Cells were collected at 10, 20, 30, 60, and 120 minutes, respectively, while the functional agent (0.2%) was added to the cultured cells. The collected cells were washed 3 to 4 times with PBS (phosphate buffered saline) and scraped off using a scraper. Cells were harvested using centrifugation, 100 μl of PBS at 4 ° C was added, and the cells were disrupted with an ultrasonic pulverizer for 30 seconds. 코티닌 분석은 HITACHI L-2000 (HITACHI Co. Japan)을 이용하였다. 분석조건으로 니코틴 정량을 위한 이동상은 7 % CH3OH, 2 mM NaH2PO4, 0.2 % 인산(phosphoric acid) 및 1 mM 헵탄 술폰산 나트륨(heptane sulfonate sodium)을 사용하였으며, 코티닌 정량을 위한 이동상은 2 % CH3OH, 2 mM NaH2PO4, 0.1 % 인산 및 1 mM 헵탄 술폰산 나트륨을 사용하였다. 준비한 샘플을 80 ㎕주입(injection)하여 UV 260 nm에서 유속(flow rate)을 1 ㎖/분으로 하여 검출하였다. 니코틴은 아세트아닐리드(acetanilide)의 피크 높이와 비교하여 정량하였으며, 코티닌은 코티닌 표준품((-)-Cotinine, Sigma사, USA, C-0430)을 가지고 표준곡선을 가지고 검량하였다. 그 결과, 도 3에서 볼 수 있는 것처럼, 기능성 제제는 대조군과 비교하여 니코틴 함량은 감소하고, 코티닌 함량은 증가시키는 것으로 나타났다.Cotinine analysis was performed using HITACHI L-2000 (HITACHI Co., Japan).
