상기에서 박층 크로마토그래피를 수행함으로써 수득된 5개 분획물(OC1, OC2, OC3, OC4 또는 OC5)을 대상으로 디스크확산법(disc diffusion method, DD)을 수행하여 이들의 항균활성을 확인하였다.The five fractions (OC1, OC2, OC3, OC4 or OC5) obtained by performing thin layer chromatography above were subjected to a disk diffusion method (DD) to confirm their antibacterial activity.
먼저, 스타필로코커스 오레우스 균주를 실험예 1-1에 기재된 바와 동일한 방법 및 조건으로 준비하여 현탁액을 제조하였다. 100 ㎕의 제조된 현탁액을 취하여 MH 고체 배지에 멸균된 면봉을 이용하여 고르게 도포하고, 이를 배양하였다. 한편, 실시예 2의 5개 분획물을 250, 500 또는 1,000 ㎍/㎖의 농도가 되도록 DMSO에 희석한 뒤, 직경 6 ㎜의 BBL TM 블랭크 종이 디스크(BBL TM blank paper discs, BD, 미국)에 점적하고, 이를 상온에서 완전히 건조시켜 준비하였다. 상기 배양된 고체 배지에 준비된 디스크를 올리고, 37℃하에서 16시간 동안 추가 배양하였다. 이때, 양성 대조군으로 암피실린을 동일한 농도로 사용하였다. 배양 후, 확인된 결과를 도 8에 나타내었다.First, a Staphylococcus aureus strain was prepared in the same manner and under the same conditions as described in Experimental Example 1-1 to prepare a suspension. 100 μl of the prepared suspension was taken and evenly applied to MH solid medium using a sterile cotton swab, and cultured. 도 8에 나타난 바와 같이, OC2, OC3 및 OC4 분획물을 처리한 경우 스타필로코커스 오레우스 균주의 성장이 억제되었다. 특히, OC2 분획물이 가장 우수한 항균활성을 나타내어 13 내지 14 ㎜의 클리어 존(clear zone)을 형성하였다.As shown in FIG. 8, when the OC2, OC3 and OC4 fractions were treated, the growth of the Staphylococcus aureus strain was inhibited. According to claim 3, wherein the organic solvent is any one or more selected from the group consisting of n-hexane, ethyl acetate and butanol, a pharmaceutical composition for the prevention or treatment of diseases caused by bacterial infection.
According to claim 1, wherein the bacteria is a gram-positive bacteria, a pharmaceutical composition for the prevention or treatment of diseases caused by bacterial infection.
According to claim 5, wherein the Gram-positive bacteria is Staphylococcus (Staphylococcus) sp. strain, a pharmaceutical composition for preventing or treating a disease caused by a bacterial infection.
A health functional food for the prevention or improvement of diseases caused by bacterial infection, comprising a rock extract or a fraction thereof as an active ingredient.
An antibacterial composition comprising a rockson extract or a fraction thereof as an active ingredient.
The antibacterial composition according to claim 8, wherein the antibacterial composition exhibits antibacterial activity against Gram-positive bacteria.
An antibacterial cosmetic composition comprising an extract or a fraction thereof as an active ingredient.
