새로운 유전자 재조합 백신이 3가지 지역형 바이러스에 대한 방어능을 시험하기 위해 C57BL/6 암컷 쥐를 이용하여 PD50 측정하는 실험을 진행하였다. JC-VP1, PA2-VP1 백신을 마우스에 면역 (목적동물을 위한 항원량의 1/10, 1/40, 1/160, 1/640)후 7일만에 각 목적하는 바이러스 ME-SA (O Vet), SEA(O JC), Cathay (O TWN97) 바이러스의 역가 100LD50 를 공격접종하여, 그 생존율과 체중 감소 등을 확인하여 공격접종으로 인한 영향력 즉, 방어 효과를 확인하였다. <실험예3> 본 발명에 따른 백신의 면역원성 평가<Experimental Example 3> Immunogenicity evaluation of the vaccine according to the present invention

소의 면역시험의 경우 동물 중 가장 표준이 되는 동물실험법으로 알려져 있으며, 일반적으로 백신으로 면역 후 3-4주 혈액을 가지고 백신이 야외주와 매칭되는지를 2D 중화시험을 통하여 확인하였다. 따라서 소에서의 면역실험의 경우 마우스 및 돼지에서 시험하는 백신과 동일하게 제작되었으며 1ml/ dose을 접종하여 항체 수준을 비교하였다. PA2-VP1, O JC-VP1 1회만 소에 면역한 후 8주까지 항체 수준을 확인하였다. 그 결과 접종 후 14일째 그 항체 수준은 3종 항원 모두 매우 높은 수준을 보였고 시험기간 동안 높은 역가로 유지하였다. PA2-VP1의 경우는 소에서 1주 만에 중화항체 반응성이 우수하였다. 1차 접종만으로 높은 항체 수준을 유지됨이 확인되었다. JC-VP1의 경우는 소에서 2주 만에 높은 중화항체를 유지하였으며, 1차 접종만으로도 높은 항체 수준을 유지하였다(도 3의 a, b). 돼지 면역원성 시험을 통해서 백신의 면역성을 평가하는 것은 특히 한국 상황에서는 백신의 현장에서의 유효성을 평가하는 중요한 척도이다. 따라서 돼지에서의 실험을 통해 백신의 효용성을 판단하기 위해 그룹 당 5마리 면역실험을 통해 그 효과를 평가하고자 하였고, 마우스, 소에서의 면역 실험 등에 사용된 백신 (1ml/dose)을 1회 접종 후 4주후 2회 접종하고, 4주까지는 매주 채혈하였고 이후 2주 간격으로 채혈하여 그 면역 반응성을 확인하였다(도 3의 c, d).Evaluating the immunity of vaccines through swine immunogenicity testing is an important criterion for evaluating the effectiveness of vaccines in the field, especially in Korea. JC-VP1 백신 접종한 그룹에서 BE(ME-SA) 타입으로 공격접종하였을 때 50% 방어율을 보였으나(ME-SA #4~7), 20ug(ME-SA #12~15) 으로 항원량을 증가하여 백신 접종을 하면 100%로 방어율을 올릴 수 있었다(도 5b). 이종(Heterologous) VN 역가의 경우 예상한 대로 JC(SEA type) 공격접종 그룹에서는 JC-VP1 백신접종 그룹에서 제일 높게 나타났으며(도 6의 a), BE(ME-SA) 공격접종 그룹에서도 같은 ME-SA type 인 PA2-VP1 백신을 접종한 그룹에서 가장 높게 나타났다(도 6의 b).
