Abstract
본 발명은 세포 투과능 및 근육 재생능을 가지는 이중 기능성 펩타이드 및 이의 용도에 관한 것으로, 본 발명에 따른 이중 기능성 펩타이드는 근육세포를 재생시키는 기능이 있어, 근육 재생이 필요한 질환의 예방 또는 치료에 유용하고, 세포 투과능을 가지므로 펩타이드의 세포 투과를 위해 별도의 펩타이드를 부착하거나 기타 제제를 첨가할 필요가 없어 최종적으로 효율적인 근육 재생 효과를 발휘하는바, 다양한 외과적 재생치료에 간편하게 적용이 가능하고 치료기간도 단축시킬 수 있는 장점이 있다.
Description
본 발명은 세포 투과능 및 근육 재생능을 가지는 이중 기능성 펩타이드 및 이의 용도에 관한 것으로, 더욱 상세하게는 서열번호 1 내지 8 중에서 어느 하나의 아미노산 서열로 표시되는 펩타이드를 포함하는 근육 질환 예방 또는 치료용 약제학적 조성물 및 근육 질환 치료용 조직공학적 지지체에 관한 것이다.
골격근은 자가 재생이 가능하지만 외상, 선천성 결함, 노인성 진환, 종양 절제로 인해 조직 손상이 커지면 수술과 같은 치료 접근이 필요하고, 자가 혹은 동종 근모세포 혹은 유전자 치료제를 근육 내로 주사하는 방법 등이 이용되고 있다.
근육 줄기세포와 근육이 손상된 경우 재생에 관여하는 근모세포는 근육 위성세포(satellite cells) 중 일부분에서 분리되는 것으로 알려져 있는데, 위성세포는 근육의 기저막과 근섬유 겉을 둘러싸고 있는 막 사이에 존재한다. 위성세포는 기저막에 존재하고 있다가 외부 자극 또는 스트레스로 근육이 손상되면 분열 및 증식 과정을 통해 단백질을 합성시키고, 휴지기 상태의 위성세포로 다시 돌아와 세포 수를 유지한다. 위성세포는 근육 재생 치료 방식에서 많이 활용되는데, 많은 양의 세포를 빠른 시간 내에 배양하여 얻을 수 있고, 바이러스나 기타 벡터들로 유전자 전달이 용이하며 또한 세포간 접촉하는 경우에 근육 섬유로 분화되어 성장이 멈추기 때문에 과발현에 의해 암세포로 전환될 염려가 없어 조직공학적 이용 가능성에 대해 기대되고 있다.
광범위한 골격근 결손을 치료하기 위해서는 근섬유로 직접 분화하는 위성세포와 같은 골격근 유래 줄기세포를 이식하거나 세포와 지지체를 이용한 조직공학적인 방법이 시도되고 있다. 그러나 줄기세포나 위성세포는 생체 외에서 증식이 가능하지만 배양기간이 짧아서 근육 내로 주입했을 때 증식, 분화 및 재생 능력이 낮은 것으로 보고되었다.
Kin 등이 2007년에 발표한 논문에 의하면, 토끼 다리 골격근 상처 부위에 콜라겐으로 제조된 지지체를 이식했을 경우 대조군에 비해 경미한 반흔조직과 새로운 근조직이 형성된 것을 확인한 바 있다(Shuichi Kin, et al., Regeneration of Skeletal Muscle Using In Situ Tissue Engineering on an Acellular Collagen Sponge Scaffold in a Rabbit Model, ASAIO Journal 2007.). 골격근 조직에서 in situ 재생 접근을 위해 PLA(polylactate acid) 지지체를 실험용 근조직에 이식하고 침투하는 세포를 관찰한 결과 Pax3, Pax7, MyoD 등을 발현하는 근육 위성세포와 전구세포 등이 발견되고 IGF-1의 성장인자를 처리하면 지지체로 침투하는 근육세포의 수가 증가됨을 관찰한 바 있다.
그러나 단백질성 성장인자는 반감기가 짧고, 상용화에 있어 종양 유발, 다기능성에 의한 부작용 등을 유발할 수 있는 단점이 있다. 이를 극복하기 위해 후생유전학적 분자 타겟을 이용하여, 이를 선택적으로 제어하는 물질에 의한 줄기세포 분화기술이 필요한 상황이다.
히스톤 탈아세틸화 효소 (Histone Deacetylase, HDAC)는 히스톤을 탈아세틸화시켜 염색질을 응축시키고 유전자전사를 억제하는 효소이다. 히스톤 (Histone)단백질은 DNA와 함께 뉴클레오좀 (Nucleosome)을 구성하는 요소이며 히스톤 단백질의 구조는 여러 가지 수식이 일어날 수 있는데, 히스톤 아세틸화와 메틸화는 DNA 메틸화와 함께 특정 유전자의 발현을 조절하는 중요한 후천적 기작이다. 일반적으로 특정유전자 발현이 활성화 된 경우 히스톤 꼬리에 히스톤 아세틸화효소 (Histone acetylatrasferase, HAT)에 의해 아세틸화가 일어나 있는 반면, 히스톤 탈아세틸화 효소, HDAC에 의해 탈아세틸화가 일어난 경우에는 특정유전자 발현이 비활성화, 즉 유전자 발현이 억제된 경우이다. 적절한 HDAC의 조절은 정상적인 발달단계 및 질환발생에 있어 중요한데, 암, 염증성 질환, 신경병증을 포함한 퇴행성 질환을 비롯한 다양한 질환을 치료하기 위한 HDAC의 역할 및 이들의 조절자가 연구되고 있다.
이전의 연구에서 HDAC 억제제는 염증치료에 효과가 있을 뿐 아니라 줄기세포의 신경, 뼈, 및 간조직으로의 특이적인 분화를 촉진하는 것으로 보고된 바 있다. 그러나 저분자 합성 물질이나 siRNA 등의 유전성 물질을 제외하고는 펩타이드 기반의 HDAC억제제는 전무한 상황이다.
이에 본 발명자들은 상기와 같은 종래기술의 문제점을 해결하고자 예의 연구 노력한 결과, HDAC의 발현을 억제하고 줄기세포로부터 근육세포로의 분화를 촉진하여 근육세포를 재생시킴과 동시에 세포 투과능을 가지는 이중 기능성의 특정 펩타이드를 확인하고, 본 발명을 완성하였다.
본 발명의 목적은 세포 투과능과 근육 재생능을 가지는 이중 기능성 펩타이드를 유효성분으로 포함하는 근육 질환 예방 또는 치료용 약제학적 조성물을 제공하는데 있다.
본 발명의 다른 목적은 상기 이중 기능성 펩타이드를 포함하는 근육 질환 치료용 조직공학적 지지체를 제공하는데 있다.
상기 목적을 달성하기 위하여, 본 발명은 서열번호 1 내지 8 중 어느 하나의 아미노산 서열로 표시되는 이중 기능성 펩타이드를 유효성분으로 포함하는 근육 질환 예방 또는 치료용 약제학적 조성물을 제공한다.
본 발명은 또한 상기 이중 기능성 펩타이드를 포함하는 근육 질환 치료용 조직공학적 지지체를 제공하는데 있다.
본 발명에 따른 이중 기능성 펩타이드는 근육세포를 재생시키는 기능이 있어, 근육 재생이 필요한 질환의 예방 또는 치료에 유용하고, 세포 투과능을 가지므로 펩타이드의 세포 투과를 위해 별도의 펩타이드를 부착하거나 기타 제제를 첨가할 필요가 없고, 펩타이드 자체의 세포 투과능으로 단시간 내에 세포의 이행, 증식 및 분화를 촉진하여 최종적으로 효율적인 근육 재생 효과를 발휘하는바, 다양한 외과적 재생치료에 간편하게 적용이 가능하고 치료기간도 단축시킬 수 있는 장점이 있다.
도 1은 근육 전구세포 (C2C12)에 서열번호 1 내지 8의 펩타이드 및 서열번호 9의 대조 펩타이드를 처리하고 10분 후에 세포 내 투과 정도를 확인하기 위해 Confocal microscope로 분석한 결과를 나타낸 것이다(서열번호 1: P0, 서열번호 2: P1, 서열번호 3: P2, 서열번호 4: P3, 서열번호 5: P4, 서열번호 6: P5, 서열번호 7: P6, 서열번호 8: P7, 서열번호 9: C1).

다른 식으로 정의되지 않는 한, 본 명세서에서 사용된 모든 기술적 및 과학적 용어들은 본 발명이 속하는 기술분야에서 숙련된 전문가에 의해서 통상적으로 이해되는 것과 동일한 의미를 갖는다. 일반적으로 본 명세서에서 사용된 명명법은 본 기술 분야에서 잘 알려져 있고 통상적으로 사용되는 것이다.
본 발명에서는, 세포 투과능 및 근육 재생능을 가지는 이중 기능성 펩타이드를 제조하고, 근육 전구세포 및 중간엽 유래 줄기세포(Mesenchymal stem cell, MSC)에 이를 처리하는 실험을 수행하였다. 그 결과, 상기 이중기 능성 펩타이드가 세포 내로 투과되며, HDAC 5의 발현이 감소하고, 근육세포의 마커 단백질의 발현이 증가함을 확인하였다.
따라서, 본 발명은 일 관점에서 서열번호 1 내지 8 중에서 어느 하나의 아미노산 서열로 표시되는 이중 기능성 펩타이드를 유효성분으로 포함하는 근육 질환 예방 또는 치료용 약제학적 조성물에 관한 것이다.

본 발명에 있어서, 서열번호 2는 서열번호 1의 시스테인 (cysteine)을 메티오닌(methionine)으로 치환한 것으로, 인접한 시스테인-시스테인 또는 떨어진 시스테인-시스테인 구조에 존재하는 S-S 결합에서 산화를 방지하기 위해, 시스테인을 메티오닌으로 치환한 것이다. 서열번호 1에 시스테인은 모두 3개이며, 서열번호 2 내지 8은 이를 메티오닌으로 치환한 것이다.
본 발명에서 상기 펩타이드는 세포 투과능과 근육 재생능을 가지는 이중 기능성인 것을 특징으로 할 수 있다.
본 발명에서 상기 펩타이드는 유전자 재조합 기법에 의해 생산되는 단백질에 비해 대량으로 제조할 수 있다는 점에서 펩타이드 합성법(solid phase peptide synthesis)을 이용하여 제조되는 것이 바람직하나, 이에 한정되지는 않는다.
한편, 본 발명자들은 상기 펩타이드가 세포 내로 투과 가능하고, 근육 재생 효과가 우수함을 in vitro 실험을 통해 확인하였다.
본 발명의 일 실시예에 따르면, 상기 이중 기능성 펩타이드는 세포 내로 투과 가능한 아미노산 서열로 표시된다. 기존에 활용되었던 안티센스올리고뉴클레오타이드는 세포 내로 투과 가능성이 전무하기 때문에, 세포 내의 HDAC 5와 결합하여 발현을 저해하기 위해서는 반드시 바이러스성 혹은 비바이러스성 유전자 전달체를 사용해야 하며, 이러한 바이러스성 전달체는 안전성 우려에서 임상적 실용화에 제한점이 있었다. 그러나 본 발명에 따른 펩타이드는 자체적으로 세포 내로 투과 가능하기 때문에 부가적인 전달체를 사용할 필요가 없다.
본 발명의 "조성물"은 특정 성분을 포함하는 산물뿐만 아니라, 특정 성분의 배합에 의해 직접 또는 간접적으로 만들어지는 임의의 산물을 포함하는 것으로 간주된다.
본 발명에 있어서, 상기 조성물에는 약제학적으로 허용가능한 담체를 추가로 함유할 수 있고, 상기 담체는 식염수, 멸균수, 링거액, 완충식염수, 덱스트로즈 용액, 말토덱스트린 용액, 글리세롤, 에탄올로 구성된 군에서 선택된 하나 이상인 것을 특징으로 할 수 있으나, 이에 한정되는 것은 아니다.
본 발명에 있어서, 상기 조성물에는 부형제, 완충제, 항균성 보존제, 계면활성제, 산화방지제, 긴장성 조정제, 방부제, 증점제 및 점도개질제로 구성된 군에서 선택된 하나 이상의 보조제를 추가로 함유하는 것을 특징으로 할 수 있으나, 이에 한정되는 것은 아니며, 상기 각각의 성분은 업계에서 통상적으로 사용되는 원료를 당해 분야에서 통용되는 범위 내에서 적절히 변형하여 사용할 수 있다.
본 발명에 있어서, 상기 조성물은 경구투여용 제제, 주사용 제제 및 국소이식용 겔제로 이루어진 군에서 선택될 수 있으나, 이에 한정되는 것은 아니며, 공지의 기술을 이용하여 적합한 제형으로 제제화할 수 있다 (Joseph Price Remington, Remington's Pharmaceutical Science; 17th edition, MackPublishing Company, EastonPA).
본 발명에 있어서, 상기 조성물은 의약 분야에서 통상적으로 이용되는 경로를 통해 투여될 수 있으며, 비경구 국소 투여 경로가 바람직하고, 예를 들어 근육내 경로를 통하여 투여할 수 있다.
본 발명의 "유효성분"은 일반적으로 활성 성분으로서 본 발명에 따른 세포 투과능 및 근육 재생능의 이중 기능성 펩타이드를 치료 유효량으로 함유하는 것을 의미한다.
본 발명의 "근육 질환"은 근육조직 또는 근세포가 손상을 받아서 근육조직이 소실(loss)되는 모든 질환을 의미하며,"근육 질환의 예방 또는 치료용 조성물"은 "근육 재생용 조성물"과 동일한 의미로 사용된다.
본 발명에 있어서, 상기 근육 질환은 근디스트로피 (muscular dystrophy), 근위축증(muscular atropy), 근감소증 (muscular sarcopenia), 근육염 (Myositis), 다발성 근육염 (polymyositis), 말초혈관 질환 (peripheral vascular disease) 및 섬유증 (fibrosis)으로 구성된 군에서 선택되는 것을 특징으로 할 수 있으나, 이에 한정되는 것은 아니다.
본 발명의 "치료"는 (a) 질환, 질병 또는 증상의 발전의 억제; (b)질환, 질병 또는 증상의 경감, 또는 (c) 질환, 질병 또는 증상을 제거하는 것을 의미한다.
본 발명에 있어서, 상기 조성물은 내생적 자가 줄기세포의 분화를 촉진하는 데에 사용됨으로써 그 자체로 치료용 약제학적 조성물이 될 수도 있고, 혹은 치료용 이식 줄기세포와 함께 투여되어 이식된 세포가 병변 부위에서 근육세포로 분화하도록 하는 세포치료 보조제로 적용될 수도 있으며, 줄기세포가 적용될 때 사용되는 고분자성 지지체 (scaffold)의 내외부에 탑재되어 조절 방출됨으로써 줄기세포의 기능조절에도 사용될 수 있으나, 이에 한정되는 것은 아니다.
본 발명에 있어서, 상기 조성물은 바람직하게는 치료 대상의 체중 1 kg 당 0.001 ~ 1000 mg, 더욱 바람직하게는 0.1 ~ 100 mg의 용량으로 투여하는 것을 특징으로 할 수 있으나, 이에 한정되지는 않으며, 이는 환자의 체중, 연령, 성별, 건강상태, 식이, 투여시간, 투여방법, 배설율 및 질환의 중증도 등에 따라 그 범위가 다양하며, 상기 사항을 고려하여 본 기술분야의 통상의 전문가가 결정할 수 있다.
본 발명은 다른 관점에서, 상기 이중 기능성 펩타이드를 포함하는 근육 질환 치료용 조직공학적 지지체에 관한 것이다.
본 발명에 있어서, 상기 조직공학적 지지체는 세포를 담지할 수 있는 생체적합성 고분자로 이루어진 것으로, 주입형 지지체 혹은 2,3차원 지지체를 포함하는 것을 특징으로 할 수 있으나, 이에 한정되는 것은 아니다. 주입형 지지체로는 반고형 상태로 줄기세포를 탑재하거나 탑재하지 않은 상태로 적용되며, 소재는 콜라겐, 젤라틴, 피브린젤, 알지네이트 및 히알루론산으로 구성된 군에서 선택될 수 있으나, 이에 한정되는 것은 아니다. 또한 2, 3차원 지지체로는 생체장기(심근막, 심막 등)의 탈세포 처리방법에 의해 얻어진 콜라겐 소재 지지체, 콜라겐, 합성고분자를 캐스팅한 후 나노패터닝 등의 방법으로 방향성을 제공한 것에 펩타이드를 탑재한 것을 의미하는데 이에 한정되는 것은 아니다.
이하, 실시예를 통하여 본 발명을 더욱 상세히 설명하고자 한다. 이들 실시예는 오로지 본 발명을 예시하기 위한 것으로, 본 발명의 범위가 이들 실시예에 의해 제한되는 것으로 해석되지 않는 것은 당업계에서 통상의 지식을 가진 자에게 있어서 자명할 것이다.
실시예 1: 세포 투과능 및 근육 재생능을 가지는 이중 기능성 펩타이드의 합성
서열번호 1의 펩타이드를 합성장치를 이용하여 C 말단으로부터 F-moc 고체상 화학합성방법으로 합성하였다. 즉, 블로킹 그룹(Blocking group)으로 Fmoc-(9-Fluorenylmethoxycarbonyl)이 결합된 Rink resin (0.075 mmol/g, 100 ~ 200 mesh, 1% DVB crosslinking)을 사용하여 합성하였으며, 합성기에 50 mg의 Rink Amide MBHA resin을 넣은 뒤 DMF로 resin을 스웰링(swelling) 시킨 후 Fmoc-group의 제거를 위해 20% piperidine/DMF 용액을 사용하였다. C 말단부터 서열대로 0.5M amino acid 용액(용매: DMF), 1.0M DIPEA(용매: DMF&NMP), 0.5M HBTU (용매: DMF)를 각각 5, 10, 5 당량씩 넣어 질소 기류 하에서 1~2시간 동안 반응시켰다. 상기 디프로텍션(deprotection)과 커플링(coupling) 단계가 끝날 때마다 DMF와 Methanol로 두 번씩 세척하는 과정을 거쳤다. 마지막 아미노산을 커플링(coupling) 시킨 후에도 디프로텍션(deprotection)을 해주어 Fmoc-group을 제거하였다.
합성의 확인은 닌하이드린 테스트(ninhydrin test) 방법을 이용하였고, 테스트를 거치고 합성이 완료된 resin은 건조시킨 후 Reagent K cleavage cocktail을 resin 1g 당 20ml의 비율로 넣어 3시간 shaking 시킨 후 필터링을 통해 resin과 펩타이드가 녹아 있는 cocktail을 분리하였다. 필터로 걸러진 용액을 콜드 에테르(cold ether)를 넣어주어 펩타이드를 고체상으로 결정화시키고 이를 원심분리하여 분리해내었다. 이때 에테르로 여러 번 세척과 원심분리 과정을 거쳐 Reagen K cleavage cocktail을 완전히 제거하였다. 이렇게 해서 얻어진 crude를 증류수에 녹여 액체크로마토그래피를 이용하여 분리 정제하였다. 정제된 펩타이드는 동결건조를 진행한다.

실험예 1. 세포 투과능 및 근육 재생능을 가지는 이중 기능성 펩타이드에 의한 세포 투과능 평가
6웰(well)에 근육 전구세포(C2C12)를 웰(well)당 1.5Х105개씩 이식한 후, EMEM(Eagle's minimal essential medium, ATCC)배지로 배양한다. 세포는 confluency가 50%가 될 때까지 24시간 간격으로 배지를 교체해준다. confluency가 50% 되었을 때, Essential 8 배지로 교체하고, 동시에 형광 (rhodamine)이 표지된 서열번호 1 내지 8의 펩타이드와 대조 펩타이드(서열번호 9)를 각각 10μM 처리하고, 10분 후에 세포를 고정시키고, Confocal microscope로 관찰하였다. 대조 펩타이드(서열번호 9)와 대비하여 서열번호 1 내지 8의 펩타이드가 세포 내로 투과한 것을 확인하였다 (도 1).
실험예 2.
세포 투과능 및 근육 재생능을 가지는 이중 기능성 펩타이드에 의한 HDCA 5 발현 억제 평가
근육 전구세포(C2C12)를 10파이 디쉬에 1.5Х106개씩 이식한 후, EMEM(Eagle's minimal essential medium, ATCC)배지로 배양한다. 세포는 confluency가 50%가 될 때까지 24시간 간격으로 배지를 교체해준다. confluency가 50% 되었을 때, Essential 8 배지로 교체하였다. 100μM의 서열번호 1의 펩타이드를 2시간 동안 처리한 후에, 세포 내 HDAC 5와의 결합력을 Immunoprecipitation로 확인하였다. protease inhibitor를 넣은 RIPA buffer로 harvest한 lysate를 BCA assay로 정량하였다. 1 ㎍/ml 농도의 최소 500 ㎕로 정량한 각 샘플을 micro tube에 모은 후, HDAC 5 1st antibody를 2 ㎍ 넣고 4 ℃ 냉장상태로 16시간 동안 회전 반응시켰다. protein A/G agarose beads를 20 ㎕ 넣고, 3시간 동안 4 ℃에서 반응시켰다. 이 후 beads를 원심분리하여 DPBS로 3번 세척하고 2X sample buffer를 넣고 85 ℃에서 5분간 가열하여 상층액만 수득한 뒤 western blot 하여, FITC antibody로 확인하였다. 그 결과, 서열번호 1의 펩타이드가 세포 내 HDAC 5와 결합함을 확인하였다 (도 2(a)).
상기와 동일한 조건으로 근육 전구세포(C2C12)를 배양하고, 배지를 교체해주는 동시에 서열번호 1 내지 8 각각의 펩타이드 100μM과 대조 펩타이드(서열번호 9) 100μM을 처리하고, 5일째에 HDAC 5의 발현을 웨스턴블랏으로 관찰하였다. 그 결과, 도 2(b)에서 나타난 바와 같이, 펩타이드를 처리하지 않은 군(NT)과 대조 펩타이드(서열번호 9)를 처리한 군에 비교하여, 서열번호 1 내지 8의 펩타이드를 처리하였을 때, HDAC 5의 발현이 억제된 것을 확인하였다.
실험예 3. 세포 투과능 및 근육 재생능을 가지는 이중 기능성 펩타이드에 의한 줄기세포로부터 근육세포로의 분화능 평가
중간엽 유래 줄기세포 3X106 cells 를 bacterial culture dish에 3일간 배양하여 spheroid가 형성되도록 하고, 이 때 사용하는 배지는 low glucose DMEM with 10%FBS and 1%P/S로 하였다. 3일 후에 supernatant는 없애고, spheroid 만 모아서 10ug/ml Collagen Type I 이 코팅된 cell culture dish에 seeding 하여 4일간 배양하였다. 이 때 사용하는 배지는 DMEM/F12 with 1ng/ml TGFb (Transforming growth beta), 1xNEAA (nonessential amino acids), 1xITS (insulin-transferrin-selenium)이다. 4일 후, 배지를 low glucose DMEM with 2% FBS and 10ng/ml IGF-1 (Insulin growth factor-1)으로 바꿔주고 2주간 더 분화를 시켜서 근육분화 마커를 확인하였다. 펩타이드에 의한 근육분화 마커 발현효율을 보기 위해, 펩타이드를 처리하지 않고 근육분화배지만 사용한 군 (NT), 서열번호 1 내지 8의 펩타이드 각 100uM, 대조 펩타이드(서열번호 9) 100uM을 13일 동안 처리하였다. 근육 분화 마커인 Dysotrophin, Myogenin의 유전자 발현정도는 real time RT-PCR로 측정하였다. 각 유전자의 증폭에 사용된 PCR 프라이머의 염기서열은 다음과 같다.

도 3에 나타난 바와 같이, 서열번호 1 내지 8의 펩타이드를 처리한 경우, Dystrophin, Myogenin의 유전자 발현이 증가하였고, 대조 펩타이드(서열번호 9)와 펩타이드를 처리하지 않은 경우에는 근육분화 마커 유전자의 발현이 증가하지 않았다.
또한 이 발현 마커들을 세포 면역형광분석으로 측정하였는데, 배양된 세포를 슬라이드에서 1:1 Acetone/methanol로 고정 후 tris-buffered saline (TES: Sigma)으로 세척하였다. 이후 30분간 2% bovine serum albumin (Sigma)와 5% goat serum (Sigma)이 첨가된 용액으로 고정시켰다. 이후 a-SMA, Dysotrophin에 대한 일차항체 (Santa Cruz Biotechnology, Santa Cruze, CA, USA)를 사용하여 실온에서 1시간 동안 반응시켰다. 핵은 DAPI로 염색시키고, a-SMA는 FITC가 결합된 anti-avidin을 30분동안 실온에서 반응시키고, 다시 TBS로 세척하였다. Dystrophin은 rhodamine이 결합된 anti-avidin을 30분 동안 실온에서 반응시키고, 다시 TBS로 세척하고, confocal microscope로 관찰하였다. 도 4에 나타난 바와 같이, 서열번호 1 내지 8의 펩타이드가 처리된 세포군에서 a-SMA, Dystrophin의 발현이 뚜렷하게 관찰된 바, 펩타이드에 의한 근육 분화, 재생효과가 우수함을 확인할 수 있었다.
이상으로 본 발명 내용의 특정한 부분을 상세히 기술하였는 바, 당업계의 통상의 지식을 가진 자에게 있어서 이러한 구체적 기술은 단지 바람직한 실시양태일 뿐이며, 이에 의해 본 발명의 범위가 제한되는 것이 아닌 점은 명백할 것이다. 따라서, 본 발명의 실질적인 범위는 청구항들과 그것들의 등가물에 의하여 정의된다고 할 것이다.

Claims (10)
- 서열번호 1 내지 8 중에서 어느 하나의 아미노산 서열로 표시되는 펩타이드를 유효성분으로 포함하는 근육 질환 예방 또는 치료용 약제학적 조성물.
- 제1항에 있어서, 상기 펩타이드는 세포 투과능과 근육 재생능을 가지는 이중 기능성인 것을 특징으로 하는 약제학적 조성물.
- 제1항에 있어서, 상기 펩타이드는 줄기세포로부터 근육세포로의 분화를 촉진시키는 것을 특징으로 하는 약제학적 조성물.
- 제1항에 있어서, 상기 근육 질환은 근디스트로피 (muscular dystrophy), 근위축증(muscular atropy), 근감소증 (muscular sarcopenia), 근육염 (Myositis), 다발성 근육염 (polymyositis), 말초혈관 질환 (peripheral vascular disease) 및 섬유증 (fibrosis)으로 구성된 군에서 선택되는 것을 특징으로 하는 약제학적 조성물.
- 제1항에 있어서, 상기 약제학적 조성물은 경구투여용 제제, 주사용 제제 및 국소이식용 겔제로 구성된 군에서 선택되는 것을 특징으로 하는 조성물.
- 제1항에 있어서, 상기 약제학적 조성물은 치료 대상의 체중 1 kg 당 0.001 ~ 1000 mg의 용량으로 투여하는 것을 특징으로 하는 약제학적 조성물.
- 서열번호 1 내지 8 중에서 어느 하나의 아미노산 서열로 표시되는 펩타이드를 포함하는 근육 질환 치료용 조직공학적 지지체.
- 제7항에 있어서, 상기 펩타이드는 세포 투과능과 근육 재생능을 가지는 이중 기능성인 것을 특징으로 하는 조직공학적 지지체.
- 제7항에 있어서, 상기 조직공학적 지지체는 콜라겐, 젤라틴, 피브린젤, 알지네이트 및 히알루론산으로 구성된 군에서 선택되는 어느 하나인 것을 특징으로 하는 조직공학적 지지체.
- 제7항에 있어서, 상기 조직공학적 지지체는 치료 대상의 체중 1 kg 당 0.001 ~ 1000 mg의 용량으로 투여하는 것을 특징으로 하는 조직공학적 지지체.
