상기의 방법으로 개발된 유자 및 배 식물캘러스 상등액 및 추출액의 콜라겐 생성 촉진 활성은 다음과 같이 평가하였다. 인간섬유아세포 (human fibroblast cell, CCD-986sk)를 96 well plate에 1 × 104 cell/well씩 분주하고 10% FBS가 포함된 DMEM 배양액에서 24시간 배양한 후 serum free 배지로 단계적으로 희석한 유자 및 배 식물캘러스 상등액 및 추출액 혹은 유전자 재조합 인간 형질전환성장인자 (recombinant human transforming growth factor-beta, rhTGF-β)를 각 well에 100 μL씩 가하고 24시간 동안 CO2 incubator에서 배양하였다. 이후 세포 배양액을 취해 Procollagen type I C-peptide EIA kit (Takara, Japan)을 이용하여 콜라겐 생합성 양을 측정하였다. 먼저, anti-POD conjugate solution을 100 μL씩 각 well에 첨가하고 시료 혹은 표준액을 20 μL씩 넣은 후 교반한 다음 37oC에서 3시간 반응시켰다. 이후 내용물을 제거하고 인산염완충액 (pH 7.4) 400 μL로 4회 세척하였다. 세척 후 각 well 당 기질 용액을 100 μL씩 넣고 20~30oC에서 15분간 반응시킨 뒤 ELISA stop solution을 100 μL씩 첨가하여 발색을 중지시킨 후 교반하였다. 450 nm에서 흡광도를 측정한 후 표준농도곡선을 이용하여 콜라겐 생합성 양을 산정하였다.Collagen production promoting activity of the callus supernatant and extract of citron and pear plant developed by the above method was evaluated as follows.
