Abstract
본 발명은 멜리틴과 소수성 고분자를 결합시켜 입자화한 후, 정전기적 인력을 이용하여 나노입자를 코팅하여 나노입자 복합체를 형성하므로, 멜리틴 자체의 독성을 낮출뿐 아니라, 암세포를 효과적으로 사멸시킬 수 있는 멜리틴 기반의 나노입자 복합체 및 이의 제조방법에 대한 것이다.
Description
본 발명은 멜리틴과 소수성 고분자를 결합시켜 자가조립으로 입자화한 후, 정전기적 인력을 이용하여 나노입자를 코팅하여 나노입자 복합체를 형성하므로, 멜리틴 자체의 독성을 낮출 수 있고, 암세포를 효과적으로 사멸시킬 수 있는 멜리틴 기반의 나노입자 복합체 및 이의 제조방법에 대한 것이다.
멜리틴(Melittin)은 벌 독에서 추출되며, 26개의 아미노산으로 구성된 단백질로, 세포막 파괴 및 용혈 현상을 일으키므로, 하기 특허문헌에 기재된 바와 같이 살균, 항염증제, 항암제 등으로 사용하기 위한 연구가 진행되고 있다.

하지만, 상기 멜리틴은 뛰어나 효능에도 불구하고 정상세포도 공격하는 강한 독성을 가지고 있어, 치료제로 바로 쓰기가 어려운 문제가 있다.
본 발명은 상기와 같은 문제점을 해결하기 위해 안출된 것으로,
본 발명은 멜리틴과 소수성 고분자를 결합시켜 자가조립으로 나노 입자화한 후, 정전기적 인력을 이용하여 알부민과 히알루론산과 같은 생체물질을 이용하여 나노입자의 최외각을 코팅하여 나노입자 복합체를 형성하므로, 멜리틴 자체의 독성을 낮출 수 있고, 암세포를 효과적으로 사멸시킬 수 있는 멜리틴 기반의 나노입자 복합체 및 이의 제조방법을 제공하는데 그 목적이 있다.
본 발병은 앞서 본 목적을 달성하기 위하여 다음과 같은 구성을 가진 실시예에 의해 구현된다.
본 발명의 일 실시예에 따르면, 본 발명에 따른 나노입자 복합체는 멜리틴을 포함하는 나노입자와, 상기 나노입자를 에워싸는 코팅층을 포함하는 것을 특징으로 한다.
본 발명의 다른 실시예에 따르면, 본 발명에 따른 나노입자 복합체에 있어서 상기 나노입자는 멜리틴과 화학 결합하는 작용기를 포함하는 생체적합성을 가진 소수성 고분자와, 멜리틴을 포함하는 것을 특징으로 한다.
본 발명의 또 다른 실시예에 따르면, 본 발명에 따른 나노입자 복합체에 있어서 상기 나노입자는 양전하를 가지며, 상기 코팅층은 음전하성 고분자로 이루어지는 것을 특징으로 한다.
본 발명의 또 다른 실시예에 따르면, 본 발명에 따른 나노입자 복합체에 있어서 상기 멜리틴과 상기 소수성 고분자의 작용기가 화학 반응하여, 상기 멜리틴과 소수성 고분자는 결합하여 결합체를 형성하게 되며, 상기 결합체가 자가 조립을 통해 나노입자를 형성하는 것을 특징으로 한다.
본 발명의 또 다른 실시예에 따르면, 본 발명에 따른 나노입자 복합체에 있어서 상기 나노입자는 폴리머좀 또는 리포좀과 같은 형태를 가지는 것을 특징으로 한다.
본 발명의 또 다른 실시예에 따르면, 본 발명에 따른 나노입자 복합체에 있어서 정전기적 인력에 의해, 상기 나노입자의 외 표면에 음전하성 고분자로 이루어진 코팅층이 형성되는 것을 특징으로 한다.
본 발명의 또 다른 실시예에 따르면, 본 발명에 따른 나노입자 복합체는 낮은 pH 조건(산성 조건)에서 코팅층이 제거되는 것을 특징으로 한다.
본 발명의 또 다른 실시예에 따르면, 본 발명에 따른 나노입자 복합체의 제조방법은 멜리틴과 화학 결합하는 작용기를 포함하는 소수성 고분자와, 멜리틴을 혼합하고 반응시켜 양전하를 가지는 폴리머좀 또는 리포좀을 형성하는 나노입자형성단계와, 상기 표면 양전하를 가지는 폴리머좀 또는 리포좀과 음전하성 고분자를 혼합하고 반응시켜 폴리머좀 또는 리포좀의 외 표면에 음전하성 고분자로 이루어진 코팅층을 형성하는 코팅층형성단계를 포함하는 것을 특징으로 한다.
본 발명의 또 다른 실시예에 따르면, 본 발명에 따른 나노입자 복합체의 제조방법에 있어서 상기 나노입자형성단계에서는 상기 멜리틴과 소수성 고분자의 작용기가 화학 반응하여, 상기 멜리틴과 소수성 고분자는 결합하여 결합체를 형성하게 되며, 상기 결합체가 자가 조립을 통해 폴리머좀 또는 리포좀과 같은 형태의 나노입자를 형성하는 것을 특징으로 한다.
본 발명의 또 다른 실시예에 따르면, 본 발명에 따른 나노입자 복합체의 제조방법에 있어서 상기 코팅층형성단계에서는 정전기적 인력에 의해 나노입자의 외 표면에 음전하성 고분자로 이루어진 코팅층이 형성되는 것을 특징으로 한다.
본 발명은 앞서 본 실시예에 의해 다음과 같은 효과를 얻을 수 있다.
본 발명은 멜리틴과 소수성 고분자를 결합시켜 자가조립으로 나노 입자화한 후, 정전기적 인력을 이용하여 알부민과 히알루론산과 같은 생체물질을 이용하여 나노입자의 최외각을 코팅하여 나노입자 복합체를 형성하므로, 멜리틴 자체의 독성을 낮출 수 있고, 암세포를 효과적으로 사멸시킬 수 있는 효과가 있다.

이하에서는 본 발명에 따른 멜리틴 기반의 나노입자 복합체 및 이의 제조방법을 첨부된 도면을 참조하여 상세히 설명한다. 특별한 정의가 없는 한 본 명세서의 모든 용어는 본 발명이 속하는 기술분야의 통상의 지식을 가진 기술자가 이해하는 당해 용어의 일반적 의미와 동일하고 만약 본 명세서에 사용된 용어의 의미와 충돌하는 경우에는 본 명세서에 사용된 정의에 따른다. 또한, 본 발명의 요지를 불필요하게 흐릴 수 있는 공지 기능 및 구성에 대해 상세한 설명은 생략한다. 명세서 전체에서, 어떤 부분이 어떤 구성요소를 "포함"한다고 할 때 이는 특별히 반대되는 기재가 없는 한 다른 구성요소를 제외하는 것이 아니라 다른 구성요소를 더 포함할 수 있는 것을 의미한다.
본 발명의 일 실시예에 따른 멜리틴 기반의 나노입자 복합체를 도 1 내지 13을 참조하여 설명하면, 상기 나노입자 복합체는 멜리틴을 포함하는 나노입자(1)와, 상기 나노입자(1)를 에워싸는 코팅층(2)을 포함한다.
상기 나노입자(1)는 세포 내에 섭취되어 질병(예컨대, 암 등)의 치료에 이용되는 구성으로, 멜리틴 등을 포함한다. 상기 나노입자(1)는 멜리틴과 화학 결합하는 작용기를 포함하는 생체적합성을 가진 소수성 고분자와, 멜리틴을 포함하며, 상기 멜리틴은 소수성 고분자의 작용기와 화학 반응하는 반응기를 포함할 수 있다. 상기 멜리틴의 반응기와 소수성 고분자의 작용기가 화학 반응하여, 상기 멜리틴과 소수성 고분자는 결합하여 결합체(11)를 형성하게 되며, 상기 결합체(11)가 자가 조립을 통해 폴리머좀(Polymersome) 또는 리포좀(Liposome) 형태의 나노입자(1)를 형성하게 된다. 상기 멜리틴은 강한 양전하를 가지므로, 상기 나노입자(1)는 양전하를 가지게 된다.
상기 멜리틴에 반응기를 도입할 수도 있고, 상기 멜리틴에 존재하는 아민기를 반응기로 사용될 수 있으며, 상기 소수성 고분자는 생체적합성 및 소수성을 가지는 공지의 고분자가 사용될 수 있고, 예컨대 PLA(Poly lactic acid), 인지질(Lipid) 등이 사용될 수 있다. 상기 소수성 고분자에는 도입되는 작용기는 공지의 화합물이 사용될 수 있으나, 예컨대 아민기와 NHS-amine 반응하는 NHS(N-hydroxysuccinimide)가 소수성 고분자에 도입되어 작용기로 사용될 수 있다.
상기 코팅층(2)은 상기 나노입자(1)를 에워싸는 구성으로, 상기 코팅층(2)은 생체 적합성을 가진 음전하성 고분자로 이루어지게 된다. 상기 나노입자(1)는 양전하를 가지므로, 상기 음전하성 고분자와 나노입자(1)를 반응시키는 경우, 정전기적 인력에 의해 나노입자의 외 표면에 음전하성 고분자로 이루어진 코팅층이 형성되게 된다. 상기 코팅층(2)은 음전하를 가지는 공지의 고분자가 사용될 수 있으나, 예컨대 알부민, 히알루론산(Hyaluronic acid) 등이 사용될 수 있다. 암세포에서는 gp60, CD44 등이 과발현되는데, 알부민은 gp60에 결합하고, 히알루론산은 CD44 리셉트에 결합하므로, 상기 나노입자 복합체는 암세포에 선택적으로 전달할 수 있게 된다. 상기 코팅층은 정전기적 인력에 의해 형성되어 나노입자와 약한 결합을 가지므로, 상기 나노입자 복합체가 산성 용액(예컨대, 5.0 내지 5.5의 pH)과 반응하는 경우, 상기 나노입자 복합체에서 코팅층이 제거된 나노입자(1)가 외부로 노출되게 된다. 상기 나노입자 복합체는 멜리틴과 소수성 고분자를 결합시켜 자가 조립을 통해 나노 입자화한 후, 정전기적 인력을 이용하여 나노입자를 코팅을 하여 형성되므로, 상기 나노입자 복합체는 세포의 리소좀(Lysosome, pH가 5 내지 5.5임)에 효과적으로 전달되어 상기 코팅층이 제거됨으로써, 엔도좀 또는 리소좀 안에서 멜리틴이 엔도좀과 리소좀의 지질 이중막 붕괴를 유발하여 엔도좀과 리소좀 안에 있는 분해 효소(예컨대, Cathepin B 등)가 엔도좀과 리소좀 밖 세포로 이동하여 세포의 사멸을 유도하여, 정상 세포에 대한 독성이 낮출 수 있고, 암세포를 효과적으로 사멸시킬 수 있게 된다.
본 발명의 다른 실시예에 따른 멜리틴 기반의 나노입자 복합체의 제조방법을 도 1을 참조하여 설명하면, 상기 나노입자 복합체의 제조방법은 멜리틴과 화학 결합하는 작용기를 포함하는 소수성 고분자와, 멜리틴을 혼합하고 반응시켜 양전하를 가지는 나노입자를 형성하는 나노입자형성단계와, 상기 나노입자와 음전하성 고분자를 혼합하고 반응시켜 나노입자의 외 표면에 음전하성 고분자로 이루어진 코팅층을 형성하는 코팅층형성단계를 포함한다.
상기 나노입자형성단계는 멜리틴과 화학 결합하는 작용기를 포함하는 소수성 고분자와, 멜리틴을 혼합하고 반응시켜 양전하를 가지는 나노입자를 형성하는 단계로, 상기 나노입자형성단계에서는 상기 멜리틴의 반응기와 소수성 고분자의 작용기가 화학 반응하여, 상기 멜리틴과 소수성 고분자는 결합하여 결합체(11)를 형성하게 되며, 상기 결합체(11)가 자가 조립을 통해 폴리머좀(Polymersome) 또는 리포좀(Liposome) 형태의 나노입자(1)를 형성하게 된다.
상기 코팅층형성단계는 상기 나노입자와 음전하성 고분자를 혼합하고 반응시켜 나노입자의 외 표면에 음전하성 고분자로 이루어진 코팅층을 형성하는 단계로, 상기 코팅층형성단계에서는 정전기적 인력에 의해 나노입자의 외 표면에 음전하성 고분자로 이루어진 코팅층이 형성되게 된다.
이하, 실시예를 통해서 본 발명을 보다 상세히 설명하기로 한다. 하지만, 이들은 본 발명을 보다 상세하게 설명하기 위한 것일 뿐, 본 발명의 권리범위가 이에 한정되는 것은 아니다.
<실시예 1> 멜리틴 기반의 나노입자 복합체의 제조
1. 나노입자의 형성
DMSO 800uL에 3mg의 PLA-NHS(MW:7000)를 녹여 형성한 제1용액과, DW 200uL에 1mg의 멜리틴(MW:2840)을 녹여 형성한 제2용액을 혼합하여 혼합용액을 형성하고, pH가 알카리성 조건일 때 PLA의 작용기 NHS가 멜리틴의 말단 N-terminius amine group과 반응하므로, 혼합용액의 pH 8.4가 되도록 맞추어 준 후, shaking incubation을 이용하여 25℃에서 2시간 동안 반응시켰다. 이후, Tip sonication(UP200S_Watts)을 이용하여, cycle 1, amplitude 20% 조건으로 30초간 처리하였다. 이후, 혼합용액의 DMSO 농도가 5%가 되도록 DW로 희석하고 미반응 물질을 제거하기 위해 30KDa의 amicon tube를 이용하여 4000rpm, 5분 조건으로 원심분리를 진행한 후, DW로 재분산하여 4000rpm, 5분 조건으로 2번 워싱하고, 펠릿을 1ml의 DW로 재분산 후, tip sonication을 이용하여 cycle 1, amplitude 20% 조건으로 30초간 처리하여 나노입자(이하, 'PLA-MEL'이라 함)를 얻었다.
2. 나노입자 복합체의 형성
(1) DW 2ml에 20ml의 알부민(human serum albumin)을 녹여 형성한 알부민 용액과 실시예 1의 1의 최종 용액(나노입자가 분산되어 있음) 1ml와 혼합한 후, 48시간 동안 magnetic stirring을 수행하였다. 이때, 멜리틴은 양전하를, 알부민은 음전하를 가지고 있으므로 나노입자의 표면에 알부민이 코팅되게 된다. 이후, 미반응 물질을 제거하기 위해 100KDa의 amicon tube를 이용하여 4000rpm, 5분 조건으로 원심분리를 진행한 후, DW로 재분산하여 4000rpm, 5분 조건으로 2번 워싱하고, 펠릿을 1ml의 DW로 재분산하여 나노입자에 알부민이 코팅된 나노입자 복합체(이하, 'PLA-MEL+HSA'이라 함)를 형성하였다.
(2) 알부민 대신 히알루론산(hyaluronic acid)을 사용한 것을 제외하고는 다른 조건을 실시예 1의 2의 (1)과 동일하게 하여 나노입자에 히알루론산이 코팅된 나노입자 복합체(이하, 'PLA-MEL+HA'이라 함)를 얻었다.
<실시예 2> 나노입자에 코팅층이 형성되었음을 확인
1. 상기 나노입자(PLA-MEL) 및 나노입자 복합체(PLA-MEL+HSA, PLA-MEL+HA)의 표면 전하값을 측정하여 그 결과를 도 2에 나타내었고, Dynamic light scattering(DLS) 방법을 이용하여 상기 나노입자(PLA-MEL) 및 나노입자 복합체(PLA-MEL+HSA, PLA-MEL+HA)를 측정하여 그 결과를 도 3에 나타내었고, 상기 나노입자(PLA-MEL) 및 나노입자 복합체(PLA-MEL+HSA, PLA-MEL+HA)를 TEM으로 측정하여 그 결과를 4에 나타내었다. 도 4의 (a)는 PLA-MEL의 TEM 이미지이고(scale bar : 50), 도 4의 (b)는 PLA-MEL의 TEM 이미지이며(scale bar : 100), 도 4의 (c)는 PLA-MEL+HSA의 TEM 이미지이고(scale bar : 100), 도 4의 (d)는 PLA-MEL+HA의 TEM 이미지이다(scale bar : 100).
2. 도 2를 보면, 나노입자(PLA-MEL)는 강한 양전하(20.9mV)를 띄고 있으나, 나노입자 복합체(PLA-MEL+HSA, PLA-MEL+HA) 각각은 -21.6mV, -15.9mV의 전하를 가지므로, 나노입자에 음전하를 가지는 알부민, 히알루론산이 코팅되어 있음을 알 수 있다. 또한, 도 3을 보면, 나노입자 복합체(PLA-MEL+HSA, PLA-MEL+HA)가 나노입자(PLA-MEL)에 비해 입자 크기가 큼을 확인할 수 있어, 나노입자에 코팅층이 형성되었음을 간접적으로 확인할 수 있고, 도 4의 (a) 및 (b)를 보면 나노입자가 형성되었음을 알 수 있고 도 4의 (c) 및 (d)를 나노입자 외면에 검은색으로 보이는 코팅층이 형성되었음을 알 수 있다.
<실시예 3> pH가 낮아짐에 따라 나노입자 복합체의 코팅층이 분해됨을 확인
1. pH가 낮아짐에 따라 알부민 코팅이 벗겨짐을 확인하기 위해, lysosome의 pH와 같이 DW의 pH를 5.0으로 맞춘 뒤, 상기 DW에 나노입자 복합체(PLA-MEL+HSA)를 넣어 24시간 뒤에 변화를 Dynamic light scattering(DLS) 방법을 이용하여 측정하여 그 결과를 도 5에 나타내었고, TEM으로 확인하여 그 결과를 도 6에 나타내었다.
2. 도 5을 보면, 24시간 반응 후 나노입자 복합체의 크기가 작아지고, 도 6을 보면, 알부민 코팅으로 보이는 검은 부분이 사라짐을 확인할 수 있어, pH가 낮아짐에 따라 나노입자 복합체의 코팅층이 분해됨을 알 수 있다.
<실시예 4> 나노입자 복합체의 세포 독성 및 항암 효과 확인
1. 상기 나노입자 복합체(PLA-MEL+HSA)의 세포 독성 및 항암 효과를 확인하고자, 인체혈관내피세포(HUVEC)와 유방암세포(MCF-7)에서 나노입자 복합체 처리에 의한 세포 사멸효과 및 기작을 확인하였다.
2. EGM-2 medium, 혼탁 배양 조건에서 배양한 인체혈관내피세포(HUVEC), RPMI medium, 혼탁 배양 조건에서 배양한 유방암세포(MCF-7)와 항암제 저항이 있는 유방암세포(MCF-7 ADR) 각각을 96well plate에 seeding한 후(1×104), 24시간 동안 배양하고, 농도가 20ug/ml 또는 100ug/ml인 멜리틴, 멜리틴의 농도가 20ug/ml 또는 100ug/ml인 나노입자(PLA-MEL), 멜리틴의 농도가 20ug/ml 또는 100ug/ml인 나노입자 복합체(PLA-MEL+HSA)를 처리하고 일정 시간 동안 반응시킨 후, MTT assay를 수행하여 그 결과를 도 7 내지 9에 나타내었다.
3. 도 7을 보면, 정상세포인 인체혈관내피세포(HUVEC)에 멜리틴을 처리한 경우보다, 나노입자(PLA-MEL) 또는 나노입자 복합체(PLA-MEL+HSA)를 처리한 경우 현저하게 낮은 세포 독성을 가짐을 알 수 있고, 도 8 및 9를 보면, 나노입자(PLA-MEL) 또는 나노입자 복합체(PLA-MEL+HSA)가 유방암 세포를 사멸시킴을 알 수 있다. 또한, 도 7 내지 9를 종합하면, 나노입자 복합체(PLA-MEL+HSA)가 나노입자(PLA-MEL)에 비해 상대적으로 낮은 정상세포 독성을 가지고 더 강한 항암효과를 가짐을 알 수 있다.
<실시예 5> 나노입자 복합체의 세포 섭취(Cell uptake) 시 항암 기작을 확인
1. Confocal dish에 1×105의 유방암 세포(MCF-7)를 seeding한 후, 24시간 동안 incubation을 진행하였다.
2. 실시예 5의 1에서 준비된 유방암 세포에, Alexa 555 형광(붉은색)을 labeling한 나노입자 복합체(PLA-MEL+HSA)를 3시간, 24시간 동안 섭취시킨 후 워싱을 진행하고, 리소좀 안에 있는 효소(protease)인 Cathepin B(녹색으로 염색)와 세포핵(DAPI로 염색하여 파란색을 띔)을 염색하고, confocal microscope를 이용하여 세포 이미징을 진행하여, 배율을 달리하는 그 결과를 도 10 및 11에 나타내었고, confocal 3D 이미지를 도 12에 나타내었다.
3. 실시예 5의 1에서 준비된 유방암 세포에, 나노입자 복합체(PLA-MEL+HSA)를 3시간, 24시간 동안 섭취시킨 후, TEM으로 확인하여 그 결과를 도 13에 나타내었다. 도 13에서 화살표는 세포 내의 나노입자 복합체의 위치를 나타낸다.
4. 도 10 내지 12를 보면, 나노입자 복합체(PLA-MEL+HSA)를 처리하지 않은 대조군(CTL)에서 Cathepsin B 형광(녹색)이 보이지 않음을 알 수 있는데 반해, 나노입자 복합체(PLA-MEL+HSA)를 처리한 경우(3h, 24h), 강한 Cathepsin B 형광(녹색)이 나타남을 알 수 있다. Cathepsin B는 엔도좀과 리소좀 안에 있는 분해 효소이므로, 위와 같은 사실을 통해 나노입자 복합체(PLA-MEL+HSA)가 엔도좀과 리소좀을 붕괴시켜 Cathepsin B가 배출되어 강한 형광을 나타내었다고 판단된다.
5. 도 13을 보면, 3시간 경과한 후 나노입자 복합체가 리소좀안에 유입됨을 알 수 있고, 24시간 경과한 후 세포가 사멸하여 세포핵이 붕괴되었고, 세포막의 파괴가 이루어졌음을 알 수 있다. 즉, 전체적으로 세포 형태가 사멸된 세포 또는 사멸 과정 중의 세포임을 보이고 있다.
6. 위와 같은 사실을 통해, 나노입자 복합체는 산성인 리소좀, 엔도좀 안에 유입되어 코팅이 벗겨져 멜리틴이 세포막 붕괴 및 용혈반응 유발하여 세포막/질 구조를 붕괴시킴으로써 리소좀, 엔도좀 안에 있는 효소인 Cathepin B가 배출되게 됨을 알 수 있다.
이상에서, 출원인은 본 발명의 다양한 실시예들을 설명하였지만, 이와 같은 실시예들은 본 발명의 기술적 사상을 구현하는 일 실시예일 뿐이며, 본 발명의 기술적 사상을 구현하는 한 어떠한 변경예 또는 수정예도 본 발명의 범위에 속하는 것으로 해석되어야 한다.

Claims (10)
- 멜리틴을 포함하는 나노입자와, 상기 나노입자를 에워싸는 코팅층을 포함하는 것을 특징으로 하는 나노입자 복합체.
- 제1항에 있어서,
상기 나노입자는 멜리틴과 화학 결합하는 작용기를 포함하는 생체적합성을 가진 소수성 고분자와, 멜리틴을 포함하는 것을 특징으로 하는 나노입자 복합체. - 제1항에 있어서,
상기 나노입자는 양전하를 가지며,
상기 코팅층은 음전하성 고분자로 이루어지는 것을 특징으로 하는 나노입자 복합체. - 제2항에 있어서,
상기 멜리틴과 상기 소수성 고분자의 작용기가 화학 반응하여, 상기 멜리틴과 소수성 고분자는 결합하여 결합체를 형성하게 되며, 상기 결합체가 자가 조립을 통해 나노입자를 형성하는 것을 특징으로 하는 나노입자 복합체. - 제4항에 있어서,
상기 나노입자는 폴리머좀 또는 리포좀 형태를 가지는 것을 특징으로 하는 나노입자 복합체. - 제3항에 있어서,
정전기적 인력에 의해, 상기 나노입자의 외 표면에 음전하성 고분자로 이루어진 코팅층이 형성되는 것을 특징으로 하는 나노입자 복합체. - 제6항에 있어서,
상기 나노입자 복합체는 산성 반응 조건에서 코팅층이 제거되는 것을 특징으로 하는 나노입자 복합체. - 멜리틴과 화학 결합하는 작용기를 포함하는 소수성 고분자와, 멜리틴을 혼합하고 반응시켜 양전하를 가지는 나노입자를 형성하는 나노입자형성단계와, 상기 나노입자와 음전하성 고분자를 혼합하고 반응시켜 나노입자의 외 표면에 음전하성 고분자로 이루어진 코팅층을 형성하는 코팅층형성단계를 포함하는 것을 특징으로 하는 나노입자 복합체의 제조방법.
- 제8항에 있어서,
상기 나노입자형성단계에서는 상기 멜리틴과 소수성 고분자의 작용기가 화학 반응하여, 상기 멜리틴과 소수성 고분자는 결합하여 결합체를 형성하게 되며, 상기 결합체가 자가 조립을 통해 폴리머좀 또는 리포좀 형태의 나노입자를 형성하는 것을 특징으로 하는 나노입자 복합체의 제조방법. - 제8항에 있어서,
상기 코팅층형성단계에서는 정전기적 인력에 의해 나노입자의 외 표면에 음전하성 고분자로 이루어진 코팅층이 형성되는 것을 특징으로 하는 나노입자 복합체의 제조방법.
