 시료를 혼합한 후 12,000rpm에서 5분 이상 원심분리하는 것을 특징으로 하는 방법.
- 제1항에 있어서, 얻어진 계측치에 부착율인 2,4 및 6을 곱하여, 시료중의 바이러스 유전자의 양을 최소치, 평균치 및 최대치로서 정량함을 특징으로 하는 방법.
- 제1항에 있어서, 계측치로부터 얻어진 바이러스 유전자 량을 하기식에 대입하여 시료중의 댄입자를 구하는 것을 특징으로 하는 방법.X÷ (2.7×10-6)×13 = 입자의 수/ml(식중, X는 시료의 계측된 값을 양성대조군과 비교하여 얻어진 DNA의 양이며, 그 단위는 pg이다)
- 혈청, 오줌 또는 침에 존재하는 바이러스 유전자를 정성·정량하는 키트에 있어서, 폴리에틸렌 글리콜을 함유하는 수용액 또는 영수용액으로 되고, 시료에 가하여 바이러스 입자를 분리 침강하기 위한 침전액과, 얻어진 분리 침강물을 현탁하는 현탁액을 함유하는 것을 특징으로 하는 바이러스 유전자의 정성·정량용 키트.
- 제8항에 있어서, 전기 침전액으로서 폴리에틸렌 글리콜의 최종 농도가 5-10% 함유하는 수용액 또는 염수용액을 함유하는 것을 특징으로 하는 키트.
- 제8항에 있어서, 전기 용해액으로서 TE 완충액[10mM Tris-HCl, 1mM EDTA(pH 8.0)], 증류수, SDS 수용액 및 폴리에틸렌 옥틸 페닐에테르 수용액으로된 군에서 선택된 1종을 15-16ml함유하는 것을 특징으로 하는 키트.※ 참고사항 : 최초출원 내용에 의하여 공개하는 것임.
