Abstract
본 발명은 감귤 유래의 박테리아 셀룰로오스를 주재로 하는 생분해성 및 무독성 필름 타입의 의료용 드레싱 제재에 관한 것으로, 셀룰로오스 필름은 4주 만에 효과적으로 생분해 되었고 분해된 셀룰로오스는 백서의 체내에서 어떠한 독성도 유발하지 않았으므로 생체적합한 창상치유용 드레싱 제재를 제공하는 뛰어난 효과가 있다.
Description
본 발명은 감귤 유래의 박테리아 셀룰로오스를 주재로 하는 생분해성 및 무독성 필름 타입의 의료용 드레싱 제재에 관한 것이다.
셀룰로오스는 지구상에서 가장 흔한 유기물로 1894년 Brown에 의해 최초로 식초산균이 셀룰로오스를 생산하는 것으로 보고된 후 현재까지 박테리아 셀룰로오스의 특성 및 효능에 대한 연구가 활발히 진행되고 있다. 특히 박테리아 셀룰로오스의 독특한 3차원 나노 구조는 습윤 환경을 조성해주고, 외부 환경으로부터의 감염을 예방할 수 있어 최근 창상 치유 및 세포 지지체로서의 활용 등 의료 산업에서 연구가 활발히 진행되고 있고 그 임상적 유용성도 증명되고 있다. Kucharzewski 등은 정맥성 궤양(venous ulcer)이 있는 환자에 박테리아 셀룰로오스를 적용하여 박테리아 셀룰로오스가 창상치유의 속도를 증가시켜준다고 입증하였고(Kucharzewski M, Slezak A, and Franek A: Topical treatment of non-healing venous leg ulcers by cellulose membrane. Phlebologie 32: 147, 2003), Czaja 등은 박테리아 셀룰로오스를 2도 - 3도 화상에 적용하여 창상치유의 효과를 입증하였다(Czaja W, Young D, Kawecki M, and R MBJ: The Future Prospects of Microbial Cellulose in Biomedical Applications. Biomacromolecules 8: 1, 2007). Fontana 등은 화상이나 만성 궤양(chronic ulcer)에 박테리아 셀룰로오스를 적용하여 창상치유 효과를 입증하였는데, 통증경감, 감염으로부터 창상 보호, 창상치유과정의 가속화 및 비용절감의 효과가 있다고 주장하였다(Fontana J, de Souza A, Fontana C, Torriani I, Moreschi J, Gallotti B, de Souza S, Narcisco G, Bichara J, and Farah L: Acetobacter cellulose pellicle as a temporary skin substitute. Appl Biochem Biotechnol 24-25: 253, 1990).
이상적인 드레싱 제재는 유해한 외부 환경으로부터 창상을 차단하고 보호하며, 미생물의 감염을 예방하고 초기 삼출물을 적절히 흡수하여야 하며, 염증 반응을 억제할 수 있어야 하고 물리적, 기계적 내구성이 높아야 한다. 그 동안 창상 드레싱 제재로서 이용될 수 있는 소재는 생체조직에 대하여 거부반응이 최소화되는 소재, 즉 생체조직에 대하여 불활성을 갖는 합성고분자 부류 등으로 극히 한정되어 왔다. 하지만 최근 인체 친화성이 우수한 천연 물질에서 유래한 소재들을 이용하여 창상 드레싱 제재 및 인공 피부로의 개발이 활발히 연구되고 있다. 히알루론산(hyaluronic acid), 알긴산(alginate), 스클래로글루칸(scleroglucan) 등이 대표적인 천연 물질 유래 바이오 제재들이다. 이러한 새로운 재료의 사용시에는 반드시 그 물질에 대한 생체 적합성 및 독성평가를 시행하여 그 안전성을 확보해야만 임상적으로 사용이 가능하다. 최근 한 논문에서 미더덕 껍질로부터 분리한 셀룰로오스 필름이 4주 후 마우스의 피하에서 완벽히 생분해되고 독성 또한 나타내지 않는 것을 확인한 바 있다(Jung Y, Jung S, Lee Y, Kim J, Kim J, Lee E, Choi H, Kim H, Lee J, Kang B, Wan J, and Hwang D: Biodegradability and Toxicity of Regenerated Cellulose Films Prepared from the Tunicate Styela Clava. Biomaterials Research 13: 87, 2009).
한편, 박테리아 셀룰로오스가 다양한 산업적 가능성으로 인해 주목을 받으면서 대량생산 하기 위한 많은 연구가 이루어졌는데 특히, 감즙이나 코코넛 등 소비는 적지만 생산량이 많은 잉여 과실류를 천연 재료 배양액으로 사용하여 박테리아 셀룰로오스를 대량생산 할 경우, 환경친화적일 뿐만 아니라 고부가가치를 창출 할 수 있다(Kim S, Kim J, Wee Y, Park D, and Ryu H: Production of bacterial cellulose by Gluconacetobacter sp. RKY5 isolated from persimmon vinegar. Appl Biochem Biotechnol 129-132: 705, 2006). 그 중에서도 감귤류는 국내의 제주도에서만 연간 60만 톤이 생산되어 생과와 감귤주스 이외의 가공품 개발에 의한 소비가 적어 대부분이 농축액 상태로 저장되거나 폐기되는 실정으로 잉여 과실류에 속하고 현재 다양한 가공품 개발이 요구되고 있다(Jung j, Kim j, and Choi k: Pilot production of bacterial cellulose by gluconacetobacter hansenii TL-2C. j Korean Soc Food Sci Nutr 36: 1341, 2007).
본 발명자들은 글루콘아세토박터속 균주를 이용하여 감귤에서 추출한 셀룰로오스 겔을 창상치유용으로 사용을 검토하여 대한민국 특허 제10-2011-0117794호로 출원한 바 있다.
따라서, 본 발명의 목적은 감귤 유래의 박테리아 셀룰로오스를 주재로 하는생분해성 및 무독성 필름 타입의 의료용 드레싱 제재를 제공하는데 있다.
본 발명의 상기 목적은 필름 타입의 감귤 유래 박테리아 셀룰로오스 필름 제작 단계와; 상기 제작한 박테리아 셀룰로오스 필름을 선형 창상을 유발시킨 백서에 이식하는 단계와; 육안적, 조직학적 및 생화학적 분석을 통해 상기 창상 부위에 이식한 박테리아 셀룰로오스 필름의 생분해성 및 독성 검사 단계를 통하여 달성하였다.
본 발명은 감귤 유래의 박테리아 셀룰로오스를 필름 타입으로 제공하는 효과가 있고, 제작된 필름 타입 셀룰로오스의 생분해성 및 무독성의 안전성이 확보된 생체적합한 창상치유용 드레싱 제재를 제공하는 뛰어난 효과가 있다.
도 1은 본 발명에서 제작한 필름 타입의 박테리아 셀룰로오스의 사진도이다.
도 2는 백서의 등에 길이 1.5 cm의 선형 창상을 만들어 피하에 필름 타입의 감귤 유래 박테리아 셀룰로오스(넓이 1cm2)를 이식한 후 Nylon 4-0를 이용하여 봉합하는 과정을 보여주는 사진도이다.
도 3은 필름 타입 감귤바이오겔의 이식 3일과 28일 후 조직병리학적 분석을 통한 생분해성 결과를 나타낸 것이다.
도 4는 백서에 이식한 필름 타입의 감귤바이오겔에 의한 독성검사를 간 및 신장의 조직병리학적 분석을 통해 나타낸 결과이다.
이하, 본 발명의 구체적인 내용을 바람직한 실시예와 실험예를 통하여 상세히 설명한다.
도 2는 백서의 등에 길이 1.5 cm의 선형 창상을 만들어 피하에 필름 타입의 감귤 유래 박테리아 셀룰로오스(넓이 1cm2)를 이식한 후 Nylon 4-0를 이용하여 봉합하는 과정을 보여주는 사진도이다.
도 3은 필름 타입 감귤바이오겔의 이식 3일과 28일 후 조직병리학적 분석을 통한 생분해성 결과를 나타낸 것이다.

이하, 본 발명의 구체적인 내용을 바람직한 실시예와 실험예를 통하여 상세히 설명한다.
본 발명에 사용된 실험동물로는 체중 160g - 200g의 Sprague-Dawley종 백서 수컷을 사용하였다. 실험동물은 온도 24 ± 0.5℃, 습도 55 - 65% 및 12시간 주기로 명암이 자동으로 조절되는 사육조건하에서 관리되었으며 약 14일간 환경에 적응시킨 후 실험에 사용되었다.
이하, 본 발명의 구체적인 방법을 실시예와 실험예를 들어 상세히 설명하고자 하지만 본 발명의 권리범위는 이들 예에만 한정되는 것은 아니다.
실시예 1 : 필름 타입의 감귤 유래 박테리아 셀룰로오스 필름 제작
감귤농축액은 국립원예특작과학원 감귤 시험장에서 제조한 평균당도 65 °Brix의 서귀포산 온주밀감 농축액으로 -20℃에서 저장한 후 해동하여 사용하였다. 겔 발효용 배지로는 시판 당액 9%, 감귤농축액 1%(v/v), ethanol 1%(v/v) 및 acetic acid 1%(v/v)를 첨가하여 발효용 감귤과즙배지를 조제하였다. 감귤발효액으로부터 분리한 겔 발효균 Gluconacetobacter sp. gel_KCG326(KACC 91555P)을 감귤과즙배지에 수회 반복 배양한 후 배지 표면에 형성된 겔을 종균으로 사용하였다. 배지를 50 mL 용적의 발효병에 35 mL씩 분주하고 전 배양한 겔 5%(W/V)를 접종한 후 발효병 입구를 살균된 filter paper로 덮고 28℃의 배양실에서 7일간 정치 배양하였다. 배양된 박테리아 셀룰로오스는 감귤겔에 0.1N NaOH와 0.1N HCl로 반복 처리하여 색소와 불순물을 제거한 뒤 반 정제한 박테리아 셀룰로오스를 분쇄기로 3분간 마쇄하여 증류수에 분산시킨 후 15분간 1차 가압 살균하였다. 상기 살균된 박테리아 셀룰로오스를 200 mesh screen에 담아 수분을 제거하고 40℃ dry oven에서 건조(수분함량 : 6.88%, Moisture analyzer)한 후 다시 15분간 2차 가압 살균하여 사용하였다. 도 1은 본 발명에서 제작한 두께 160㎛(Digimatic micrometer), 넓이 1㎝ × 1㎝인 필름 타입의 박테리아 셀룰로오스를 보여주고 있다.
본 발명에서 상기 필름 타입의 드레싱 제재는 가장 바람직한 넓이와 두께를 한정하였다.
실시예 2 : 감귤 유래의 박테리아 셀룰로오스 필름 이식
12마리 백서를 두 집단으로 분류하고, 한 집단은 등부위에 길이 1.5 cm의 선형 창상을 만들어 피하에 필름 타입의 감귤바오겔(넓이 1cm2)을 이식한 후 Nylon 4-0를 이용하여 봉합하고(도 2), 다른 한 집단은 창상을 낸 후 이식을 하지 않고 봉합하였다. 수술 시에는 졸레틸 50®(Virbac, Frances)을 50mg/kg으로 근육 주사하여 마취하였다.
실험예 1 : 조직병리학적 분석을 통한 감귤 유래 박테리아 셀룰로오스의 생분해성 관찰
감귤바이오겔 필름의 생분해성 관찰을 조직병리학적인 분석으로 살펴보았다. 상기 필름 이식한 부위의 피부조직을 술후 3일과 28일에 각 집단에서 3마리씩 희생하여 적출한 후 즉시 10% 포르말린에 고정하고 파라핀에 포매한 후 6 ㎛ 두께로 잘라 헤마톡실린-에오신(Hematoxylin-Eosin)으로 염색하여 관찰하였다.
실험결과 도 3에서 보는 바와 같이 이식 3일 후의 조직에서는 정상조직과 비교하여 분해되지 않은 감귤바이오겔 필름이 남아 있는 것이 관찰 되었으나 이식 28일 후 백서의 피하 조직에서는 필름이 분해되어 없어진 것을 확인하였다. 따라서 감귤바이오겔 필름은 4주 정도에 효과적으로 피하에서 생분해됨을 알 수 있었다.
실험예 2 : 감귤 유래 박테리아 셀룰로오스 필름의 독성검사
필름 타입의 감귤 유래 박테리아 셀룰로오스가 백서의 피하에서 생분해되었을 때 독성을 유발하는지를 살펴보기 위하여 실시예 2에서 필름 이식한 후 28일째 백서의 체중 및 장기(간, 신장, 비장, 폐, 심장)를 분리하여 무게를 측정하였다. [표 1]에서 보는 바와 같이 셀룰로오스를 이식한 군과 이식하지 않은 control군 사이의 몸무게와 장기 무게는 통계학적으로 유의한 차이가 없었다.

체내에 들어온 독성물질에 대한 대표적인 해독작용 기관인 간과 신장에서 독성이 나타나는지 살펴보기 위해서 독성 지표인 albumin, asparatate aminotransferase(AST), alanine aminotransferase(ALT), total bilirubin, creatinine 및 blood urea nitrogen(BUN) 단백질들의 변화를 확인해 본 결과, [표 2]에서 보는 바와 같이 그 수치에서 유의적인 변화를 발견하지 못하였다. 그뿐만 아니라, 간과 신장의 조직병리학적 관찰에서도 특이적인 이상조직을 발견하지 못하였다(도 4).

따라서, 상기 실험들을 통하여 백서의 피하에 이식한 필름 타입의 감귤 유래 박테리아 셀룰로오스가 독성을 일으키지 않는다는 것을 확인할 수 있었다.
이상에서 살펴본 바와 같이 본 발명은 체내에서 효과적으로 생분해되고 독성을 일으키지 않는 필름 타입의 감귤 유래 박테리아 셀룰로오스를 제공하는 효과가 있고, 이를 이용한 안전성이 확보된 생체적합소재를 제공하는 뛰어난 효과가 있으므로 바이오의료산업상 매우 유용한 발명인 것이다.
Claims (2)
- 감귤 유래의 셀룰로오스 겔을 0.1N NaOH와 0.1N HCl로 반복 처리하여 색소와 불순물을 제거한 뒤, 반 정제한 박테리아 셀룰로오스를 분쇄기로 3분간 마쇄하여 증류수에 분산시켜 15분간 1차 가압 살균한 후, 200 mesh screen에 담아 수분을 제거하고 40℃ dry oven에서 건조한 뒤, 다시 15분간 2차 가압 살균하여 필름 타입으로 제작된 것이 특징인 생분해성 및 무독성의 감귤 유래 박테리아 셀룰로오스를 주재로 하는 드레싱 제재.
- 제1항에 있어서, 상기 드레싱 제재는 두께 160㎛, 넓이 1㎝ × 1㎝ 임을 특징으로 하는 필름 타입의 의료용 드레싱 제재.
