도 2는 상기 CA 복합체의 표면 고정화를 확인하기 위한 공초점 형광 현미경 사진으로, (A) GFP-doc를 처리한 대조군과 (B) GFP-doc-mCbpA를 처리한 실험군을 비교한 것이다. 왼쪽은 CBM을 포함하는 mCbpA에서 세포 이미지이고, 오른쪽은 동일한 세포의 표면에 나타난 녹색 형광 프로브 이미지로, 위상차(pahse contrast)에 의해 탐지되었다.
도 3은 CA 복합체의 형성이 탄산탈수효소 활성에 미치는 영향을 나타낸 것이다. (A)는 CA 복합체와 단일 효소(cCA)의 상대적인 효소 활성 수준을, (B)는 CA 복합체와 단일 효소(cCA)의 안정성을 비교한 것이다.
도 4는 CA 복합체가 세포 생장에 미치는 영향을 나타낸 것이다. 세포 밀도(cell density)를 위해 흡광도(optical density, OD)는 540 nm 파장에서 측정되었다.
도 5는 C. vulgaris에서 CA 복합체가 말로닐-CoA의 세포 내 농도에 미치는 영향을 HPLC로 분석한 것이다.
도 6은 C. vulgaris의 지질 함량 분석 결과를 나타낸 것으로서, 지질은 클로로포름을 이용하여 추출하고, 건중량법(dry weight method)으로 분석하였다.
도 7은 지방산 생산량 및 함량을 가스 크로마토그래피(GC)로 분석한 결과를 나타낸 것으로서, (A)는 총 지방산 생산량, (B)는 총 지방산 함량을 나타낸 것이다.
Claims (13)
- 탄산탈수효소 및 도커린 모듈의 결합체;가
코헤신 모듈 및 셀룰로오스 결합 모듈(cellulose binding module, CBM)을 포함하는 소형 셀룰로오스 결합 단백질;에 결합된 탄산탈수효소 복합체.
- 제1항의 탄산탈수효소 복합체를 포함하는 이산화탄소 고정화 반응 촉진용 조성물.
- 제1항의 탄산탈수효소 복합체를 포함하는 녹조류의 지질 생산 촉진용 조성물.
- 하기 단계를 포함하는 탄산탈수효소 복합체의 제조방법:
(1) 탄산탈수효소를 암호화하는 유전자 및 도커린 모듈을 암호화하는 유전자를 포함하는 벡터를 주입한 제1형질전환체 제조단계;
(2) 코헤신 모듈을 암호화하는 유전자 및 셀룰로오스 결합 모듈을 암호화하는 유전자를 포함하는 벡터를 주입한 제2형질전환체 제조단계; 및
(3) 상기 제1 및 제2 형질전환체를 각각의 배지에서 배양하는 단계.
- 제4항에 있어서,
상기 탄산탈수효소 복합체는 상기 제1형질전환체의 배양물과 제2형질전환체의 배양물을 몰분율 2:1로 혼합하여 제조하는 것을 특징으로 하는, 탄산탈수효소 복합체의 제조방법.
- 제4항에 있어서,
상기 (1) 단계의 탄산탈수효소를 암호화하는 유전자는 서열번호 1의 염기서열을 포함하는 것을 특징으로 하는, 탄산탈수효소 복합체의 제조방법.
- 제4항에 있어서,
상기 (1) 단계의 도커린 모듈을 암호화하는 유전자는 서열번호 2의 염기서열을 포함하는 것을 특징으로 하는, 탄산탈수효소 복합체의 제조방법.
- 제4항에 있어서,
상기 (2) 단계의 코헤신 모듈을 암호화하는 유전자는 서열번호 3의 염기서열을 포함하는 것을 특징으로 하는, 탄산탈수효소 복합체의 제조방법.
- 제4항에 있어서,
상기 (2) 단계의 셀룰로오스 결합 모듈을 암호화하는 유전자는 서열번호 4의 염기서열을 포함하는 것을 특징으로 하는, 탄산탈수효소 복합체의 제조방법.
- 제4항에 있어서,
상기 (1) 및 (2) 단계의 형질전환체는 대장균(Escherichia coli)인 것을 특징으로 하는, 탄산탈수효소 복합체의 제조방법.
- 제4항에 있어서,
상기 (1) 및 (2) 단계의 벡터는 pColdⅡ플라스미드 벡터인 것을 특징으로 하는, 탄산탈수효소 복합체의 제조방법.
- 하기 단계를 포함하는 이산화탄소 고정화 방법:
(1) 제1항의 탄산탈수효소 복합체를 제조하는 단계; 및
(2) 상기 제조된 탄산탈수효소 복합체에 이산화탄소를 공급하여 상기 이산화탄소를 중탄산염으로 전환시키는 단계.
- 녹조류에 제1항의 탄산탈수효소 복합체를 처리하는 단계를 포함하는, 녹조류의 지질 생산성을 증가시키는 배양 방법.
