Abstract

Description
본 발명은 2016년 2월 4일에 출원된 미국 가출원 62/291,361,의 이익(benefit)을 주장하며, 이는 본 명세서에서 기재된 것처럼 모든 목적을 위하여 참고 문헌으로 포함된다.
본 발명은 질병 치료를 위한 항체-약물 상승작용(Antibody-Drug Synergism, ADS) 화합물 및 이의 사용방법에 관한 것이다.
항-혈관신생(Anti-angiogenesis) 전략은 삼출성 황반변성(exudative AMD) (또한, 습성 황반변성(wet AMD)으로도 알려짐)과 같은 안구 신생혈관(ocular neovascular) 질병 치료에 효과적이다. 현재, 여러 혈관내피성장인자(VEGF)-중화(neutralizing) 생물학적 약물(biologic drugs)이 시중에 나와있다. 원래, 암을 위해 Genetech에 의하여 개발된 VEGF-A에 대한 항체인 베바시주맙(bevacizumab)(avastin)은 초기에 FDA 승인 없이(off-label) 습성 AMD에 처방되었다. 발명의 실시예
실시예로서, 베바시주맙은 유리액 내에서 3-4일의 반감기로 가수분해하는 링커에 의하여 닌테다닙에 연결된다. 이 ADS에서, 베바시주맙은 VEGF 차단 활성을 유지한다. 유리액으로 유리체강내 주입하면, 닌테다닙은 ADS 화합물로부터 천천히 방출되고 모(parent) ADS 화합물이 제거되기(cleared) 전에 효과적인 농도를 유지한다. Claims (19)
- 피험자의 제 1 표적을 차단하는 고전(classic) 항체 또는 변형된 생물학적 분자인 항체를 제공하는 단계;
피험자의 상기 제 1 표적 또는 제 2 표적을 차단하는 저분자제(small molecule agent)인 약물을 제공하는 단계;
상기 항체 및 상기 약물을 링커(linker)로 연결하여 항체-약물 상승작용(Antibody-Drug Synergism, ADS) 화합물을 형성하는 단계; 및
상기 ADS 화합물로 질병을 치료하는 단계를 포함하되,
여기에서, 상기 링커는 일정 시간 동안 피험자 내에서 가수분해되어 상기 항체 및 상기 약물이 동시에 그들의 기능을 발휘하도록 하고,
여기에서, 상기 ADS 화합물은 상기 ADS 화합물의 상승작용 때문에 상기 항체 또는 상기 약물의 단독 보다 더 우수한 효능을 구현하는 것인 피험자의 질병 치료에서의 상승작용 및 향상된 효능을 생성하는 방법. - 제 1항에 있어서,
상기 질병은 안 질환(an ocular disease), 피부 질환(a dermatological disease), 또는 관절 질환(a joint disease )인 것인 방법. - 제 2항에 있어서,
상기 안 질환은 비정상적인 혈관신생(abnormal angiogenesis) 및 혈관 누수(vessel leakage)를 포함하는 신생혈관 질병인 것인 방법. - 제 3항에 있어서,
상기 ADS 화합물은 유리체강내(intravitreal), 전안방내(intracameral), 맥락막위(suprachoroidal), 결막하(subconjunctival), 테논낭하(subtenon), 또는 국소 부위(topical)의 안구(ocular)를 통해 피험자의 눈으로 전달되거나(delivered) 주사되는(injected) 것인 방법. - 제 1항에 있어서,
상기 항체는 VEGF, PDGF 및 PIGF의 군의 일원(family members) 및 이들의 수용체로 이루어진 군으로부터 선택되는 하나 또는 그 이상의 전구-혈관신생 인자(pro-angiogenesis factors)의 활성을 차단하는 것인 방법. - 제 5항에 있어서,

상기 항-혈관신생 억제제는 스쿠알라민(Squalamine)인 것인 방법. - 제 1항에 있어서,
상기 링커는 1 내지 24시간, 1 내지 28일, 또는 1 내지 4개월의 반감기로 가수분해되는 것인 방법. - 피험자의 제 1 표적을 차단하는 고전 항체 또는 변형된 생물학적 분자인 항체;
