Abstract
Description
도 2는 본 발명의 일 실험예에 있어서, 라카아제(10 U/g-기질)를 포함하거나 포함하지 않은 배지에서 볏짚을 처리할 때, 침지된 볏짚(CRS)으로부터 유래된 페놀 화합물의 pH에 따른 가용화 정도를 나타내는 그래프이다(대조군 (●); pH 4.0 (○); pH 4.5 (▼); pH 5.0 (△); 및 pH 5.5 (■)).
도 3은 본 발명의 일 실험예에 있어서, 라카아제를 포함하는 배지에서 볏짚을 처리할 때, 침지된 볏짚으로부터 유래된 페놀 화합물의 pH 4.5에서 라카아제 투여량에 따른 가용화 정도를 나타내는 그래프이다((5 U/g-기질 (●); 10 U/g-기질 (○); 15 U/g-기질 (▼); 20 U/g-기질 (△)).

Claims (10)
- 전처리 공정 또는 당화 공정 중에 바이오매스에서 생성되는 독성 물질을 라카아제로 감소시키는 무독화 과정에 있어서,
대상 바이오매스와는 상이하며 천연 중개자(mediator)로서 p-쿠마르산(p-coumaric acid)을 함유하는 2차 바이오매스 기질을 첨가하여 라카아제에 중개자를 보충하는 단계를 포함하며,
상기 2차 바이오매스 기질은 밀기울로서, p-쿠마르산이 1.5mM 로 공급되도록 첨가하는 것을 특징으로 하는 라카아제의 무독화 활성을 증진시키는 방법. - 제 1항에 있어서, 상기 2차 바이오매스 기질은 상기 1차 바이오매스 기질의 중량 대비 0.098% 이하로 포함되는 것을 특징으로 하는 라카아제의 무독화 활성을 증진시키는 방법.
- 바이오매스와, 상기 바이오매스와는 상이하며 p-쿠마르산을 함유하는 2차 바이오매스 기질을 혼합하여 분쇄하고, 분쇄된 바이오매스들을 침지시키는 제 1 단계; 및
배양 배지에 리그노셀룰라아제를 생산하는 곰팡이 컨소시엄을 접종시켜 배양하여 생산된 효소 혼합물 및 라카아제(laccase)로 상기 침지된 바이오매스 혼합물을 동시에 전처리 및 당화시켜 당화물을 제조하는 제 2 단계;를 포함하며,
상기 2차 바이오매스 기질은 밀기울로서, p-쿠마르산이 1.5mM로 공급되도록 첨가하는 것을 특징으로 하는 바이오매스의 동시 전처리 및 당화 방법. - 제 3항에 있어서, 상기 2차 바이오매스 기질은 상기 1차 바이오매스 기질의 중량 대비 0.098% 이하로 포함되는 것을 특징으로 하는 바이오매스의 동시 전처리 및 당화 방법.
- 제 3항에 있어서, 상기 곰팡이 컨소시엄은 Pholiota adiposa와 Armillaria gemina의 혼합물인 것을 특징으로 하는 바이오매스의 동시 전처리 및 당화 방법
- 제 3항에 있어서, 상기 당화는 pH 5.0, 35℃에서 90 내지 110 RPM의 속도로 교반하며, 36시간 동안 수행되는 것을 특징으로 하는 바이오매스의 동시 전처리 및 당화 방법.
- 제 3항에 있어서, 상기 제 2 단계는 보호제로서 계면활성제를 추가로 첨가하는 것을 특징으로 하는 바이오매스의 동시 전처리 및 당화 방법.
- 제 7항에 있어서, 상기 계면활성제는 폴리옥시에틸렌소르비탄모노라우레이트(polyoxyethylene sorbitan monolaurate, Tween-20)인 것을 특징으로 하는 바이오매스의 동시 전처리 및 당화 방법.
- Pholiota adiposa 및 Armillaria gemina의 곰팡이 컨소시엄, 라카아제, 및 p-쿠마르산을 포함하는 2차 바이오매스 기질을 유효성분으로 포함하며, 상기 2차 바이오매스 기질은 밀기울이며, p-쿠마르산이 1.5mM로 공급되도록, 상기 1차 바이오매스 기질의 중량 대비 0.098%로 포함되는 것을 특징으로 하는 바이오매스의 동시 전처리 및 당화용 조성물.
- 제 9항에 있어서, 상기 조성물은 폴리옥시에틸렌소르비탄모노라우레이트(polyoxyethylene sorbitan monolaurate, Tween-20)인 계면활성제를 더 포함하는 것을 특징으로 하는 바이오매스의 동시 전처리 및 당화용 조성물.
