Abstract

실시예
방법 및 재료
표 1은 해당 펩타이드, 폴리펩타이드 및 융합 단백질의 서열번호를 보여준다.
유세포 분석. 비장 세포를 37℃에서 2시간 동안 항원성 자극제로 자극한 다음, 2시간 동안 37℃에서 50㎍/㎖의 브레펠딘(Brefeldin) A 및 모넨신(Monensin)으로 처리하였다. 세포를 수확하고, 0.5% BSA 함유 PBS로 세척하고, 동시에 APC/Cy7-접합 항-CD3 항체, PerCP/Cy5.5-접합 항-CD4 항체, FITC-접합 항-CD8 항체, PE-접합 항-CD44 항체 및 APC-접합 항-CD62L 항체로 염색하였다. 세척 후, 세포를 투과화하고, 고정시키고 동시에 PE-접합 항-IFN-γ 항체, PE/Cy7-접합 항-IL-2 항체 및 eFluor450-접합 항-TNF-α 항체로 세포 내부적으로 염색하였다. 효소-결합 면역점(ELISpot) 분석. 비장 세포를 항원성 자극제의 존재 또는 부재 하에서 2×105 세포/웰의 세포 밀도로 전처리한 쥐 IFN-γ 포획 96-웰 플레이트(CTL IMMUNOSPOT®)에 3반복으로 접종하였다. 37℃ 배양 24시간 후 세포를 버렸다. 세척 후, 2시간 동안 상온에서 비오틴-접합 항-쥐 IFN-γ 항체를 사용하여 포획된 IFN-γ를 검출하였으며 제조사의 지침에 따라 IFN-γ-면역점을 전개하였다. IMMUNOSPOT® S5 마이크로 분석기(CTL)를 사용하여 IFN-γ-면역점의 스캐닝 및 계수를 수행하였다.
간접 효소-결합 면역흡착 분석(ELISA). 수집한 전혈 시료를 30 내지 60분 동안 4℃에 그대로 둔 다음 10분 동안 5,000g로 원심분리하여 응혈을 펠렛화하였다. 혈청 시료를 -20℃에 보관하였다. 항원-특이적 항체 결합을 위해 정제한 코팅 단백질을 구아니딘 코팅 완충제(2M 구아니딘 염산, 500mM Na2HPO4, 25mM 구연산염, pH 4.0 내지 4.4)로 희석하고 96-웰 플레이트에 1㎍/웰로 분배하였다. 4℃에서 밤새 항온처리한 다음, 96-웰 플레이트를 1% BSA 함유 PBS로 37℃에서 1시간 동안 블로킹하였다. 혈청 시료를 해동한 다음, 1% BSA 함유 PBS로 10배 연속 희석하였다. 코팅 단백질을 100㎕의 1000배 희석된 혈청 시료와 함께 37℃에서 2시간 동안 항온처리하였다. 인산염 완충 염수 TWEEN®-20(PBST)으로 4회 세척한 다음, 항원-특이적 항체를 겨자무 과산화효소(horseradish peroxidase: HRP)-접합 염소 항-생쥐 IgG(1:10,000 희석, Cat#31430, Thermo Fisher Science)로 37℃에서 30분 동안 검출하였다. PBST로 4회 세척한 다음, HRP-매개 발색을 100㎕의 TMB 기질의 존재 하에서 촉매화하였고 100㎕의 1N HCl로 반응중지시켰다. 혈청 시료 내 항원-특이적 항체의 상대적 역가를 450㎚에서의 흡광도로 결정하였다.
통계적 분석. t-검정을 사용하여, p<0.05일 때 결과를 유의한 것으로 간주하였다.
실시예 1
발현 벡터의 제작

E7을 다양한 병원체 또는 암 기원의 임의의 다른 관심 항원(들)으로 대체하고 도 4의 플라스미드에 삽입하여 임의의 관심 항원을 포함하는 융합 단백질을 발현하기 위한 도 3과 유사한 발현 벡터를 생성할 수 있다.

실시예 2
단백질 발현
발현 벡터를 보유한 대장균 BL21 세포를 37℃에서 선별 항생제를 함유하는 ZY 배지(10g/ℓ 트립톤 및 5g/ℓ 효모 추출물)에서 배양하였다. 배양물이 초기 로그 단계(OD600=2 내지 5)에 도달했을 때, 아이소프로필-1-티오-β-D-갈락토파이란노시드(isopropyl-1-thio-β-D-galactopyranoside: IPTG)(0.5 내지 2mM)로 융합 단백질의 발현을 유도하였다. IPTG 유도 4시간 후 세포를 수확하고 초음파 처리로 분쇄하였다. 봉입체(inclusion body)를 분리하고 가용화 완충액(6M 구아니딘 염산염, 20mM 인산 칼륨, 500mM NaCl, 20mM 이미다졸, 1mM DTT, pH 7.4)에 용해하여 과발현된 융합 단백질을 회수하였다. 정제 후, 20 내지 50배 부피의 투석 완충액(10mM PBS)으로 4℃에서 밤새 투석하여 융합 단백질의 리폴딩(refolding)을 수행하였다. 리폴딩된 융합 단백질을 환원(다이티오트레이톨(dithiothreitol) 존재 하; +DTT) 및 비-환원(다이티오트레이톨 부재 하; -DTT) 조건 하에서 SDS-PAGE 분석하여 적절하게 리폴딩되었는지를 평가하였다.
실시예 3
융합 단백질의 면역원성 분석
정제한 CD40L47-261-TPE-E7, 18sCD40L-TPE-E7, E7-TStx-18sCD40L 및 RAP1-E7 융합 단백질의 면역원성 분석을 추가로 수행하여 이들의 생물학적 활성을 평가하였다.
암컷 C57BL/6NCrlBltw 생쥐(5 내지 6주령)를 무작위로 다음과 같은 5개의 군(n=5)으로 나누었다: (A) 위약(즉, PBS); (B) CD40L47-261-TPE-E7(100㎍) 융합 단백질; (C) 18sCD40L-TPE-E7(100㎍) 융합 단백질; (D) E7-TStx-18sCD40L(100㎍) 융합 단백질; 및 (E) RAP1-E7(100㎍) 융합 단백질. 융합 단백질을 PBS로 투석하였다. CpG1826(50㎍)을 동물군 B 내지 E에 보조제로 사용하였다. 각 군을 0일에서 부터 7일 간격으로 3회 피하(s.c.) 면역화하였다. 0, 7 및 14일째에 혈액 시료를 수집하였다. 21일째에, 혈액 시료를 수집하고 비장 세포를 FBS(10%) 및 PSA 함유 RPMI 1640 배지에 재현탁하였다.
비장 세포를 사용하여 항원 자극의 존재 및 부재 하에서 CD8+ 및 CD4+ 기억 T 세포의 세포내 사이토카인 유도(IFN-γ, IL-2 및 TNF-α)를 분석하였다. 간략하게, 각 동물군으로부터의 비장 세포를 항원 E7 단백질(2㎍/㎖의 HPV16 E7 펩타이드 풀)로 또는 이 단백질 없이 처리하고 유세포 분석으로 분석하였다. 각 생쥐군의 세포내 사이토카인 유도의 정도 또는 수준을 상대적인 사이토카인 유도로 나타내었으며, 이는 자극 항원 E7의 존재 하에서 사이토카인+/CD8+ 및 사이토카인+/CD4+ 비장 세포의 빈도를 자극하지 않은(처리하지 않은) 대조군의 것에 대해 정규화하여 얻었다.

혈액 시료를 사용하여 ELISA로 혈청 HPV16 E7-특이적 및 HPV18 E7-특이적 항체의 수준을 분석하였으며, 여기서 정제한 HPV16 E7 및 HPV18 E7 재조합 단백질을 각각 코팅 단백질로 사용하였다.
도 6은 HPV16 E7 펩타이드 풀을 사용한 비장 세포의 항원 자극 후 사이토카인 유도를 보여준다. CD40L47-261-TPE-E7, 18sCD40L-TPE-E7 또는 E7-TStx-18sCD40L 융합 단백질로 면역화한 동물로부터의 CD8+ 기억 T 세포에서의 IFN-γ 및 TNF-α의 상대적 사이토카인 유도는 RAP1-E7 융합 단백질 또는 위약을 처리한 동물로부터 것에 비해 유의하게 증가한 것으로 나타났으나, IL2의 경우는 그렇지 않았다. 융합 단백질로 처리한 동물군으로부터의 CD4+ 기억 T 세포에서의 IFN-γ, IL2 또는 TNF-α의 상대적 사이토카인 유도는 위약군에 비해 약간, 하지만 유의적이지는 않게 증가한 것으로 나타났다.
이는 본 발명의 융합 단백질이 HPV16 E7 항원의 자극에 반응하여 CD8+ 기억 T 세포에서 IFN-γ와 TNF-α의 분비를 유도함에 있어서 기존 기술의 융합 단백질에 비해 우수하다는 결론을 도출하였다.
도 7은 시험관 내에서 수행한 것으로서 HPV16 E7 펩타이드 풀로 자극한 비장 세포의 IFN-γ+ 면역점을 보여준다. CD40L47-261-TPE-E7, 18sCD40L-TPE-E7, E7-TStx-18sCD40L 또는 RAP1-E7로 면역화한 동물군으로부터의 IFN-γ-분비 비장 세포의 빈도는 위약군에 비해 유의적으로 증가하였다. 특히 E7-TStx-18sCD40L은 CD40L47-261-TPE-E7보다 유의적으로 높은 빈도의 IFN-γ-분비 세포를 유도하였다(p=0.035).
이러한 결과는 본 발명의 융합 단백질이 항원성 HPV16 E7 펩타이드 풀을 사용한 자극 시 또는 자극 후 IFN-γ-분비 T 세포 개체군을 유의적으로 증가시킬 수 있음을 나타낸다.
도 8은 0, 7 및 14일째에 다양한 융합 단백질로 면역화한 동물에서의 혈청 HPV16 E7-특이적 항체 수준의 결과를 보여준다. CD40L47-261-TPE-E7, 18sCD40L-TPE-E7 또는 E7-TStx-18sCD40L로 접종한 동물의 혈청 HPV16 E7-특이적 항체 수준은 7일째의 2차 접종 후에 증가하기 시작하여, 14일째의 3차 접종 후에 더욱 증가하였고, 21일째에는 위약 또는 RAP1-E7로 처리한 동물보다 높았다.
RAP1-E7(RAP1-CD28convPEt- E7-K3) 융합 단백질은 2번의 접종(0일째 및 7일째) 후 HPV16 E7-특이적 항체 수준을 유도하는데 실패하였다. 14일째의 3차 접종 후 혈청 HPV16 E7-특이적 항체를 유도하기 시작하였고, 혈청 항체 수준은 21일째에 앞서 언급한 본 발명의 융합 단백질로 처리한 동물에 비해 단지 미미한 정도였다. 위에 기술된 것과 같은 요법 및 면역화 일정을 사용하여 동물을 RAP1-CD28convPEt-HBx-K3("RAP1-HBx"로 지칭함)로 접종하였을 때 HBx-특이적 항체를 유도하는 것에서도 유사한 패턴이 관찰되었다. RAP1-E7에서 E7 항원을 HBx 항원으로 대체하여 RAP1-HBx 융합 단백질을 생성하였다. RAP1-HBx 융합 단백질은 14일째의 3차 접종 후 혈청 HBx-특이적 항체 수준을 유도하였고, 21일째의 혈청 항체 수준은 단지 미미한 정도였다(데이터 미제시).
대조적으로, 본 발명의 융합 단백질은 0 및 7일째의 2회의 접종 후 혈청 HPV16 E7-특이적 항체 수준을 유도하였다(도 8).
도 9는 0, 7 및 14일째에 다양한 융합 단백질로 면역화한 동물에서의 혈청 HPV18 E7-특이적 항체 수준을 보여준다. CD40L47-261-TPE-E7, 18sCD40L-TPE-E7 및 E7-TStx-18sCD40L 융합 단백질은 혈청 HPV18 E7-특이적 항체 수준을 위약군에 비해 유의적으로 증가시켰다. 위에 기술된 HBx-preS1-TStx-18sCD40L 융합 단백질의 면역원성 분석을 수행하였다. 이의 결과, HBx-보유 융합 단백질은 적어도 2회의 면역화 후 HBx-특이적 T 세포-매개 및 체액성 면역 반응을 효과적으로 유도할 수 있는 것으로 나타났다(데이터 미제시).

실시예 4
효능 분석
HPV 감염 생쥐 모델에서 융합 단백질과 면역 관문 억제제의 병용
CD40L47-261-TPE-E7, 18sCD40L-TPE-E7 및 E7-TStx-18sCD40L 융합 단백질을 각각 면역 관문 억제제 항체(항-PD-1 항체)와 병용하고, 생쥐 HPV16 종양 모델에서 각 병용물의 효능에 대해 시험하였다.
암컷 C57BL/6NCrlBltw 생쥐(5 내지 6주령)를 무작위로 다음과 같은 5개의 군으로 나누었다: (A) 위약(PBS, n=4); (B) 항-PD-1 항체(100㎍; 카탈로그 번호 BE0146, Bio X Cell, Inc.)와 CD40L47-261-TPE-E7(25㎍)의 병용(n=5); (C) 항-PD-1 항체(100㎍)와 18sCD40L-TPE-E7(25㎍)의 병용(n=4); (D) 항-PD-1 항체(100㎍)와 E7-TStx-18sCD40L(25㎍)의 병용(n=5); 및 (E) 항-PD-1 항체 단독(100㎍)(n=5). B군 내지 D군에서, 융합 단백질을 PBS에 용해시키고 CpG1826(50㎍)을 동물 면역화에 보조제로 사용하였다. 도 10은 면역화 일정, 병용 요법 및 투여량을 보여준다.
C57BL/6 생쥐의 폐 상피 세포로부터의 HPV16 E6- 및 E7-발현 종양 세포주(TC-01)를 사용하여 생쥐 HPV16 종양 모델을 확립하였다. 종양 세포는 FBS(10%) 및 페니실린/스트렙토마이신/암포테리신 B(50유닛/㎖) 함유 RPMI 1640 배지에서 37℃, 5% CO2에서 배양하였다. 생쥐에 적용하기 위해, 0일째에 각 생쥐의 좌측 옆구리에 종양 세포(0.1㎖ 내 1×105)를 피하 주사하였다. 7, 14 및 21일째에 각 군의 생쥐에 시험 물품(즉, 위약, 항-PD-1 항체 또는 병용 요법)을 3회 투여하였다. 각 투여에서, 융합 단백질은 피하로 제공하였고, 항-PD-1 항체는 복강 내로 제공하였다. 생존한 모든 생쥐는 39일째에 희생시켰다.
종양 크기를 다음의 변형 타원체 공식을 기반으로 캘리퍼 측정값을 곱하여 1주에 2회 결정하였다: 종양 부피 = 1/2(길이×폭 2 ). 생존율 및 무종양 비율을 계산하였다. 길이가 2㎝를 초과하는 종양이 있는 생쥐는 죽은 것으로 간주하였고 측정 가능하거나 감지 가능한 종양 덩어리가 없는 생쥐는 무-종양으로 간주하였다. 각 비교의 유의성은 t-검정을 사용하여 계산하였고, p<0.05인 경우 결과를 유의한 것으로 간주하였다.
접종한 종양이 위약군에서 빠르게 발달하여 25일째에 2마리가 죽었고 이에 따라 21일째까지만 위약군의 데이터를 나타내었다(도 11). CD40L47-261-TPE-E7, 18sCD40L-TPE-E7 또는 E7-TStx-18sCD40L과의 병용으로 항-PD-1 항체를 제공받은 동물군의 종양 덩어리는 적어도 전체 실험 기간 동안에는 거의 완전하게 억제되었다(마지막날은 39일째임). 항-PD-1 항체만을 제공받은 동물군의 종양은 초기에는 잘 제어되었지만, 면역화를 중단한 후에 빠르게 성장하였다. 이러한 결과는 본 발명의 병용물이 종양 성장을 효과적으로 억제할 수 있음을 나타낸다.
위약군에서 모든 생쥐가 35일째에 죽었고, PD-1 만을 제공받은 군은 35일째에 20%의 생존율로 감소한 반면, 각각 18sCD40L-TPE-E7 및 E7-TStx-18sCD40L과의 병용으로 항-PD-1 항체를 제공받은 동물군은 39일째에 100%의 생존율을 유지하였고, CD40L47-261-TPE-E7과의 병용으로 항-PD-1을 제공받은 군은 39일째에 80%의 생존율을 유지하였다(도 12). 이러한 결과는 본 발명의 병용물이 동물 종양 모델에서 생존율을 효과적으로 유지할 수 있음을 나타낸다.
전체 실험 기간 동안(39일째) 위약군 및 PD-1 단독 처리군에서 무-종양 동물을 발견할 수 없었다(도 13). 특히, 본 발명의 병용물을 제공받은 동물군에서 한마리가 측정 가능하거나 감지 가능한 종양없이 생존하고 있음을 발견하였다. 이들 무-종양 생쥐에서 종양 덩어리는 본 발명의 병용물로 3회 면역화를 완료한 후 바로 모두 없어졌다. 따라서, 본 발명의 병용물은 종양의 성장을 강력하게 억제하고 뛰어난 치료 효능을 나타낼 수 있다.

위에 기술된 바와 같이 확립된 생쥐 HPV16 종양 모델에서 E7-TStx-18sCD40L 융합 단백질과 면역 관문 자극제 항체(항-CD137 항체)의 병용을 시험하였다.
동물군 나누기 및 면역화 일정은 표 2에 나타내었다. 암컷 C57BL/6NCrlBltw 생쥐(5 내지 6주령)를 무작위로 다음과 같은 4개의 군(각 군의 n=5)으로 나누었다: (1) A군(위약; PBS); (2) B군(E7-TStx-18sCD40L); (3) C군(항-CD137 항체; 카탈로그 번호 BE0239, Bio X Cell, Inc.); 및 (4) D군(E7-TStx-18sCD40L과 항-CD137 항체의 병용).
생쥐에 적용하기 위해, 0일째에 각 생쥐의 좌측 옆구리에 0.1㎖에 1Х106의 높은 농도를 갖는 HPV 종양 세포를 피하 접종하였다. 위약군에서, 14 및 21일째에 PBS를 제공하였다. 각각의 접종(표 2)을 위해, E7-TStx-18sCD40L 융합 단백질을 PBS에 용해시키고, CpG1826(50㎍/1회 투여량)을 첨가한 후 피하 주사하였다. 항-CD137 항체는 복강 내로 제공하였다. 모든 생쥐는 41일째에 희생시켰다.
종양은 위약 및 항-CD137 항체 단독 처리군에서 빠르게 발달하였고, 30일째 부근에 모든 생쥐가 죽어서, 더 이상 데이터를 기록할 수 없었다. 놀랍게도, 항-CD137 항체 단독 처리군은 여기서 종양 억제에 기여하지 않는 것으로 나타났다(도 14).
E7-TStx-18sCD40L을 단독으로 제공받은 군에서 종양 덩어리는 초기에는 잘 제어되었지만, 두 번째의 면역화 후 7일째에 천천히 성장하기 시작하였다. 특히, 단 2회의 접종이 이루어졌고 더 많은 양의 종양 세포를 접종하였음에도(앞서 언급된 PD-1 병용 연구에서 사용된 양의 10배) 융합 단백질의 종양 억제 효과가 마지막 면역화 후 적어도 1주 동안 유지되었다.
항-CD137 항체와 병용하여 E7-TStx-18sCD40L을 제공받은 군에서 종양 덩어리가 억제되었고 42일째에 실험이 끝날 때까지 지속적인 종양 위축이 발견되었다. 관찰 기간이 더 길어지면 종양 덩어리가 완전하게 제거될 수 있음을 합리적으로 추측할 수 있었다. 기대와 달리, 항-CD137 항체는 E7-TStx-18sCD40L 융합 단백질에 상승적 효과를 나타냈다. 이러한 결과는 본 발명의 병용물이 종양의 성장을 효과적으로 억제할 수 있음을 나타낸다.
E7-TStx-18sCD40L을 제공받은 군이 40%의 생존율로 감소하였고, 위약 및 항-CD137 항체 단독 처리군 모두 0% 생존율로 감소한 반면, 항-CD137 항체와 E7-TStx-18sCD40L의 병용물을 제공받은 군은 마지막 관찰일까지 100%의 생존율을 유지하였다(도 15). 이러한 결과는 본 발명의 병용물이 생존율을 효과적으로 유지할 수 있음을 나타낸다.
따라서, 항-PD-1 길항제 항체 또는 항-CD137 작용제 항체와 같은 T 세포를 활성화할 수 있는 면역 관문 항체와 E7-보유 융합 단백질의 병용물은 우수한 종양 억제 효과를 나타낼 수 있으며 종양 또는 비정상적인 세포 증식을 치료하는데 사용될 수 있다.

효능 분석: HPV 감염 생쥐 모델에서 HBx-preS1-T

-18sCD40L 융합 단백질과 면역 관문 항체의 병용
HBV-감염 생쥐 모델에서 면역 관문 억제제(항-PD-1 항체) 또는 면역 관문 자극제(항-CD137 항체)와 HBx-preS1-TStx-18sCD40L의 병용물의 효능에 대해 시험하였다.
생쥐를 다음과 같은 몇개의 군으로 나누었다: 대조군(PBS), 백신군(HBx-preS1-TStx-18sCD40L), PD-1군(항-PD-1 항체), CD137군(항-CD137 항체), PD-1 콤보군(항-PD-1 항체 및 HBx-preS1-TStx-18sCD40L) 및 CD137 콤보군(항-CD137 항체 및 HBx-preS1-TStx-18sCD40). 융합 단백질에 CpG1826을 첨가하고 피하로 제공하였다. 항체는 복강 내로 제공하였다.
HBV-감염 생쥐 모델은 미국 특허 제10,058,606 B2호에 기술된 AAV-HBV 생쥐 모델이며 필요한 경우 적절하게 변형할 수 있다. 간략하게, 수컷 생쥐(5 내지 6주령)을 사용하여 장기간 B형 간염 바이러스를 보유한 동물 모델을 확립하였다. pAAV/HBV1.2 플라스미드와 염수의 혼합물을 고속 모드에서 고압(유체 역학 주사: HDI)으로 생쥐 꼬리 정맥에 정맥 주사하여 플라스미드가 세포막을 관통하여 간세포에 진입하도록 하였다. 플라스미드-보유 간세포는 B형 간염 바이러스 단백질을 발현한다. 바이러스는 간세포 안에서 조립되어 혈액으로 방출될 것이다. 이러한 동물 모델은 만성 B형 간염 증상으로 고통받는 인간 환자를 모방한다.
고압 주사 28일 후, 7일 간격으로 0, 7, 14일째에 3회 투여의 융합 단백질 및/또는 표시된 항체를 각 군의 생쥐에 제공하였다. 고압 주사와 같은 날, 그리고 0, 7, 14, 21, 32 및 42일째에 체중을 측정하였다. 알라닌 아미노기전달효소(ALT), 빌리루빈, 바이러스 DNA 및 표면 항원(HBsAg)의 분석을 위해 혈액을 수집하였다. 첫 번째 접종 후 82일째에 생쥐를 희생시키고 간 핵심 항원(HBcAg)의 양을 분석하였다.
항-PD-1 항체 또는 항-CD137 항체와 병용한 HBx-preS1-TStx-18sCD40의 투여는 HBV 감염 상 뛰어난 치료 효과를 나타낼 것으로 기대된다. 예를 들어, 상기 병용은 바이러스 DNA, HBsAg 및 HBcAg를 감소시키고, HBx-특이적 면역 반응을 유도할 것으로 기대된다. 상기 병용(즉, HBx-보유 융합 단백질과 면역 관문 조절제의 병용)의 사용은 간세포에서의 B형 간염 바이러스의 증식과 HBV 환자에서의 B형 간염 바이러스의 감염을 효과적으로 억제할 것으로 기대된다.
요약하면, 새로운 항원-보유 융합 단백질은 이의 독특한 단백질 설계 및 작용 기작으로 인해 강력한 항원-특이적 T 세포 면역 반응을 유도할 수 있다. 면역 관문 조절제는 억제성 면역 관문(예를 들어, PD-1, PD-L1, PD-L2, CTLA-4, LAG3, TIGIT, CD96, CD122R, TIM3, VISTA, CEACAM1, SIGLEC-7, SIGLEC-9, SIGLEC-15, KIRi, CD200R, BTLA 및 ILT2)에 길항(antagonize)하여 T 세포 기능을 복구하거나 자극성 면역 관문(예를 들어, CD137, OX40, GITR, ICOS, CD27, CD28, CD40, KIRs, CD226 및 CD244)에 작용(agonize)하여 T 세포를 활성화할 수 있다. 적어도 하나의 적합한 항원이 융합 단백질에 적용되고 적합한 면역 관문 조절제가 선택되는 경우, 이들 모두 T 세포 면역력 향상에 기여하고 이들이 병용으로 사용되는 경우 우수한 치료 효능 달성할 수 있었다. 따라서, 본 발명은 종양 및 감염성 질환을 치료하기 위한 임의의 기존 공개문헌에 개시되지 않은 병용을 사용하는 독창적인 발명적 개념을 제공한다.
