Abstract
본 발명에 따른 약학적 조성물은 전신경화증의 예방 또는 치료에 유용하게 사용할 수 있다.
Description
본 발명은 전신경화증의 예방 또는 치료용 약학적 조성물을 제공하기 위한 것이다.
전신경화증(systemic sclerosis; SSc)은 피부, 혈관, 내부 장기의 비후(두꺼워짐)나 경화(딱딱해짐)를 일으키는 퇴행성 질환으로서, 자가면역 류마티스 질환에 속한다. 이 질환은 만성적이고 오랜 기간에 걸쳐 진행될 수 있다. 전신경화증의 원인은 아직 밝혀지지 않았지만 특정한 화학물질(톨루엔, 벤젠, 비닐 클로라이드, 실리카 등)도 발병과 연관이 있다는 보고도 있으며, 병력, 이학적 검사, 조직 검사, 혈액 검사를 종합하여 진단한다. 상부 하부 위장관 검사, X-ray 검사, 폐 기능 검사, 심전도, 심초음파 검사 등으로 장기 내부의 손상 정도를 확인할 수 있다.
전신경화증은 일반적으로 예후가 나쁘지 않으나, 폐, 신장, 또는 심장으로 전이되는 경우 생명을 위협할 수 있다. 간질성 폐 질환 및 폐동맥 고혈압이 진행되어 호흡 곤란 등이 일어날 수 있으며, 심각한 고혈압을 동반한 신장 위기가 발생할 수 있다.
전신경화증은 내부의 면역체계의 오류에 의해 발생되는 자가면역질환의 한 종류이다. 전신경화증은 류마티스내과에서 진료, 자가면역저해에 의한 다양한 증상을 호전하기 위한 치료를 진행한다. 전신경화증은 피부 침범 부위 정도에 따라 제한성(limited) 또는 미만성(diffuse cutaneous) 전신경화증으로 크게 분류되는데, 전신경화증 환자의 일부는 면역조절 장애와 함께 전신경화증 연관 폐질환(SSc-ILD) 증세가 나타난다. 전신경화증 연관 폐질환은 주로 미만성 전신경화증에서 보다 흔하게 관찰된다. 또한, 이 중 25 ~ 30%의 환자는 진행성 전신경화증 연관 폐질환 (progressive SSc-ILD)으로 발전되어 전신경화증 환자의 주요 사망 원인으로 알려져 있다(Lancet, 2017; 390:1685-1699).
전신경화증 연관 폐질환은 전신경화증 환자에게서 발생하는 호흡기 질환으로, 호흡기내과에서 진료, 폐기능을 개선하기 위한 치료를 진행한다. 전신경화증 연관 폐질환은 전신경화증 환자의 조기 사망률과 관련이 높고, 미국내 조사 결과 전신경화증 환자의 폐 섬유증에 의한 사망률이 대략 20년 사이 6%에서 33%로 5배 이상으로 증가하여 간질성 폐질환이 전신경화증 관련 사망의 가장 빈번한 원인이 되었고, EULAR의 조사 결과에서도 35% 이상의 전신경화증 환자의 사망률에 영향을 주고 있음을 밝혔다. 전신경화증이 있는 환자에서 간질성 폐질환의 발병 또는 진행에 대한 위험 요소에는 dcSSc(diffuse cutaneous systemic sclerosis)의 존재, 아프리카계 미국인, 발병 시 나이(고령), 더 짧은 질병 기간 및 anti-Scl-70/anti-topoisomerase I 항체 및 anti-centromere 항체의 부재 등 아직까지도 밝혀지지 않은 다양한 이유에 의해서 간질성 폐질환 발병 조건이 이루어졌을 때 발병한다. 그러나 이러한 위험 요소 중 어느 것도 절대적인 것은 아니며, 전신경화증이 유발된다고 해서 무조건 전신경화증 연관 폐질환이 유발되는 것이 아니다. 또한, 모든 환자가 호흡기 증상을 보이는 것은 아니므로 진단에 의사와 연구자들은 다양한 방법을 통해 이를 조기 진단 및 치료제 개발에 열을 올리고 있다(Respiratory Research volume 20, Article number: 13 (2019)).
특히, 최근 임상시료를 사용한 연구들을 보면, 전신경화증 연관 폐질환은 전신경화증 환자로부터 유래하지만 혈액내 다양한 cytokine 및 IGFBP-1, MMP-9, 및 H3.1 등과 같은 다양한 Biomarker의 유의미한 차이를 보이고 있어 환자의 질환적 특성이 다름을 보여주는 것으로 생각된다. 이러한 연구들은 현재 전신경화증 코호트에서 전신경화증 연관 폐질환을 진단하기 위한 Biomarker로 활용하기 위해 지속적을 진행되고 있으며, 이는 두 질병이 연관성이 높지만 독립된 분류로 볼 수 있는 기반이 될 수 있는 데이터이다. 또한, MMP-7 및 MMP-9이 혈액내 특이적으로 전신경화증 연관 폐질환 환자에서 전신경화증 환자보다 증가되어 있다는 사실은 폐 섬유화적 발병의 이유 및 1/3에 이르는 전신경화증 연관 폐질환 환자의 빠르고 높은 사망률을 뒷받침하는 증거일 뿐만 아니라, 전신경화증 연관 폐질환과 전신경화증을 임상적 뿐만 아니라 병리학적으로도 구분할 수 있는 근거로 해석된다((i) Clin Epigenetics. 2020 Aug 17;12(1):124, (ii) Sarcoidosis Vasc Diffuse Lung Dis. 2015 Sep 14; 32(3):228-36, (iii) European Respiratory Journal 2021 58: 2101560).
전신경화증은 현재까지 병증의 진행을 중단시킬 수 있는 치료법이 없으며, 일부 약물을 통해 특정 증상을 완화하고 기관 손상을 감소시킬 수 있다. 비스테로이드성 항염증제(NSAID)가 관절 통증 완화에 도움이 되며, 칼슘 통로 차단제는 레이노 현상의 증상을 완화시킬 수 있으나, 위장 문제를 야기할 수 있다. 코르티코스테로이드를 통해 근육염 증상 완화, 면역억제제를 통한 폐 염증 완화, 고혈압 약물을 통한 급성 신장 손상 및 혈압 상승 완화할 수 있으나, 일부 증상의 완화 효과에 불과한 한계점이 있으며, 전반적인 증상 완화 약물은 존재하지 않는다.
전신경화증 관련 폐질환에 대해서는 오페브®(닌테다닙, Nintedanib) 및 악템라®(토실리주맙, tocilizumab)가 치료제로 허가되었다. 닌테다닙은 특발성폐섬유증 치료제로 폐섬유증 환자의 폐기능을 개선하는 것으로 알려져 있다. 하지만, 닌테다닙은 폐섬유화 개선에만 효과가 있고 피부경화 증상에 대해서는 치료효과 입증에 실패하여, 이미 진행형 간질성 폐질환이 동반되는 말기 전신경화증 연관 폐질환 환자에 폐기능 개선을 위한 보조 치료제로만 처방이 되고 있다. 토실리주맙은 류마티스 관절염을 치료하기 위한 면역억제제로 류마티스 질환의 증상을 완화해주는 것으로 알려져 있다. 하지만 토실리주맙은 섬유화 완화/개선을 입증하지 못하여, 심각한 폐기능 저해가 아직 발생하지 않은 초기 단계의 전신경화증 연관 폐질환 환자에게 예방 차원의 보조 치료제로 사용되고 있다. 즉, 허가된 두 약물 모두 전신경화증은 물론, 전신경화증 동반 폐질환 환자들에게 근본적인 치료 효과를 제공하지 못하여 first-line treatment로 처방되고 있지 않고, 여전히 환자들은 cyclophosphamide, mycophenolate와 같은 부작용이 심한 면역억제제에 의존해야한다.
한편, PRS(prolyl-tRNA synthetase)는 아미노아실-tRNA 합성효소(aminoacyl-tRNA synthetase; ARS) 패밀리 효소군 중의 하나로서, 단백질 합성을 위해 아미노산을 활성화시키는 역할을 한다. 즉, ARS는 아미노아실 아데닐레이트(AA-AMP)를 형성한 후 활성화된 아미노산을 대응하는 tRNA의 3번 말단으로 이동시키는 역할(translational function)을 수행한다. ARS는 단백질 합성에 핵심적인 역할을 수행하기 때문에, ARS 저해는 모든 세포의 생장과 성장을 억제한다. 이에 따라, ARS는 항생제나 세포 과발현을 억제해야 하는 질병 치료제의 유망한 타겟으로 인식되고 있다(Nature, 2013, 494: 121-125).
PRS는 EPRS(Glutamyl-Prolyl-tRNA Synthetase) 형태의 다중합성효소 복합체(multisynthetase complex, MSC) 상태로 존재하고 기능한다. 특히, 다양한 MSC 중에서도 EPRS는 혈관신생(angiogenesis)의 핵심인자인 VEGF A(vascular endothelial growth factor A)의 생성을 억제하는 전사침묵자(translational silencer)로서 기능을 하며, 또한 다양한 고형암과 밀접한 관련성이 있다고 보고되었다(Nat. Rev. Cancer, 2011, 11, 708-718).
이에 본 발명자들은 전신경화증의 예방 또는 치료 방법을 예의 연구한 결과, 후술할 특정 PRS 저해제가 전신경화증의 예방 또는 치료에 유용함을 확인하여, 본 발명을 완성하였다.
본 발명은 전신경화증의 예방 또는 치료에 유용하게 사용할 수 있는 약학적 조성물을 제공하기 위한 것이다.
상기 과제를 해결하기 위하여, 본 발명은 하기와 같은 전신경화증의 예방 또는 치료용 약학적 조성물을 제공한다:

상기 화학식 1로 표시되는 화합물 또는 이의 약학적으로 허용 가능한 염은, 한국특허 등록번호 제10-2084772호에 기재된 화합물로서, 구체적으로 해당 명세서의 실시예 40으로 기재된 물질이다.
본 발명에 따른 약학적 조성물은 전신경화증 중에서도, 특히 전신경화증 연관 폐질환(systemic sclerosis associated interstitial lung disease; SSc-ILD)의 예방 또는 치료에 유용하게 사용될 수 있다. 특히, 본 발명에 따른 약학적 조성물은 전신경화증 연관 폐질환의 피부 경화 완화 및 폐기능 개선의 이중 효과를 가진다. 본 발명에서 사용하는 용어 "폐기능 개선"은 실질적인 폐의 기본적인 기능인, 혈액을 체내 공급하는 능력이 개선됨을 의미하며, 이는 폐 섬유화 개선으로부터 당연히 따라오는 효과는 아니며, 폐 섬유화 지표와 별도로 체내 산소포화도를 통하여 확인할 수 있다.
본 발명에서 사용되는 용어 "예방"은 본 발명의 조성물의 투여로 상기 질환의 발생, 확산 및 재발을 억제시키거나 지연시키는 모든 행위를 의미하고, "치료"는 본 발명의 조성물의 투여로 상기 질환의 증세가 호전되거나 이롭게 변경되는 모든 행위를 의미한다.
한편, 상기 화학식 1로 표시되는 화합물은 약학적으로 허용가능한 염의 형태로 사용할 수 있으며, 염으로는 약학적으로 허용 가능한 유리산(free acid)에 의해 형성된 산부가염이 유용하다. 유리산으로는 무기산과 유기산을 사용할 수 있다. 무기산으로는 염산, 브롬산, 황산, 인산 등을 사용할 수 있고, 유기산으로는 구연산, 초산, 젖산, 말레인산, 글루콘산, 메탄술폰산, 숙신산, 4-톨루엔술폰산, 글루탐산, 또는 아스파르트산 등을 사용할 수 있다. 바람직하게는, 상기 화학식 1로 표시되는 화합물의 약학적으로 허용 가능한 염은 염산염이다.
또한, 상기 화학식 1로 표시되는 화합물은 결정 형태 또는 비결정 형태로 제조될 수 있으며, 결정 형태로 제조될 경우, 임의로 수화되거나 용매화될 수 있다. 본 발명에서는 상기 화학식 1로 표시되는 화합물의 화학양론적 수화물뿐만 아니라 다양한 양의 물을 함유하는 화합물이 포함될 수 있다. 상기 화학식 1로 표시되는 화합물의 용매화물은 화학양론적 용매화물 및 비화학양론적 용매화물 모두를 포함한다.
본 발명의 약학적 조성물은 표준 약학적 실시에 따라 경구 또는 비경구 투여 형태로 제형화할 수 있다. 이들 제형은 유효성분 이외에 약학적으로 허용 가능한 담체, 보조제 또는 희석액 등의 첨가물을 함유할 수 있다.
적당한 담체로는 예를 들어, 생리식염수, 폴리에틸렌글리콜, 에탄올, 식물성 오일 및 아이소프로필미리스테이트 등이 있고, 희석액으로는 예를 들어 락토즈, 덱스트로즈, 수크로즈, 만니톨, 솔비톨, 셀룰로즈 및/또는 글리신 등이 있으나, 이들로 한정되는 것은 아니다. 또한, 본 발명의 화합물들은 주사 용액의 제조에 통상적으로 사용되는 오일, 프로필렌글리콜 또는 다른 용매에 용해시킬 수 있다. 또한, 국소 작용을 위해 본 발명의 화합물을 연고나 크림으로 제형화할 수 있다.
본 발명의 화합물들의 약학적 투여 형태는 이들의 약학적으로 허용 가능한 염 또는 용매화물의 형태로도 사용될 수 있고, 또한 단독으로 또는 다른 약학적 활성 화합물들과 결합뿐만 아니라 적당한 집합으로 사용될 수 있다. 일례로, 본 발명에 따른 약학적 조성물은 전신경화증의 예방 또는 치료에 사용되는 다른 유효 성분을 추가로 포함할 수 있다. 이러한 다른 유효 성분의 예로는, 오페브®(닌테다닙, Nintedanib) 또는 악템라®(토실리주맙, tocilizumab)를 들 수 있다. 또한, 본 발명에 따른 약학적 조성물이 다른 유효 성분을 추가로 포함하는 경우, 상기 화학식 1로 표시되는 화합물, 또는 이의 약학적으로 허용 가능한 염과, 다른 유효 성분의 중량비는 1:0.1 내지 1:10이 바람직하다.
본 발명의 화합물들은 일반적인 식염수, 5% 덱스트로스와 같은 수용성 용매 또는 합성 지방산 글리세라이드, 고급 지방산 에스테르 또는 프로필렌글리콜과 같은 비수용성 용매에 화합물을 용해시키거나, 현탁시키거나 또는 유화시켜 주사제로 제형화 될 수 있다. 본 발명의 제형은 용해제, 등장화제(isotonic agents), 현탁화제, 유화제, 안정화제 및 방부제와 같은 통상의 첨가제를 포함할 수 있다.
본 발명에 따른 약학적 조성물을 경구 또는 비경구적 경로를 통해 투여될 수 있다. 투여 방법에 따라, 본 발명에 따른 조성물은 상기 화학식 1로 표시되는 화합물, 또는 이의 약학적으로 허용 가능한 염을 0.001 내지 99 중량%, 바람직하게는 0.01 내지 60 중량% 함유할 수 있다.
본 발명의 약학 조성물은 쥐, 생쥐, 가축 및 인간 등을 비롯한 포유동물에 다양한 경로로 투여될 수 있다. 투여의 모든 방식은 예상될 수 있는데, 예를 들어, 경구, 직장 또는 정맥, 근육, 피하, 자궁내 경막 또는 뇌혈관(intracerebroventricular) 주사에 의해 투여될 수 있다.
상술한 바와 같이 본 발명에 따른 약학적 조성물은 전신경화증의 예방 또는 치료에 유용하게 사용할 수 있다.

실시예
한국특허 등록번호 제10-2084772호의 실시예 40과 동일한 방법으로 하기 화합물을 제조하였고, 이하 '활성 성분' 또는 'Example'로 명명하였다.

실험예 1: 3D skin organoid 모델에서의 피부경화 완화 효과 확인
전신피부경화증 환자의 iPSC 유래 섬유아세포를 이용하여 문헌(Lancet. 2009 Nov 21;374(9703):1745-53)에 기재된 방법에 따라 3D skin organoid를 제작하였고, 활성 성분 1 uM을 DMSO 용액으로 처리하였을 때, 3D skin organoid의 두께가 감소됨을 H&E 면역화학염색을 통해 확인하였다(도 1). 대조군은 DMSO만 처리하였다.
실험예 2: 3D skin organoid 이식 마우스 모델에서의 피부경화 완화 효과 확인
앞서 구축된 전신피부경화증 환자 iPSC 유래 3D skin organoid를 SCID 마우스에 이식하여 전신피부경화증 마우스 모델을 구축하였다. 마우스 피부조직을 1 x 2 cm로 자른 후, 3D skin organoid 피부조직을 마우스 피부에 봉합하였다. 피부이식 방법은 임상에서 사용되는 tie-over dressing 방법을 이용하여 마우스 등에 이식하였다. 봉합부위에 거즈를 올린 뒤, 봉합사로 묶고 밴드로 드레싱하였다,
전신피부경화증 skin organoid 이식 마우스 모델에 활성 성분 3 mg/kg, 10 mg/kg를 2주간 매일 피하투여를 진행하였다. 음성대조군(NC)은 정상피부조직에 생리식염수를, 양성대조군(SSc)은 전신피부경화증 skin organoid 이식 피부조직에 생리식염수를 투여하였다. 전신피부경화증의 1차 평가지표인 피부 두께를 면역화학염색을 통해 측정하여 그 결과를 도 2에 나타내었다. 도 2에 나타난 바와 같이, 활성 성분에 의해 피부 조직의 두께가 유의적으로 감소하는 것을 확인하였다 (***p<0.001).
활성 성분에 의해 전신피부경화증 피부조직에서 섬유화 인자가 감소하는지 여부를 확인하기 위하여 면역화학염색을 수행하고 그 결과를 도 3에 나타내었다. 도 3에 나타난 바와 같이, 활성 성분에 의해 섬유화인자 Collagen I, Collagen III, αSMA가 유의적으로 감소하는 것을 확인하였다 (*p<0.05, ***p<0.001).
실험예 3: 전신경화증 연관 폐질환(SSc-ILD) 마우스 모델에서 치료 효과 확인
C57BL/6 마우스 7 주령을 입수하여 5일 이상 실내에서 순화시켰다. 전신경화증 연관 폐질환 모델은 osmotic pump를 주입한 모델을 design 하였다. 그 이유로, 만일 osmotic pump를 사용한 모델링이 아니면, 매일 시험 동물의 등에 BLM을 피하 주사로 4주간 주입해야 하는 모델이며, 이러한 시험은 투여 부위에 매우 극심한 통증을 줄 수 있기 때문이다.
도 4에 나타난 바와 같이, osmotic pump를 이용한 약물의 투여 속도는 0.5 ul/hr이었고(7 days), osmotic pump의 크기는 1.5 cm이었다. 절개 부위를 봉합하고, osmotic pump 삽입한 다음, 1주일간 bleomycin 투약 후, 10일째 osmotic pump를 제거하였다. 1주일간 BLM 투여 마우스 모델에서 전신경화증 연관 폐질환이 유발되었음을 확인하였으며, 구체적인 실험군은 하기 표 1과 같다.

실험예 3-1: Body weight (%)
활성 성분 투여 28 days 결과를 기반으로(표 2 및 도 5), 몸무게의 변화는 활성 성분 3 mg/kg에서부터 유의한 결과를 나타내었으며, 30 mg/kg에서는 p<0.01로 vehicle 대비 몸무게가 유의하게 증가된 것을 확인하였다.

실험예 3-2: Lung weight
Lung weight의 변화는 30 mg/kg에서만 유의한 경향성을 보였으나, body weight로 보정 시 10 mg/kg에서부터 유의성이 나타났으며 30 mg/kg에서는 p<0.01 유의성이 높아짐을 확인하였다(표 3 및 도 6).

실험예 3-3: 폐기능(Lung Function) 평가(SpO
2

본 폐기능 평가는 전신경화증 연관 폐질환 환자에서 본 활성물질의 치료효과를 보여주기 위한 실험으로서, 일반적인 폐섬유화 개선 효과와는 달리, 실질적으로 전신경화증 연관 폐질환 환자의 증상과 삶의 질을 좌우하는 가장 큰 영향을 끼치는 가장 직접적이고 중요한 평가 지표이다. 이는 체내 산소 농도를 측정함으로써 전신경화증 연관 폐질환 동물 모델에서 폐의 기능 개선 효과를 가장 직접적으로 보여줄 수 있는 실험이다.
28일째 마우스의 복부 쪽을 통해 SpO2 측정 장치(Berry, Veterinary Pulse Oximeter)로 측정하였다. 그 결과를 도 7에 나타내었다.
도 7에 나타난 바와 같이, 활성 성분 10 mg/kg에서부터 유의한 결과를 나타냈으며, 활성 성분 30 mg/kg에서는 p<0.001로 vehicle 대비 산소포화도가 높아진 것으로 나타났다. 또한, 활성 성분 30 mg/kg 투여군에서는 비교군인 닌테다닙 60 mg/kg 투여군과 동등한 정도의 산소포화도 회복률을 나타냄을 확인하였다.
실험예 3-4: 피부 두께(Skin thickness) 평가
28일째 마우스에서 피부 조직을 추출하여 H&E 및 Mansson’s Trichrome (MT stain)을 진행하였다. Stain이 진행된 사진을 imagej를 통해 random하게 10 군데의 skin thickness를 측정하였다(최소 200~300 um 간격을 두고 진행). 최대한 사진의 전체 부위를 고르게 측정하였고, 측정시 stain 조직이 손상이 되어있는 부위는 배제하였다. 그 결과를 도 8 및 표 4에 나타내었다.
도 8에 나타난 바와 같이, BLM 군에서는 skin의 dermis외에 Adipose가 있어야 할 부분에도 Collage이 가득 매워지며 잘 유도된 것으로 확인되었다. 활성 성분 10 mg/kg부터 유의한 결과를 보이는 것으로 확인되었으며, 활성 성분 30 mg/kg에서는 P<0.0001 이하로 매우 유의한 결과를 나타내었다. 또한, 전체적으로 활성 성분을 투여하였을 때 MT stain 염색 정도가 낮은 것을 통하여 피부 내 콜라겐 밀도가 감소하였음을 확인하였다. 특히, 활성 성분 3 mg/kg에서 skin thickness는 크게 감소하지 않았으나 skin의 collagen의 density가 감소하는 경향성을 보이는 것이 확인되었다. 특히, 활성 성분 10 mg/kg에서 닌테다닙 60 mg/kg보다 우수한 skin thickness 개선 효과가 확인되었다.

실험예 3-5: 체내 콜라겐 함량 측정
마우스 체내 hydroxyproline 양을 측정하여, 콜라겐 함량을 간접적으로 측정하고자 하였다. INSOLUBLE Collagen Assay(Biocolor, S2000)를 이용하여 분석하였으며, 구체적으로 하기와 같았다.
먼저, 냉동된 피부 조직 10 mg을 100 ul Fragmentation Reagent와 함께 분쇄하였다. 37% HCl 100 ul를 첨가하고 65℃에서 3시간 incubation 하였다. 30분 간격으로 조직 붕해를 돕기 위해 튜브 내용물을 흔들어 주었다. 12,000 rpm으로 10분간 원심 분리하였다. 원심분리 후 1.5 ml tube로 옮겼다. 10~100 ul 사이의 sample에서 50 ul을 취하고 증류수를 첨가하여 100 ul에 맞춰서 sample을 준비하였다.
상기 준비된 sample 100 ul에 1 ml dye를 넣어 30분 동안 혼합하였다. 12,000 rpm으로 10분간 원심 분리한 후 새로운 tube로 옮겼다. 750 ul의 ice-cold Acid-Salt Wash Reagent로 Dye를 제거하였다. 12,000 rpm으로 10분간 원심 분리한 후 새로운 tube로 옮겼다. 튜브를 vortex mixer를 사용하여 콜라겐 결합 염료를 용액과 섞고, 콜라겐 결합 염료를 10분 내에 용해시켜, 측정 준비를 완료하였다(2 ~ 3 시간 내에 측정을 완료하여야 한다). 흡광도를 측정할 준비가 될 때까지 튜브 뚜껑을 닫아두었다.
각 시료의 200 ul를 96 micro-well plate의 개별 well로 옮기고, 560 nm에서 흡광도를 측정하였다. Hydroxyproline 값을 normal 군에 대비하여 ratio 값으로 결과를 표시하였으며, 그 결과를 표 5 및 도 9에 나타내었다.
BLM 군을 확인한 결과 Saline 군에 비해 skin의 Hydroxyproline이 2배 이상 증가된 것을 확인하여 모델링이 잘 이루어진 것을 확인하였다. 또한, 활성 성분 10 mg/kg부터 유의한 결과를 나타내었으며, 활성 성분 30 mg/kg에서는 P<0.001 이하로 매우 유의한 결과를 나타내었다.

상기 실험 결과를 통하여 확인할 수 있는 바와 같이, 본 발명의 활성성분은 전신경화증 치료 효과와 전신경화증 연관 폐질환의 폐기능 저해 개선이라는 전혀 상이한 두가지 효과를 나타내었다. 이러한 이중 효과는 어느 한가지에 대한 치료 효과를 통하여 예측할 수 없는 부분이며, 본 발명 활성물질의 특징으로 파악된다.
