Abstract
A glycoamylase, which retained >=90% of its activity when held at 70≰C for 10 min. at pH 4.5 and had max. activity at ~ 75≰C, was isolated from a Talaromyces duponti culture. Thus, T. duponti G 45-632 was cultured for 7 days at 40≰C in a yeast ext. starch agar medium and the glucoamylase was isolated from the culture, after filtering off the mycelia. The glucoamylase was purified by DEAE-cellulose, CM-cellulose, and sephadex G-150 chromatog. Finally, the purified glucoamylase was immobilized by binding with AE-cellulose using glutaraldehyde. Immobilized glucoamylase had 2,000 units of activity per g carrier. In a saccharification test, using a 25% soln. of starch hydrolysate at 60≰C, immobilized glucoamylase produced a saccharified soln. contg. 96.5% dextrose.
