Abstract
본 발명은 울금으로부터 추출되는 울금추출물을 유효성분으로 함유하여 간기능 개선 효능을 갖도록 이루어진 울금과 용암해수를 이용한 간기능 개선용 울금추출분말소재 및 이를 이용한 간기능 개선용 조성물에 관한 것이다.
본 발명인 울금과 용암해수를 이용한 간기능 개선용 울금추출분말소재 및 이를 이용한 간기능 개선용 조성물은 울금을 알코올과 용암해수를 이용하여 추출하되, 건조 울금 1㎏ 중량대비, 50% 에탄올(주정) 92.5중량%와 용암해수 7.5중량%로 이루어진 혼합(추출)용매 8L를 이용하여 60℃에서 7시간 추출하고, 보조첨가물을 첨가한 후 분무건조시켜 분말화시킨다.

본 발명은 울금으로부터 추출되는 울금추출물을 유효성분으로 함유하여 간기능 개선 효능을 갖도록 이루어진 울금과 용암해수를 이용한 간기능 개선용 울금추출분말소재 및 이를 이용한 간기능 개선용 조성물에 관한 것이다.
만성 간질환은 국내에서 성인병 및 사망의 중요한 원인으로 부각되고 있으며, 간은 완충능력이 큰 생체기관으로 질환의 초기 단계에 자각증상이 없어 질병이 상당히 진전된 상태에 발견되는 경우가 많을 뿐 아니라 효과적인 치료제 등이 부적한 실정이다.
질환의 원인으로는 알코올, 약물, 화학약품, 바이러스성 간염, 담도질환과 같은 대사성 질환과 자가면역 질환 등이 있으며, 또한, 간은 인체에서 혈액의 저장과 순환, 혈액량 조절과 해독작용의 중추로서 최근 산업화에 따라 배출량이 늘어나는 유해물질에 노출되는 기회가 증가하면서 끊임없는 해독작용에 시달리고 있다.
알코올성 간질환은 단순 지방간염부터 심각한 질환인 섬유화, 경변증 및 간암에 이르는 폭넓은 범위를 보이는 일련의 질환군이며, 전 세계적으로 만성간질환의 가장 흔한 형태로 높은 유병률과 사망률을 나타내는 질환이다.
알코올성 지방간은 만성 음주의 가장 흔한 형태로 나타나는 질환으로 간세포 내 중성지방의 비정상적인 축적을 특징으로 하며, 간세포내 지방 축적의 주요 기전은 신생지질생성의 주요 조절인자인 sterol regulatory element-binding protein 1c(SREBP1c)의 유전자 발현이 증가하고 이와 반대로 유리 지방산(free fatty acid)의 합성과 수송 조절에 중요한 역할을 하는 peroxisome proliferator-activated receptor α(PPARα)의 유전자 발현감소를 통해 지방 대사 불균형에서 기인한다.
에탄올은 직접적으로 SREBP1c 유전자 전사를 증가시키고, 간세포 내에서 PPARα 활성을 억제하고, 간접적으로는 세포질 세망(Endoplasmic reticulum, ER)의 스트레스 반응과 싸이토크롬 효소(cytochrome P450 2E1, CYP2E1) 발현 증가에 의해 유래되는 산화적인 스트레스와 엔도카나비노이드 신호전달 기전을 통해 지방 대사의 불균형을 초래한다.
본 출원인은 울금 및 용암해수 활용을 높여 지역경제를 이바지하고자 하는 목적과, 간 기능 개선을 위하여 아래와 같은 선행특허기술를 제공하였다.
한국공개특허 10-2014-0084502호인 알로에와 울금을 이용한 혼합발효물을 유효성분으로 하는 간 기능 개선용 조성물과 그 제조방법 및 이를 이용한 간 기능 개선용 제제, 한국등록특허 10-1381850호인 울금 추출물을 유효성분으로 하는 항 콜레스테롤용 조성물과 그 울금 추출물의 추출방법, 한국등록특허 10-1577002호인 울금과 용암해수를 이용한 항콜레스테롤 또는 간기능 개선용 조성물과 기능성 식품.
본 발명의 출원인은 다양한 생약제를 이용한 소재탐색을 통한 울금과 제주지역내의 용암해수를 활용하여 간 기능 개선에 효능을 가지는 간기능 개선용 울금추출분말소재 개발 및 이를 이용하는 간기능 개선용 조성물를 제공하고자 한다.
또한, 본 발명은 출원인이 제공한 선행기술을 보완하고자 한다.
또한, 알코올성 지방간은 만성 음주의 가장 흔한 형태로 나타나는 질환으로 간세포 내 중성지방의 비정상적인 축적을 특징으로 현재, 알코올성 손상으로부터 간 보호에 도움을 주는 기능성원료가 국한되어 있으므로, 안전한 국내 소재로 가격 및 일일섭취량에서 경쟁력을 갖는 알코올성 간 손상 개선 소재를 개발하고자 한다.
이와 같은 목적을 달성하고자,
본 발명인 울금과 용암해수를 이용한 간기능 개선용 울금추출분말소재는 울금을 알코올과 용암해수를 이용하여 추출하되, 건조 울금 1㎏ 중량대비, 50% 에탄올(주정) 92.5중량%와 용암해수 7.5중량%로 이루어진 혼합(추출)용매 8L를 이용하여 60℃에서 7시간 추출하고, 보조첨가물을 첨가한 후 분무건조시켜 분말화시킴을 특징으로 한다.
상기 혼합(추출)용매를 이용하여 추출한 추출물은 여과과정과 농축과정를 거친 후 보조첨가물을 첨가한 후 분무건조시켜 분말화시키되, 상기 보조첨가물 고형분 함량 기준으로 카세인나트륨 20중량%와 덱스티린 20중량%이다.
다른 발명인 울금과 용암해수를 이용한 간기능 개선용 조성물은, 울금을 알코올과 용암해수를 이용하여 추출하되, 건조 울금 1㎏ 중량대비, 50% 에탄올(주정) 92.5중량%와 용암해수 7.5중량%로 이루어진 혼합(추출)용매 8L를 이용하여 60℃에서 7시간 추출하고, 보조첨가물을 첨가한 후 분무건조시켜 분말화시킨 울금추출분말을 포함함을 특징으로 한다.
상기 혼합(추출)용매를 이용하여 추출한 추출물은 여과과정과 농축과정를 거친 후 보조첨가물을 첨가한 후 분무건조시켜 분말화시키되, 상기 보조첨가물 고형분 함량 기준으로 카세인나트륨 20중량%와 덱스티린 20중량% 임을 특징으로 한다.
이와 같이 이루어진 본 발명인 울금과 용암해수를 이용한 간기능 개선용 울금추출분말소재 및 이를 이용하는 간기능 개선용 조성물은 간기능 개선용 울금추출분말소재가 분무건조로 분말화되어 이용되어 이용이 용이한 장점이 있다.
또한, 첨가물인 보조첨가물은 오일성분에 의한 분말화가 잘 이루어지지 않는 것을 해소할 수 있는 한편, 분말화 이후 수분 흡수를 방지하여 보관에 대한 안정성을 높일 수 있는 효과를 가진다.
이와 같이 이루어진 본 발명인 울금과 용암해수를 이용한 간기능 개선용 울금추출분말소재는 실험을 통하여 안정성이 있음을 확인하였으며, 간기능 개선에 효과가 있음을 확인하였음으로, 알코올에 손상된 간 개선에 널리 이용될 수 있으며, 이로 인하여 지역경제 발전에 이바지 할 수 있을 것으로 예상된다.

본 명세서 및 청구범위에 사용된 용어나 단어는 통상적이거나 사전적인 의미로 한정해서 해석되어서는 아니 될 것이며, 본 명세서에 기재된 실시 예와 도면에 도시된 구성은 본 발명의 가장 바람직한 일 실시 예에 불과할 뿐이고 본 발명의 기술적 사상을 모두 대변하는 것은 아니므로, 본 출원시점에 있어서 이들을 대체할 수 있는 다양한 균등물과 변형 예들이 있을 수 있음을 이해하여야 한다.
본 발명은 용암해수가 포함된 추출용매를 사용하여 울금을 추출하고, 추출된 울금추출물을 유효성분으로 함유하는 울금과 용암해수를 이용한 간기능 개선용 울금추출분말소재 및 이를 이용한 간기능 개선용 조성물에 관한 것이다.
본 발명인 울금과 용암해수를 이용한 간기능 개선용 울금추출분말소재는 울금을 알코올과 용암해수를 이용하여 추출하고, 추출한 추출물은 여과과정과 농축과정를 거친 후 보조첨가물을 첨가한 후 분무건조방식으로 분말화한다.
상기 혼합(추출)용매는 50% 에탄올(주정) 92.5중량%와 용암해수 7.5중량%로 이루어지며, 건조 울금 1㎏ 중량대비 혼합(추출)용매는 8L를 이용한다.
상기 용암해수를 이용한 울금의 추출과정은 추출용기에 건조 울금 1㎏ 중량대비 혼합(추출)용매는 8L를 넣고 60℃에서 7시간 추출한다.
상기 보조첨가물은 분무건조과정 전에 첨가하는 것으로 고형분 함량 기준으로 카세인나트륨 20중량%와 덱스티린 20중량%이 첨가된다.
상기 보조첨가물은 용암해수를 이용한 울금의 추출과정으로 추출되고 농축과정을 통하여 농축된 울금추출농축액에는 오일성분이 많이 함유되어 있어 분말화 가 잘 이루워지지 않는 단점이 있다. 이러한 단점을 해결하고자 분무건조 공정전에 보조첨가물인 카제인나트륨과 덱스트린 첨가한다. 상기 첨가된 보조첨가물은 분말화 이후에 수분 흡수가 잘 되지 않도록 하여 보관 안정성을 높이는 효과를 발휘한다.
상기 용암해수는 용암해수 원수를 직접 사용할 수 있으며, 용암해수를 처리하여 획득되는 ED미네랄수, RO염수, RO탈염수가 사용될 수 있다.
즉, 상기 용암해수로는 용암해수 원수, ED미네랄수, RO염수, RO탈염수 중 선택되는 하나가 사용될 수 있으며, 바람직하게는 ED미네랄수를 사용하는 것이 바람직하다.
상기 용암해수는, 「바닷물이 현무암층에 의해 여과돼 육지 지하로 흘러든 물」을 칭하며, 우리나라 제주도에서는 제주도 동부해안지역과 서부해안지역에 분포하고 있으며, 일일/계절변동 및 물리적 특성 변화가 적고, 수온은 16~18℃로 변함이 없으며, pH(수소이온농도)는 평균 75범위로서 해양심층수(78) 및 일반해수(80~86) 비교했을 때 안정한 특징을 가지고 있다. 또한 대장균, 암모니아성 질소, 페놀류, 수은, 카드뮴 등 유해성분이 검출되지 않아 산업화 적용에 장애요인이 없으며, 실험동물모델로 사망률에 미치는 급성독성이 없고, 간, 신장, 심장, 근육 등의 손상과 관련 있는 혈액지표 분석결과 조직이나 장기에 미치는 독성이 없다.
또한 일반해수, 해양심층수, 용암해수 원수는 해수를 기원으로 하기 때문에 주요미네랄(Mg, Ca 등) 성분은 유사하고 암반층에 의해 육지 지하로 스며들면서 인체에 유용한 미량 미네랄을 함유하고 있는 것을 특징으로 한다.
상기 ED미네랄수는, 용암해수를 전기투석을 거친 수(물)로 이온교환막(cation 교환막과 anion 교환막)을 사용하여 용암해수 중 염소와 나트륨을 제거하고 마그네슘과 칼슘 성분 등 몸에 이로운 이온은 그대로 유지시키는 수(물)을 칭하며, 용암해수는 ED 미네랄수와 염수로 분리된다.
상기 RO염수 및 RO 탈염수는, 용암해수를 역삼투막을 사용하여 획득한 것으로, 일반적으로 해수는 역삼투막의 방법에 의하여 담수와 염수로 분리된다. 즉, 용암해수를 역삼투막의 방법으로 처리하여 염수인 RO염수와 담수인 RO탈염수(물)를 획득한다.
상기와 같이 이루어진 울금과 용암해수를 이용한 간기능 개선용 울금추출분말소재는 간기능 개선용 조성물의 주요성분으로 사용될 수 있다.
본 발명의 울금과 용암해수를 이용한 간기능 개선용 울금추출분말소재를 유효성분으로 하는 간기능 개선용 조성물은, 투여를 위해서 상기 기재한 유효성분 이외에 추가로 약학적으로 허용가능 한 담체를 1종 이상 포함하여 제조할 수도 있다.
상기 약학적으로 허용 가능한 담체로는, 식염수, 멸균수, 링거액, 완충 식염수, 덱스트로오스 용액, 말토덱스트린 용액, 글리세롤, 에탄올 등이 있으며, 이들 성분 중 1 성분 이상을 혼합하여 사용할 수 있으며, 필요에 따라 항산화제, 완충액, 정균제 등 다른 통상의 첨가제를 첨가할 수도 있다.
또한, 희석제, 분산제, 계면활성제, 결합제 및 윤활제를 부가적으로 첨가하여 수용액, 현탁액, 유탁액 등과 같은 주사용 제형, 환약, 캡슐, 과립 또는 정제로 제제화할 수 있다.
[실시 예]
- 용암해수 울금추출분말 획득.
건조 울금 1㎏에 혼합(추출)용매 8L를 넣고 60℃에서 7시간 추출한 후 여과 및 농축을 진행하여 용암해수 울금추출물을 획득한다.
상기 혼합(추출)용매는 50% 에탄올(주정) 92.5중량%와 ED미네랄수 7.5중량%를 혼합한 혼합물을 사용하였다.
획득한 상기 용암해수 울금추출물에 고형분 함량 기준으로 카세인나트륨(Sodium Caseinate) 20중량%와 덱스티린(dextrin) 20중량%를 첨가한 후 분무건조과정을 통하여 용암해수 울금추출분말 250g을 획득하였다.
상기 용암해수는 제주도 해역내에서 획득되는 제주용암해수를 가공한 ED미네랄수를 사용하였다.

2. 실험방법
1) 알코올 유도 간 손상 동물모델 제작
알코올은 5 g/kg body weight로 1주일 동안 매일 1회 경구투여 하여 유도하였다.
2) 혈액 및 간 균질액의 조제
알코올 마지막 처리 이후부터 24시간 절식시킨 후 대동맥으로부터 채혈하고, 채취한 혈액은 실온에 30분간 방치한 후 3,000 rpm에서 10분간 원심분리 하였다. 간 균질액은 간 조직 무게(g)에 10배의 0.1 M Tris HCl buffer(pH 7.4)를 가하고 유리-테프론분쇄기로 균질화하여 준비하고, 그 단백질량은 Bradford 방법으로 정량하여 항산화 효소 활성 측정에 이용하였다.
3) 혈청 중 간 기능 지표효소 활성 측정
Alanine amintransferase(ALT), aspartate aminotransferase(AST) 및 alkaline phosphatase(ALP) 활성도는 각 기질과 효소반응을 이용한 비색법에 의해 제조된 assay kit로 측정하였다. 총 콜레스테롤, HDL-콜레스테롤, LDL-콜레스테롤과 중성지방은 효소법을 이용한 자동 분석기를 사용하였고, FFA는 ACS-ACOD 효소 방법을 이용하였며 인슐린은 Radioimmunoassay(RIA) kits를 이용하여 측정하였다.
4) 항산화 효소 활성 측정
간조직 중 catalase(CAT) 활성은 20 mM 과산화수소를 기질로 균질액 내의 CAT에 의해 감소되는 과산화수소량을 측정하는 방법, superoxide dismutase(SOD) 활성은 xanthin과 xanthine oxidase의 반응에서 형성된 superoxide anion radical이 tetrazolium blue와 formazan을 형성하는 원리를 이용한 방법, glutathione-S-transferase(GST)의 활성은 reduced gluathione이 1-chloro-2,4-dinitrobenzene (CDNB)과 반응하는 원리를 이용한 방법으로 측정하였다.
5) GST colorimetric activity assay
간조직 100mg을 GST assay buffer 0.5ml로 homogenize하여 10,000g, 4℃에서 15분동안 원심분리하여 그 상등액을 본 실험에 사용하였다.
6) Superoxide Dismutase (SOD) activity assay
간조직을 150 mM KCl 관류시켜 적혈구를 제거하였다. 0.1M Tris/HCl, 0.5% Triton X-100, 5 mM β-ME, 0.1 mg/ml PMSF로 homogenize하여 14,000g, 4℃에서 5분동안 원심분리하여 그 상등액을 본 실험에 사용하였다.
위와 같이 처리하고 37℃에서 20분간 배양한 후 450nm의 파장에서 흡광도를 측정한다.
7) CYP2E1, CYP1A2, CYP2B1 및 CYP1A1의 효소활성 측정
CYP2E1 효소 활성은 colorimetric method로 특이적 기질인 p-nitrophenol(PNP)를 이용하여 간 micrisome 획분에서 측정하였다.
CYP1A2, CYP2B1, CYP1A1는 각각 7-ethoxyresorufin-O-deethylase(EROD), 7-pentoxyresorufin-O-dealkylase(PROD), 7-methoxyresorufin-O-deethylase(MROD) 활성측정으로 Pohl과 Fouts의 방법에 의해 간 microsome 획분을 이용하여 측정하였다.
8) 혈중 에탄올 및 아세트알데히드 농도 측정
혈중 에탄올과 아세트알데히드의 농도를 측정하기 위하여 에탄올 투여 전 및 투여 후 1, 3, 5, 7시간 후에 안와채혈을 통해 혈액을 채취하여 1,900×g에서 20분간 원심 분리하여 혈청만을 분리하였고, 알코올 측정 kit와 아세트알데히드 kit를 사용하여 NADH량의 변화를 340nm에서 흡광도로 측정하였다.
9) 간 조직에서의 ADH, ALDH 활성 측정
간 조직을 분리하여 분리된 간 조직은 액체질소에 급속 냉동시킨 후, 시험 당일 50mg씩 나누어 ADH activity kit와 ALDH activity kit에 포함되어 있는 각각의 assay buffer 200μL에 넣어 균질화하였고, 450nm에서 각각의 활성을 측정하였다.
10) 간 조직 유전자 발현 분석
각 실험동물로부터 적출한 간 조직에서의 유전자 발현 양상을 real-time PCR 증폭법을 사용하여 관찰하였다. 간 조직(0.05g)을 RNAzolB(Tel-Test)용액으로 Total RNA를 추출한 뒤 cDNA 합성 및 real-time PCR 분석을 하였다.
Rat Probe & Oligonucleotire primers for real time PCR amplification
유전자 발현은 TaqMan probe(FAM dye-labeled, ABi, USA)를, internal standard를Mouse GAPDH probe set; Endogenous Control(VIC®/ MGB Probe, Probe limited) from Applied Biosystems(4352338E)을 사용하였고, primer의 최종 농도가 200 nM이 되게 반응시켰다.
Real time quantitative PCR의 조건은: pre-denaturation은 2 min at 50 ℃, 10 min 94 ℃, 그리고 40 cycles을 0.15 min at 95 ℃, 1 min at 45 ℃에서 수행하였다.
11) Hepatic Hostological Examinations, Oil Red O staining
포르말린 용액에 의하여 고정된 간 조직은 기본 실험 방법 및 조직 절편으로 만든 후 파라핀으로 포매하고, hematoxylin과 eosin으로 염색하고 형태학적 변화의 차이를 관찰하였다. 중성지방 생성량을 확인하기 위해 intracytoplasmic lipids를 측정하는데 적합한 Oil Red O를 활용하였으며, 염색하여 생성량의 변화의 차이를 관찰하였다.
12) 급성 알코올 투여시 울금추출물 시료에 의한 혈중 알코올농도 변화(숙취분석); 혈중 에탄올 및 아세트알데히드 농도 측정
혈중 에탄올과 아세트알데히드의 농도 를 측정하기 위하여 8주령 SD 흰쥐에 3일간 울금추출물 시료(200mg/kg)와 상쾌한 아침 시료(200mg/kg)를 경구투여하였다. 3일째 울금추출물 시료와 상쾌한 아침 시료를 투여 후 1시간30분 후 Ethanol (2g/kg)을 경구투여 하였다. 에탄올 투여 30분, 60분, 90분, 120분. 240분, 360분에 꼬리끝부분 절단을 통해 혈액(0.5ml)을 채취하고 원심분리하여 혈청만을 분리하였다. 알코올 측정 kit와 아세트알데히드 kit를 사용하여 NADH량의 변화를 340nm 에서 흡광도로 측정하였다.
4. 실험결과
1) 알코올 유도 만성 간 손상 동물모델 제작 및 유효성 평가
① 체중 변화에 미치는 영향
알코올성 간 손상 개선 효능을 갖는 50% 에탄올 울금추출물 및 용암해수 혼합후보소재에 대하여 알코올성 간 손상 동물모델에서의 유효성 평가를 진행하여 효능을 검증을 실시하였다.
알코올은 5g/kg EtOH(50% with saline)농도로 투여한 대조군으로 하였고, 5주간 양성대조군으로 UDCA와 울금-용암해수 50% 에탄올(주정)추출물(C&JM_50E) 400㎎/㎏, 200㎎/㎏, 100㎎/㎏ 투여하였다. 1주 간격으로 체중변화를 측정한 결과, 정상군에 비하여 알코올 투여 대조군(EtOH_CTL)이 2주 이후 체중감소가 나타났고, UDCA와 울금-용암해수 50% 에탄올(주정)추출물(C&JM_50E) 농도에 따른 체중의 변화는 나타나지 않았다(도 3 참조).
② 간 무게 변화에 미치는 영향
알코올성 간 손상 개선 효능을 갖는 50% 에탄올 울금추출물 및 용암해수 혼합후보소재에 대한 간손상 보호효능을 관찰하고자 5주간 알코올 및 UDCA, C&JM_50E를 투여한 후 부검하여 간 무게를 측정하였다.
도 4에서와 같이 정상군 (SD rat_Nr)에 비하여 에탄올 대조군 (EtOH_CTL)이 통계학적 유의성 있게 감 무게가 증가하였고, UDCA와 C&JM_50E(400mg/kg, 200mg/kg, 100mg/kg,) 투여군에서 에탄올 대조군(EtOH_CTL)에 비하여 유의성 있게 감소를 나타내었다.
이러한 결과는 만성알콜성 투여는 간손상에 의한 염증증가로 간무게가 증가하고, UDCA와 C&JM_50E 투여에 의한 항산화 작용으로 간보호효과가 있음을 알 수 있다.
③ 혈청중 지질생화학적(lipid profile)분석
㉠ 총콜레스테롤과 중성지방 측정 변화
혈청중 지질생화학적 분석은 UDCA 및 C&JM_50E복합물를 5주간 투여 후 16시간 식이를 제거하여 공복상태에서 채혈하여 혈청 중 지질생화학검사를 측정하였다(도 5 참조).
도 5의 위표는 혈청 중 총콜레스테롤(total cholesterol) 수준을 분석한 결과로 정상군(SD rat_Nr)에 비하여 에탄올 대조군(EtOH_CTL)의 총콜레스테롤 수준이 22.5% 이상 유의성 있게 증가를 나타내었고, 실험군에서는 UDCA와 C&JM_50E복합물 100 mg/kg에서 통계학적 유의성 있게 감소하였다. 그러나 C&JM_50E복합물 400mg/kg과 200mg/kg 투여군은 에탄올 대조군 (EtOH_CTL)에 비하여 총콜레스테롤 수준이 감소를 나타냈지만, 통계학적 유의성은 없었다.
도 5의 아래표는 혈청 중 중성지방(triglyceride) 수준을 분석한 결과로 정상군(SD rat_Nr)에 비하여 에탄올 대조군(EtOH_CTL)의 중성지방(triglyceride) 수준이 2배이상 유의성 있게 증가를 나타내었고, 실험군에서는 C&JM_50E복합물 400 mg/kg, 200 mg/kg, 100 mg/kg, 50 mg/kg에서 통계학적 유의성있게 2배이상 감소하였다. 그러나 UDCA 투여군은 에탄올 대조군(EtOH_CTL)에 비하여 중성지방(triglyceride) 수준이 감소를 나타냈지만 통계학적 유의성은 없었다.
㉡ 저밀도 콜레스테롤 (LDL cholesterol, LDL-c)과 고밀도 콜레스테롤 (HDL cholesterol, HDL-c) 수준 측정 변화
혈청중 지질생화학적 분석은 UDCA 및 C&JM_50E복합물를 5주간 투여 후 16시간 식이를 제거하여 공복상태에서 채혈하여 혈청 중 지질생화학검사를 측정하였다(도 6 참조).
도 6의 위표는 혈청 중 저밀도 콜레스테롤(LDL-c) 수준을 분석한 결과로 정상군 (SD rat_Nr)에 비하여 에탄올 대조군 (EtOH_CTL)의 저밀도 콜레스테롤(LDL-c)이 79.2% 이상 유의성 있게 증가를 나타내었고, 실험군에서는 UDCA와 C&JM_50E복합물 100mg/kg에서 통계학적 유의성있게 저밀도 콜레스테롤(LDL-c)이 감소하였다. 그러나 C&JM_50E복합물 50 mg/kg 투여군은 에탄올 대조군(EtOH_CTL)에 비하여 저밀도 콜레스테롤(LDL-c) 수준이 감소를 나타냈지만, 통계학적 유의성은 없었다.
도 6의 아래표는 혈청 중 고밀도 콜레스테롤(HDL-c) 수준을 분석한 결과로 정상군(SD rat_Nr)에 비하여 에탄올 대조군(EtOH_CTL)의 고밀도 콜레스테롤(HDL-c) 수준이 19.7% 이상 유의성 있게 증가를 나타내었고, 실험군에서는 UDCA와 C&JM_50E복합물 모든 투여 농도에서 에탄올 대조군(EtOH_CTL)과 차이가 나타나지 않았다.
㉢ 젖산탈수소효소(LDH)과 총단백질(Total protein, T-P) 수준 측정 변화
혈청중 지질생화학적 분석은 UDCA 및 C&JM_50E복합물를 5주간 투여 후 16시간 식이를 제거하여 공복상태에서 채혈하여 혈청 중 조직손상의 지표인 젖산탈수소효소(LDH)와 총단백질(T-P) 수준를 측정하였다(도 7 참조).
도 7의 위표는 혈청 중 젖산탈수소효소(LDH) 수준을 분석한 결과로 정상군(SD rat_Nr)에 비하여 에탄올 대조군(EtOH_CTL)의 젖산탈수소효소(LDH)이 8.8배 이상 유의성 있게 증가를 나타내었고, 실험군에서는 UDCA와 C&JM_50E복합물 (400, 200, 100, 50 mg/kg)에서 통계학적 유의성있게 젖산탈수소효소(LDH)이 감소하였다.
도 7의 아래표는 혈청 중 총단백질 (T-P) 수준을 분석한 결과로 정상군(SD rat_Nr)에 비하여 에탄올 대조군(EtOH_CTL)의 총단백질이 10.2% 이상 유의성 있게 증가를 나타내었고, 실험군에서는 UDCA와 C&JM_50E복합물 (400, 200, 100, 50 mg/kg)에서 통계학적 유의성 있게 총단백질(T-P)이 감소하였다.
㉣ 알칼리인산분해효소(ALP)과 알부민(Albumin) 수준 측정 변화
혈청중 간기능검사 분석은 UDCA 및 C&JM_50E복합물를 5주간 투여 후 16시간 식이를 제거하여 공복상태에서 채혈하여 혈청 중 간기능검사를 측정하였다(도 8 참조).
도 8의 위표는 혈청 중 알칼리인산분해효소(ALP) 수준을 분석한 결과로 정상군(SD rat_Nr)에 비하여 에탄올 대조군(EtOH_CTL)의 알칼리인산분해효소(ALP)이 56.9% 이상 유의성 있게 증가를 나타내었고, 실험군에서는 C&JM_50E 복합물 400mg/kg, 200mg/kg, 100mg/kg에서 통계학적 유의성 있게 알칼리인산분해효소(ALP)가 감소하였다. 그러나 UDCA와 C&JM_50E복합물 50mg/kg 투여군은 에탄올 대조군 (EtOH_CTL)에 비하여 알칼리인산분해효소(ALP) 수준이 감소를 나타냈지만, 통계학적 유의성은 없었다.
도 8의 아래표는 혈청 중 알부민(Albumin) 수준을 분석한 결과로 정상군(SD rat_Nr)에 비하여 에탄올 대조군(EtOH_CTL)의 알부민(Albumin) 수준이 10.2% 이상 유의성 있게 증가를 나타내었고, 실험군에서는 C&JM_50E복합물 400mg/kg, 200 mg/kg에서 통계학적 유의성 있게 알부민이 감소하였다. 그러나 UDCA와 C&JM_50E복합물 100mg/kg, 50mg/kg 투여군은 에탄올 대조군(EtOH_CTL)에 비하여 알부민(Albumin) 수준이 감소를 나타냈지만, 통계학적 유의성은 없었다.
㉤ 혈청중 간기능 아스파르테이트 아미노전달효소(AST)와 알라닌 아미노전이효소(ALT) 수준 측정 변화
혈청중 간기능검사 분석은 UDCA 및 C&JM_50E 복합물를 5주간 투여 후 16시간 식이를 제거하여 공복상태에서 채혈하여 혈청 중 간기능검사를 측정하였다(도 9 참조).
혈청 중 아미노전달효소(AST) 수준을 분석한 결과로 정상군(SD rat_Nr)에 비하여 에탄올 대조군(EtOH_CTL)의 아미노전달효소(AST)이 46.6% 이상 유의성 있게 증가를 나타내었고, 실험군에서는 UDCA와 C&JM_50E복합물 400mg/kg, 200mg/kg에서 통계학적 유의성 있게 아미노전달효소(AST)가 감소하였다. 그러나 C&JM_50E 복합물 100mg/kg, 50mg/kg 투여군은 에탄올 대조군(EtOH_CTL)에 비하여 아미노전달효소(AST) 수준이 감소를 나타냈지만, 통계학적 유의성은 없었다.
혈청 중 알라닌 아미노전이효소(ALT) 수준을 분석한 결과로 정상군(SD rat_Nr)에 비하여 에탄올 대조군(EtOH_CTL)의 알라닌 아미노전이효소(ALT) 수준이 89.3% 이상 유의성 있게 증가를 나타내었고, 실험군에서는 UDCA와 C&JM_50E 복합물 400mg/kg, 200mg/kg, 100mg/kg, 50mg/kg에서 통계학적 유의성 있게 알라닌 아미노전이효소(ALT)가 감소하였다.
④ 간 조직에서의 ADH와 ALDH 활성
에탄올에 의해 만성 간손상이 유도된 동물모델을 대상으로 UDCA 및 C&JM_50E복합물의 간 손상 보호 효과를 검증하고자, UDCA 및 C&JM_50E복합물이 알코올 분해효소의 활성에 미치는 영향을 확인하기 위하여 부검 후 간 조직과 혈청에서 ADH의 활성을 측정하였다(도 10 참조).
도 10의 왼쪽 표는 간 조직에서 ADH 활성을 측정한 결과로 정상군(SD rat_Nr)에 비하여 에탄올 대조군(EtOH_CTL)의 간 조직에서 ADH 활성이 50.3% 이상 유의성 있게 감소를 나타내었고, 실험군에서는 UDCA와 C&JM_50E 복합물 400mg/kg, 200mg/kg, 100mg/kg에서 통계학적 유의성 있게 간 조직에서 ADH 활성이 증가하였다. 그러나 C&JM_50E 복합물 50mg/kg 투여군은 에탄올 대조군(EtOH_CTL)에 비하여 간 조직에서 ADH 활성이 증가를 나타냈지만, 통계학적 유의성은 없었다.
도 10의 오른쪽 표는 혈청에서 ADH 활성을 측정한 결과로 정상군(SD rat_Nr)에 비하여 에탄올 대조군 (EtOH_CTL)의 간 조직에서 ADH 활성이 14.5% 이상 유의성 있게 감소를 나타내었고, 실험군에서는 UDCA와 C&JM_50E 복합물 400mg/kg, 200mg/kg에서 통계학적 유의성 있게 혈청에서 ADH 활성이 증가하였다. 그러나 C&JM_50E 복합물 100mg/kg, 50mg/kg 투여군은 에탄올 대조군(EtOH_CTL)에 비하여 혈청에서 ADH 활성이 증가를 나타냈지만, 통계학적 유의성은 없었다.
⑤ 간 조직에서의 ALDH 활성
에탄올에 의해 만성 간손상이 유도된 동물모델을 대상으로 UDCA 및 C&JM_50E 복합물의 간 손상 보호 효과를 검증하고자, ALDH 활성은 Bostian 방법을 변형 하여 측정하였으며, NADH의 생성속도를 지표로 사용하였다. UDCA 및 C&JM_50E복합물이 알코올 분해효소의 활성에 미치는 영향을 확인하기 위하여 부검 후 간 조직에서 ALDH의 활성을 측정하였다(도 11 참조).
도 11은 간 조직에서 ALDH 활성을 측정한 결과로 정상군(SD rat_Nr)에 비하여 에탄올 대조군(EtOH_CTL)의 간 조직에서 ALDH 활성이 43.0% 이상 유의성 있게 감소를 나타내었고, 실험군에서는 UDCA와 C&JM_50E 복합물 400mg/kg, 200mg/kg에서 통계학적 유의성 있게 간 조직에서 ALDH 활성이 증가하였다. 그러나 C&JM_50E 복합물 100mg/kg, 50mg/kg 투여군은 에탄올 대조군(EtOH_CTL)에 비하여 간 조직에서 ALDH 활성이 증가를 나타냈지만, 통계학적 유의성은 없었다.
⑥ 간 조직에서 항산화 효소계 활성 측정
㉠ 간 조직과 혈청에서 glutathione peroxidase(GSH-Px) 활성 측정 변화
에탄올에 의해 만성 간손상이 유도된 동물모델을 대상으로 UDCA 및 C&JM_50E복합물의 간 손상 보호 효과를 검증하고자, UDCA 및 C&JM_50E복합물이 알코올 분해효소의 활성에 미치는 영향을 확인하기 위하여 부검 후 간 조직과 혈청에서 GSH-Px 활성을 측정하였다(도 12 참조).
도 12의 왼쪽 표는 간 조직에서 GSH-Px 활성을 측정한 결과로 정상군(SD rat_Nr)에 비하여 에탄올 대조군(EtOH_CTL)의 간 조직에서 GSH-Px 활성이 18.1% 이상 유의성 있게 감소를 나타내었고, 실험군에서는 UDCA와 C&JM_50E 복합물 400mg/kg, 200mg/kg, 100mg/kg에서 통계학적 유의성 있게 간 조직에서 GSH-Px 활성이 증가하였다. 그러나 C&JM_50E 복합물 50mg/kg 투여군은 에탄올 대조군(EtOH_CTL)에 비하여 간 조직에서 GSH-Px 활성이 증가를 나타냈지만, 통계학적 유의성은 없었다.
도 12의 오른쪽 표는 혈청에서 GSH-Px 활성을 측정한 결과로 정상군(SD rat_Nr)에 비하여 에탄올 대조군(EtOH_CTL)의 간 조직에서 GSH-Px 활성이 30.8% 이상 유의성 있게 감소를 나타내었고, 실험군에서는 C&JM_50E 복합물 400mg/kg, 200 mg/kg에서 통계학적 유의성 있게 혈청에서 GSH-Px 활성이 증가하였다. 그러나 UDCA, C&JM_50E 복합물 100mg/kg, 50mg/kg 투여군은 에탄올 대조군(EtOH_CTL)에 비하여 혈청에서 GSH-Px 활성이 증가를 나타냈지만, 통계학적 유의성은 없었다.
㉡ 간 조직과 혈청에서 glutathione S-transferase(GST) 활성 측정 변화
에탄올에 의해 만성 간손상이 유도된 동물모델을 대상으로 UDCA 및 C&JM_50E복합물의 간 손상 보호 효과를 검증하고자, UDCA 및 C&JM_50E 복합물이 알코올 분해효소의 활성에 미치는 영향을 확인하기 위하여 부검 후 간 조직과 혈청에서 GST활성을 측정하였다(도 13 참조).
도 13의 왼쪽 표는 간 조직에서 GST 활성을 측정한 결과로 정상군(SD rat_Nr)에 비하여 에탄올 대조군(EtOH_CTL)의 간 조직에서 GST 활성이 50.6% 이상 유의성 있게 감소를 나타내었고, 실험군에서는 UDCA와 C&JM_50E 복합물 400mg/kg , 200mg/kg에서 통계학적 유의성 있게 간 조직에서 GST 활성이 증가하였다. 그러나 C&JM_50E 복합물 100 mg/kg, 50mg/kg 투여군은 에탄올 대조군(EtOH_CTL)에 비하여 간 조직에서 GST 활성이 차이가 없었다.
도 13의 오른쪽 표는 혈청에서 GST 활성을 측정한 결과로 정상군(SD rat_Nr)에 비하여 에탄올 대조군(EtOH_CTL)의 간 조직에서 GST 활성이 63.8% 이상 유의성 있게 감소를 나타내었고, 실험군에서는 UDCA와 C&JM_50E복합물 400mg/kg, 200mg/kg, 100mg/kg, 50mg/kg에서 통계학적 유의성 있게 혈청에서 GST 활성이 증가하였다.
㉢ 간 조직과 혈청에서 superoxide dismutase(SOD) 활성 측정 변화
에탄올에 의해 만성 간손상이 유도된 동물모델을 대상으로 UDCA 및 C&JM_50E복합물의 간 손상 보호 효과를 검증하고자, UDCA 및 C&JM_50E 복합물이 항산화 효소 활성에 미치는 영향을 확인하기 위하여 부검 후 간 조직과 혈청에서 SOD 활성을 측정하였다(도 14 참조).
도 14의 왼쪽 표는 간 조직에서 SOD 활성을 측정한 결과로 정상군(SD rat_Nr)에 비하여 에탄올 대조군(EtOH_CTL)의 간 조직에서 SOD 활성이 21.4% 이상 유의성 있게 감소를 나타내었고, 실험군에서는 UDCA와 C&JM_50E 복합물 400mg/kg, 200mg/kg, 100mg/kg, 50mg/kg에서 통계학적 유의성 있게 간 조직에서 SOD 활성이 증가하였다.
도 14의 오른쪽 표는 혈청에서 SOD 활성을 측정한 결과로 정상군(SD rat_Nr)에 비하여 에탄올 대조군(EtOH_CTL)의 간 조직에서 SOD 활성이 66.9% 이상 유의성 있게 감소를 나타내었고, 실험군에서는 UDCA와 C&JM_50E 복합물 400mg/kg, 200mg/kg, 100mg/kg, 50mg/kg에서 통계학적 유의성 있게 혈청에서 SOD 활성이 증가하였다.
㉣ 간 조직과 혈청에서 catalase(CAT) 활성 측정 변화
에탄올에 의해 만성 간손상이 유도된 동물모델을 대상으로 UDCA 및 C&JM_50E 복합물의 간 손상 보호 효과를 검증하고자, UDCA 및 C&JM_50E복합물이 항산화 효소 활성에 미치는 영향을 확인하기 위하여 부검 후 간 조직과 혈청에서 CAT활성을 측정하였다(도 15 참조).
도 15의 왼쪽 표는 간 조직에서 CAT 활성을 측정한 결과로 정상군(SD rat_Nr)에 비하여 에탄올 대조군(EtOH_CTL)의 간 조직에서 CAT 활성이 8.5% 이상 유의성 있게 감소를 나타내었고, 실험군에서는 UDCA와 C&JM_50E 복합물 400mg/kg, 200mg/kg, 100mg/kg, 50mg/kg에서 통계학적 유의성 있게 간 조직에서 CAT 활성이 증가하였다.
도 15의 오른쪽 표는 혈청에서 CAT 활성을 측정한 결과로 정상군(SD rat_Nr)에 비하여 에탄올 대조군(EtOH_CTL)의 간 조직에서 CAT 활성이 60.9% 이상 유의성 있게 감소를 나타내었고, 실험군에서는 UDCA와 C&JM_50E 복합물 400mg/kg, 200mg/kg, 100mg/kg, 50mg/kg에서 통계학적 유의성 있게 혈청에서 CAT 활성이 증가하였다.
⑦ 간 조직에서 real-time PCR 유전자 발현 분석
㉠ 간 조직 염증사이토카인 유전자 발현 분석
UDCA 및 C&JM_50E복합물이 간조직 mRNA유전자 발현에 미치는 영향을 확인하기 위하여 부검 후 간 조직을 real-time PCR 분석하였다(도 16 참조).
도 16은 간 조직에서 IL-1β(왼쪽 표), IL-6(중간 표), 그리고 TNF-α (오른쪽 표) mRNA를 분석한 결과로 정상군(SD rat_Nr)에 비하여 에탄올 대조군(EtOH_CTL)의 간 조직에서 IL-1β(왼쪽 표), IL-6(중간 표), 그리고 TNF-α (오오른쪽 표) mRNA 유전자 발현이 유의성 있게 증가를 나타내었고, 실험군에서는 UDCA와 C&JM_50E 복합물 400mg/kg, 200mg/kg에서 통계학적 유의성 있게 간 조직에서 IL-1β, IL-6, 그리고 TNF-α mRNA 유전자 발현이 감소하였다. 그러나 C&JM_50E 복합물 100mg/kg, 50mg/kg 투여군은 에탄올 대조군(EtOH_CTL)에 비하여 간 조직에서 IL-1β, IL-6, 그리고 TNF-α mRNA 유전자 발현이 감소를 나타냈지만, 통계학적 유의성은 없었다.
㉡ 간 조직 염증인자 NOS-II, COX-2 mRNA 유전자 발현 분석
UDCA 및 C&JM_50E복합물이 간조직 NOS-II, COX-2 mRNA유전자 발현에 미치는 영향을 확인하기 위하여 부검 후 간 조직을 real-time PCR 분석하였다(도 17 참조).
도 17은 간 조직에서 NOS-II mRNA(왼쪽 표), 그리고 COX-2 mRNA(오른쪽 표)를 분석한 결과로 정상군(SD rat_Nr)에 비하여 에탄올 대조군(EtOH_CTL)의 간 조직에서 NOS-II mRNA, 그리고 COX-2 mRNA 유전자 발현이 유의성 있게 증가를 나타내었고, 실험군에서는 UDCA와 C&JM_50E 복합물 400mg/kg, 200mg/kg에서 통계학적 유의성 있게 간 조직에서 NOS-II mRNA, 그리고 COX-2 mRNA 유전자 발현이 감소하였다. 그러나 C&JM_50E 복합물 100mg/kg, 50mg/kg 투여군은 에탄올 대조군(EtOH_CTL)에 비하여 간 조직에서 NOS-II mRNA, 그리고 COX-2 mRNA 유전자 발현이 감소를 나타냈지만, 통계학적 유의성은 없었다.
㉢ 간 조직 염증인자 MMP-2, 그리고 MMP-9 mRNA 유전자 발현 분석
UDCA 및 C&JM_50E복합물이 간조직 MMP-2, 그리고 MMP-9 mRNA유전자 발현에 미치는 영향을 확인하기 위하여 부검 후 간 조직을 real-time PCR 분석하였다(도 18 참조).
도 18은 간 조직에서 MMP-2 mRNA(왼쪽 표), 그리고 MMP-9 mRNA(오른쪽 표)를 분석한 결과로 정상군(SD rat_Nr)에 비하여 에탄올 대조군(EtOH_CTL)의 간 조직에서 MMP-2 mRNA, 그리고 MMP-9 mRNA 유전자 발현이 유의성 있게 증가를 나타내었고, 실험군에서는 UDCA와 C&JM_50E 복합물 400mg/kg, 200mg/kg에서 통계학적 유의성 있게 간 조직에서 MMP-2 mRNA, 그리고 MMP-9 mRNA 유전자 발현이 감소하였다. 그러나 C&JM_50E 복합물 100mg/kg, 50mg/kg 투여군은 에탄올 대조군(EtOH_CTL)에 비하여 간 조직에서 MMP-2 mRNA, 그리고 MMP-9 mRNA 유전자 발현이 감소를 나타냈지만, 통계학적 유의성은 없었다.
⑧간조직의 조직학적 분석
UDCA 및 C&JM_50E복합물이 간조직에 치는 영향을 확인하기 위하여 부검 후 간 조직학적 검사를하였다(도 19참조).
알코올성 간질환 초기의 조직학적 변화는 간세포의 지방증(steatosis)으로 작은 지방방울에서 큰 지방방울로 진행되면서 지방간염(steatohepatitis), 알코올성 간질환의 마지막 단계인 간조직의 섬유화에 의한 간경변증(steatofibrosis)으로 진행된다(Lefkowitch et al., 2005). 이러한 알코올성 간질환에서 UDCA 및 C&JM_50E 복합물의 간손상 개선 활성을 직접적으로 확인하고자 간 조직을 염색하였다.
도 19는 간 조직검사를 H&E, Oil red O 염색으로 분석한 결과로 정상군(SD rat_Nr)에 비하여 에탄올 대조군(EtOH_CTL)의 간 조직에서 periportal region에 더 많은 양의 lipid droplet이 축적되어 있는 macrosteatosis와 microsteatosis가 관찰되었다. 또한, 지방성 염증의 한 증상으로 보이는 neutorophil이 peripheral 영역에서 관찰되었다. 실험군에서는 UDCA와 C&JM_50E 복합물 400mg/kg, 200mg/kg 투여로 지방구 크기, 지방 축적과 지방성 염증이 농도 의존적으로 개선되는 것으로 나타났다.
2) 급성 알코올 투여시 울금-용암해수 50% 에탄올(주정)추출물(C&JM_50E)에 의한 혈중 알코올농도 변화
① 혈중 에탄올 농도 측정(숙취분석)
혈중 에탄올과 아세트알데히드의 농도 를 측정하기 위하여 8주령 SD 흰쥐에 3일간 울금-용암해수 50% 에탄올(주정)추출물(C&JM_50E) 200mg/kg과 상쾌환 아침(easytomorrow) 시료 200mg/kg를 경구투여하였다.
3일째 울금-용암해수 50% 에탄올(주정)추출물(C&JM_50E) 200mg/kg과 상쾌환 아침 시료 200mg/kg 시료를 투여 후 1시간30분 후 Ethanol 2g/kg을 경구투여 하였다.
에탄올 투여 30분, 60분, 90분, 120분. 240분, 360분에 꼬리끝부분 절단을 통해 혈액(0.5ml)을 채취하여 1,900×g에서 20분 간 원심 분리하여 혈청만을 분리하였고, 알코올 측정 kit를 사용하여 NADH량의 변화를 340nm 에서 흡광도로 측정한 결과이다.
울금-용암해수 50% 에탄올(주정)추출물 200 mg/kg과 상쾌환 아침 시료 200 mg/kg가 알코올 분해 대사에 미치는 영향을 알아 보기 위해 SD rat에 알코올을 투여하기 30분 전에 시료를 경구 투여하고, 알코올 투여 전 및 투여 후 30, 60, 90, 120, 240, 360분 후에 혈액을 채취해 에탄올 농도를 측정하였다.
도 20에서와 같이 측정 kit의 Master reaction mix와 blood serum의 반응시간을 0.5시간, 1시간, 2시간, 4시간으로 측정한 결과, 1시간 이후 비슷한 반응결과를 나타내었다. Reaction반응 1시간(1.0h Reaction) 분석결과를 보면 알코올 숙취유도 대조군은 알코올 섭취 후 6시간 이후도 혈중 에탄올 농도가 크게 감소되지 않았고, 상쾌환 아침 시료 200mg/kg 투여군은 알코올 투여 후 30분 혈중 에탄올 수준에 비하여 55.2% 에탄올 농도가 급격히 감소하였고 알콜올 투여 6시간에서 숙취유도 대조군(Ethanol_CTL)에 비하여 상쾌환 아침 시료 투여군의 혈중 에탄올 농도가 60.7% 급격히 통계적으로 유의하게 감소하는 결과를 보였다. 그리고 Reaction반응 1시간(1.0h Reaction) 분석결과를 보면 울금-용암해수 50% 에탄올(주정)추출물 200mg/kg투여군은 알코올 투여 후 30분 혈중 에탄올 수준에 비하여 34.7% 에탄올 농도가 급격히 감소하였고 알콜올 투여 6시간에서 숙취유도 대조군(Ethanol_CTL)에 비하여 울금-용암해수 50% 에탄올(주정)추출물 200 mg/kg 투여군의 혈중 에탄올 농도가 34.9% 급격히 통계적으로 유의하게 감소하는 결과를 보였다.
② 혈중 아세트알데히드 농도 측정(숙취분석)
혈중 에탄올과 아세트알데히드의 농도 를 측정하기 위하여 8주령 SD 흰쥐에 3일간 울금-용암해수 50% 에탄올(주정)추출물(C&JM_50E) 200mg/kg과 상쾌환 아침 (easytomorrow) 시료 200 mg/kg을 경구투여한다. 3일째 울금-용암해수 50% 에탄올(주정)추출물 200mg/kg과 상쾌환 아침 시료 200mg/kg을 투여 후 1시간30분 후 Ethanol(2g/kg)을 경구투여 한다. 에탄올 투여 30분, 60분, 90분, 120분. 240분, 360분에 꼬리끝부분 절단을 통해 혈액(0.5ml)을 채취하여 1,900×g에서 20분 간 원심 분리하여 혈청만을 분리하였고, 아세트알데히드 kit를 사용하여 NADH량의 변화를 340nm 에서 흡광도로 측정하였다.
혈중 알코올이 분해될 때 생성되는 아세트알데히드(acetaldehyde)는 숙취의 주된 원인으로 알코올 대사가 빠르게 진행되지 않아 쌓이는 많은 양의 아세트알데히드(acetaldehyde)는 지방간과 간 손상을 유발하는 것으로 알려져 있다. 실험동물에 30% 알코올을 투여 후 알코올 대사 중간산물인 아세트알데히드(acetaldehyde)의 농도를 확인한 결과로, 울금-용암해수 50% 에탄올(주정)추출물(C&JM_50E) 200mg/kg과 상쾌환 아침(easytomorrow) 시료(200 mg/kg)이 알코올 분해 대사에 미치는 영향을 알아 보기 위해 SD rat에 알코올을 투여하기 30분 전에 시료를 경구 투여하고, 알코올 투여 전 및 투여 후 30, 60, 90, 120, 240, 360분 후에 혈액을 채취해 아세트알데히드(acetaldehyde) 농도를 측정하였다.
도 21에서와 같이 측정 kit의 Master reaction mix와 blood serum의 반응시간을 0.5시간, 1시간, 2시간, 4시간으로 측정한 결과, 1시간 이후 비슷한 반응결과를 나타내었다. Reaction반응 1시간 (2.0h Reaction) 분석결과를 보면 알코올 숙취유도 대조군은 알코올 섭취 후 6시간 이후도 혈중 아세트알데히드(acetaldehyde) 농도가 크게 감소되지 않았고, 상쾌환 아침(easytomorrow) 시료(200 mg/kg)투여군은 알코올 투여 후 90분후 혈중 아세트알데히드(acetaldehyde) 수준에 비하여 48.7% 아세트알데히드(acetaldehyde) 농도가 급격히 감소하였고 알콜올 투여 6시간에서 숙취유도 대조군(Ethanol_CTL)에 비하여 상쾌환 아침(easytomorrow) 시료 (200mg/kg)투여군의 혈중 아세트알데히드(acetaldehyde) 농도가 59.1% 급격히 통계적으로 유의하게 감소하는 결과를 보였다. 그리고 Reaction반응 1시간(2.0h Reaction) 분석결과를 보면 울금-용암해수 50% 에탄올(주정)추출물(C&JM_50E) 200mg/kg투여군은 알코올 투여 후 60분후 혈중 아세트알데히드(acetaldehyde) 수준에 비하여 50.7% 아세트알데히드(acetaldehyde) 농도가 급격히 감소하였고 알콜올 투여 6시간에서 숙취유도 대조군(Ethanol_CTL)에 비하여 울금-용암해수 50% 에탄올(주정)추출물(C&JM_50E) 200mg/kg투여군의 혈중 아세트알데히드(acetaldehyde) 농도가 36.7% 급격히 통계적으로 유의하게 감소하는 결과를 보였다.
이상의 결과로 보아 시험 물질인 울금-용암해수 50% 에탄올(주정)추출물(C&JM_50E)은 에탄올의 분해뿐 아니라, 흡수를 지연시켜 혈중 에탄올 및 아세트알데히드(acetaldehyde)의 농도를 효과적으로 감소시키며, 장시간 지속적인 효과를 나타냄을 알 수 있다.
정리해보면,
울금-용암해수 50% 에탄올(주정)추출물(C&JM_50E) 400㎎/㎏, 200㎎/㎏, 100㎎/㎏ 투여로 SOD와 Catalase 활성이 대조군에 비해 유의성 있게 증가된 것으로 보아 항산화 효소들을 효과적으로 활성화시켜 알코올성 산화스트레스로부터 간 보호 효과를 갖는다.
지질과산화는 활성산소종들에 의해 매개된 기작으로서 여러 동물 인체실험들을 통하여 다양한 종류의 간 손상을 일으키는 것으로 알려져 왔다. GSH는 생명체에 존재하는 주요 비단백 thiol로서 인체의 산화적 손상에 대한 방어체계에서 중요한 역할을 담당한다.
과산화물을 환원시키는 다양한 효소 과정에서 각종 효소들과 함께 세포 내외적으로 주요한 역할을 하는 비효소계 항산화제인 GSH는 redox 반응 및 해독작용을 통하여 정상 세포들의 구조 및 기능을 유지시켜준다.
GSH 함량은 SD rat_Nr(정상군)에 비해 EtOH_CTL(대조군)에서 감소하였으며, 울금-용암해수 50% 에탄올(주정)추출물(C&JM_50E) 400㎎/㎏, 200㎎/㎏ 투여군에서 유의성 있게 증가된 것으로 보아 알코올 섭취로 인해 GSH 함량이 감소되는 것을 보호했을 것이라고 사료된다.
그리고 혈청중 alcohol과 acetaldehyde 농도 및 total cholesterol, HDL-cholesterol, LDL-cholesterol, triglyceride, free fatty acid, total protein, albumin 농도 및 AST, ALT, LDH 활성를, 알코올 대사과정의 효소인 ADH와 ALDH의 활성, 그리고 간 조직의 병리조직학적((H&E, oil red o) 관찰로 알코올성 지방간에 대한 개선 효과를 검증하여 울금-용암해수 50% 에탄올(주정)추출물(C&JM_50E) 400㎎/㎏, 200㎎/㎏ 투여군에서 간손상을 억제하는 유의성 있는 결과를 확인하였다.
또한, 급성으로 알코올을 투여한 알코올성 지방간 휜쥐모델에서 혈중 alcohol및 acetaldehyde농도 측정하여 울금-용암해수 50% 에탄올(주정)추출물(C&JM_50E) 400㎎/㎏, 200㎎/㎏ 투여군에서 EtOH_CTL(대조군)에 비하여 유의성 있게 감소하였다.
간세포의 염증과 관련된 cytokine 유전자인 TNF-α, TGF-β1, IL-1β, IL-6, IL-8 등의 발현정도와 간 손상 정도와는 상당한 연관성이 있다고 알려져 있으며, 모두 간조직의 감염이 시작 또는 진행되는 과정에서 그 발현이 증가한다.
염증 cytokine 유전자 발현 분석은 염증 인자인 NOS-II, COX-2, TNF-α, IL-1 β, IL-6을 통하여 mRNA 유전자발현 분석을 하였으며, 울금-용암해수 50% 에탄올(주정)추출물(C&JM_50E) 400㎎/㎏, 200㎎/㎏투여군에서 대조군에 비해 IL-1β, TNF-α, NOS-II mRNA 유전자발현은 유의성 있게 감소되었으며, IL-6와 COX-2 mRNA 유전자 발현도 감소되는 것을 확인하였다. 또한 간성상세포와 쿠퍼세포에서 생성되는 간 섬유화 관련 유전자인 MMP-2, MMP-9 mRNA 유전자 발현은 울금-용암해수 50% 에탄올(주정)추출물(C&JM_50E) 400㎎/㎏, 200㎎/㎏, 투여군에서 대조군에 비하여 감소되는 것으로 나타났다.
그리고 급성 알코올 숙취유도 동물모델에서 울금-용암해수 50% 에탄올(주정)추출물(C&JM_50E)은 에탄올의 분해뿐 아니라, 흡수를 지연시켜 혈중 에탄올 및 아세트알데히드(acetaldehyde)의 농도를 효과적으로 감소시키며, 장시간 지속적인 효과를 나타내었다.
즉, 울금-용암해수 50% 에탄올(주정)추출물(C&JM_50E)은는 알코올에 의한 간 손상을 보호하는데 기여할 수 있다.
Claims (4)
- 울금을 알코올과 용암해수를 이용하여 추출하되,
건조 울금 1㎏ 중량대비,
50% 에탄올(주정) 92.5중량%와 용암해수 7.5중량%로 이루어진 혼합(추출)용매 8L를 이용하여 60℃에서 7시간 추출하고, 보조첨가물을 첨가한 후 분무건조시켜 분말화시킴을 특징으로 하는 울금과 용암해수를 이용한 간기능 개선용 울금추출분말소재. - 제1항에 있어서,
상기 혼합(추출)용매를 이용하여 추출한 추출물은 여과과정과 농축과정를 거친 후 보조첨가물을 첨가한 후 분무건조시켜 분말화시키되,
상기 보조첨가물 고형분 함량 기준으로 카세인나트륨 20중량%와 덱스티린 20중량% 임을 특징으로 하는 울금과 용암해수를 이용한 간기능 개선용 울금추출분말소재. - 울금을 알코올과 용암해수를 이용하여 추출하되,
